Envelope-dimer epitope 1 (EDE1) antibody (C10) treatment significantly reduces Zika virus replication in the male and female reproductive tracts.

No effective therapy or vaccine exists to protect against the next Zika virus (ZIKV) outbreak. ZIKV has been detected in multiple organs of infected people, including immune-privileged sites like the brain, eyes, and reproductive tract. ZIKV replication in the reproductive tract is of high concern; ZIKV can be transmitted sexually or to the developing fetus of pregnant women, resulting in severe congenital defects. Here, we show that ZIKV-infected immunocompetent mice rapidly controlled viremia with no viral rebound following T cell depletion. In contrast, mice genetically deficient in B and T

Oxidative Stress and Interferon Signaling Drive Differential Pathogenesis of Ancestral and Contemporary Zika Viruses in Human Cerebral Organoids.

UNLABELLED: Neurotropic Zika viruses (ZIKV) cause serious human disease with pandemic potential. Pathogenesis severities resulting from Asian/American versus African ZIKV lineage infections range from mild to severe, respectively; however, mechanisms underlying differential ZIKV pathogenesis remain unclear, as do effective therapeutic strategies. The limitations of mechanistic understanding are due in part to the challenges of comparing data generated in disparate experimental models, as well as approaches that did not test both ancestral and contemporary ZIKV infections. The goal of this work

The modified mRNA vaccine protects immunocompromised AG129 mice from lethal challenge and multi-tissue infection by Zika virus.

The multiple epidemics of Zika virus (ZIKV) posed a substantial threat to public health. Clinical evidence suggests that ZIKV could break through the blood-brain, blood-placenta, and blood-testis barriers, leading to severe outcomes such as congenital malformations in newborns and Guillain-Barré syndrome in adults. Currently, there are no specific treatments for ZIKV infection. To address the antibody-dependent enhancement (ADE) of dengue virus (DENV) infection induced by ZIKV vaccination, we designed two modified prM-E RNAs (ZA and ZB) with specific mutations either shielding or disrupting th

Gestational inoculation by the Zika virus causes cognitive impairment and Na+K+-ATPase activity imbalance in frontal cortex of adult male and female Wistar rat's offspring.

It has been recently described the Congenital Zika Syndrome (CZS). Children from pregnant women who were infected by the virus have expressed a set of symptoms, particularly involving neurological disorders such as microcephaly. Animal models have been conducted aiming to enhance the knowledge about the CZS and giving support for future studies proposing prevention and treatment for this condition. The aim of the present study was to advance in establishing a rat model of CZS observing the impact of zika inoculation during pregnancy on the cognitive processes and nervous tissue damage in the m

Zika virus impairs autophagic flux in trabecular meshwork, and inhibition of autophagy restricts ocular viral transmission and associated pathology.

UNLABELLED: Zika virus (ZIKV) is the lone member of Flavivirus family known to cause congenital glaucoma following in utero exposure. The molecular mechanisms of ZIKV-induced glaucoma remain elusive, with no known therapeutic modalities. Autophagy plays a dual role in viral infections and glaucoma pathogenesis. However, how ZIKV interferes with autophagic pathways in the trabecular meshwork (TM) and whether autophagy contributes to ZIKV-induced ocular complications remains elusive. Here, we investigated the role of autophagy in ZIKV ocular pathogenesis utilizing a primary human trabecular mesh

The burden of musculoskeletal symptoms in caregivers of children with congenital Zika syndrome: a cross-sectional study.

The continuous daily care required by children with congenital Zika syndrome (CZS) may overload the lives of caregivers. However, little is known about the factors associated with musculoskeletal symptoms experienced by caregivers. This study aimed to investigate the prevalence of musculoskeletal symptoms in caregivers of children with CZS and identify its associated factors. This cross-sectional study included caregivers of children with CZS from northeast Brazil. Musculoskeletal symptoms were assessed using the Nordic Musculoskeletal Questionnaire. Information about caregivers, pregnancy, ch

Variation in virion phosphatidylserine content drives differential GAS6 binding among closely related flaviviruses.

UNLABELLED: Many enveloped viruses engage phosphatidylserine (PS) receptors to enter cells, a phenomenon known as "apoptotic mimicry." We previously reported that Zika virus (ZIKV), but not closely related West Nile virus (WNV) or dengue virus (DENV), utilized AXL to infect cells because only ZIKV could bind the AXL ligand GAS6, a PS-binding protein. In this study, we investigated the mechanisms underlying the differential ability of these viruses to bind GAS6. Although immature virions expose larger patches of the viral membrane than do mature ones, our data show that virion maturity levels d

Zika virus induces persistent phenotypic changes in natural killer cells distinct from dengue virus infection.

Zika virus (ZIKV) is a mosquito-borne orthoflavivirus of global concern due to its ability to cause congenital neurological defects in infants. Natural killer (NK) cells are innate lymphocytes that can directly kill virus-infected cells. It is thought that NK cells are protective against ZIKV, although the mechanisms by which NK cells detect and respond to ZIKV are not well understood. Here, we evaluated NK cell receptor expression on isolated NK cells from a Panamanian cohort of ZIKV-infected participants, and corresponding NK receptor ligand expression on participant PBMCs and ZIKV-infected

Novel Thiazolidinedione Derivatives as Potential ZIKV Antiviral Inhibitors.

Zika virus (ZIKV) remains a pressing global health concern due to its association with congenital Zika syndrome and the current lack of approved antiviral therapies. In this study, we evaluated the antiviral activity of three novel thiazolidinedione derivatives, GQ-402, GQ-396, and ZKC-10, against ZIKV in vitro and investigated their potential molecular targets through in silico analysis. GQ-402 exhibited the highest antiviral potency, with an IC50 of 15.7 µM, while ZKC-10 achieved the most substantial reduction in viral RNA levels, as determined by RT-qPCR. Molecular docking studies identifie

Long-Term Nutritional Deficits and Growth Patterns in Children with Congenital Zika Virus Syndrome: Evidence from a Brazilian Cohort.

Children with Congenital Zika Virus Syndrome (CZVS) experience severe neurological and nutritional impairments. Although immediate clinical consequences are well-documented, long-term anthropometric and nutritional outcomes remain poorly understood. This study assessed longitudinal anthropometric and nutritional outcomes in children affected by CZVS. A cohort of 38 children aged ≥ 5 years diagnosed with CZVS was followed at a reference center in Northeast Brazil. Anthropometric measures (weight, height, BMI, head circumference) were collected using standardized methods, including digital scale

Association between prenatal Zika virus exposure and neurodevelopmental outcomes in offspring: a protocol for a systematic review and meta-analysis of observational studies.

INTRODUCTION: Zika virus (ZIKV) infection during pregnancy has been associated with adverse birth outcomes, including congenital Zika syndrome, characterised by features such as microcephaly. However, the broader neurodevelopmental influence of prenatal ZIKV exposure, especially among offspring without congenital anomalies at birth, remains poorly understood. While previous studies, including a review, have explored neurodevelopment in ZIKV-exposed children, comparative pooled estimates between exposed and unexposed groups remain scarce. This systematic review and meta-analysis aims to evaluat

Heterologous prime-boost Zika virus vaccination induces comprehensive humoral and cellular immunity in mouse models.

Zika virus (ZIKV) remained poorly studied until an outbreak in 2015 linked the virus to severe neurological disorders and congenital malformations. Currently, there are no antiviral drugs or vaccines available. We have previously demonstrated that a simian adenovirus vector vaccine (ChAdOx1 prMEΔTM) and a virus-like particle-based vaccine bearing E proteins locked in covalent dimers (VLP-cvD) are effective against ZIKV infection in animal challenge models. In this study, we further explored the efficacy of these vaccines, either individually or in combination, using a heterologous prime and bo

Neuroteratogenic mechanisms of Zika virus (ZIKV) infection: Insights into fetal brain development disruption and congenital Zika syndrome: A systematic review.

The Zika virus (ZIKV), a mosquito-borne Flavivirus, has emerged as a global health threat due to its severe neuroteratogenic effects, particularly during pregnancy. This mechanistic-to-translational synthesis examines how ZIKV disrupts fetal brain development and links molecular events to clinical manifestations of Congenital Zika Syndrome (CZS). Our review advances prior summaries by presenting a unifying neural progenitor cell (NPC)-centric pathway framework that integrates viral entry, host signaling disruption, cell fate outcomes, and imaging correlates. Once transmitted vertically, the vi

A VLPs based vaccine protects against Zika virus infection and prevents cerebral and testicular damage.

Still, Zika virus (ZIKV) infection poses a substantial public health risk, especially for pregnant women and their fetuses, as it can result in congenital abnormalities and fetal mortality during pregnancy. Despite significant advances in understanding and combating ZIKV, considerable challenges remain in the fight against this flavivirus. A crucial component of this effort is the development of vaccines, none of which have yet been licensed for human use. Here, we present a comprehensive study of a novel ZIKV vaccine candidate based on virus-like particles (VLPs), designed to provide broad im

Zika Virus: A Review of Biology, Clinical Impacts, and Coinfections.

The Zika virus (ZIKV) gained prominence as a significant global pathogen after the 2015-2016 outbreaks associated it with an increase in neurological complications in adults and congenital malformations. Different mechanisms have been proposed by which ZIKV may cross the blood-brain barrier and reach the central nervous system to cause neuroinflammation. Although ZIKV infection triggers a robust immune response, the virus has developed different strategies to escape it. Furthermore, although the virus is present in areas with cocirculation of other pathogenic agents, few studies have evaluated

A Zika Virus-Like Particle Vaccine Mitigates Early Pregnancy Loss In Rhesus Macaques.

Zika virus (ZIKV) is an arthropod-borne Orthoflavivirus that caused a major outbreak in the Americas in 2015-16. In Brazil, up to 46% of ZIKV positive pregnancies resulted in congenital Zika syndrome (CZS). CZS is characterized by a wide range of neurologic birth defects and miscarriage in up to 7.6% of affected pregnancies. With no current licensed ZIKV vaccines, we sought to evaluate a Zika virus-like particle (VLP) vaccine candidate in a rhesus macaque (RM) pregnancy model. VLPs were produced in mammalian cells expressing the pre-membrane-envelope region of the Asian-lineage ZIKV strain PRV

Neural behavior during cystometry of children with congenital Zika virus: Pilot study.

INTRODUCTION: Understanding the neural control of bladder function is essential for advancing neuroscience and improving urological outcomes, especially in children with neurological impairments such as Congenital Zika Syndrome (CZS). Recent evidence of urological dysfunctions in CZS patients highlights the need for integrated neurological and urodynamic investigations. Thus, this exploratory pilot study aimed to evaluate the feasibility and safety of simultaneous electroencephalography (EEG) and urodynamic studies in children with CZS, and to characterize the patterns of bladder and brain act
