Association of Influenza Vaccination During Pregnancy with Health Outcomes in Mothers and Children: A Population-Based Cohort Study.

Immunization rates of maternal influenza vaccination during pregnancy remain suboptimal, with concerns about potential harm to the mothers and their offspring. We conducted a population-based cohort study, using mother-child linked database in Korea: (a) maternal cohort between December 2019, and March 2022; (b) neonatal cohort between September 2020, and June 2021. Exposure was defined as influenza vaccination during pregnancy. Study outcomes included gestational outcomes, vaccine-related adverse events, and other health outcomes in mothers and childbirth and immune-related health outcomes in

Insights into the Absence of Lymphoma Despite Fulminant Epstein-Barr Virus Infection in Patients with XIAP Deficiency.

X-linked Lymphoproliferative Syndromes (XLP), which arise from mutations in the SH2D1A or XIAP genes, are characterized by the inability to control Epstein-Barr Virus (EBV) infection. While primary EBV infection triggers severe diseases in each, lymphomas occur at high rates with XLP-1 but not with XLP-2. Why XLP-2 patients are apparently protected from EBV-driven lymphomagenesis, in contrast to all other described congenital conditions that result in heightened susceptibility to EBV, remains a key open question. To gain insights, we cross-compared newly EBV infected versus immune stimulated B

Emergence of a novel porcine pestivirus with potential for cross-species transmission in China, 2023.

Pestiviruses, RNA viruses belonging to the Flaviviridae family, have a broad host range. Their pathogenicity varies greatly and they have caused significant economic losses in animal husbandry. In this study, a novel pestivirus, porcine abortion-associated pestivirus (PAAPeV), was isolated from pigs in China in 2023. Clinically, PAAPeV causes abortions in sows and leads to congenital tremors and death in piglets. PAAPeV replicates efficiently in ST cells. Morphologically, PAAPeV virions are spherical particles with a diameter of approximately 80 nm. Phylogenetic analysis reveals that PAAPeV is

A Review of the Recent Epidemiology of Zika Virus Infection.

Zika virus (ZIKV) is a flavivirus transmitted primarily by the bite of infected Aedes species mosquitoes. Although typically asymptomatic or causing mild symptoms and infrequent neurological disease in older children and adults, infection during pregnancy can result in severe congenital malformations and neurodevelopmental deficits. We conducted a review of published literature and official data sources to describe recent Zika epidemiological trends, building on WHO updates posted in 2019 and 2022. Globally, cases declined after the height of ZIKV transmission in the Americas in 2015-2016; how

Practice recommendations for respiratory syncytial virus prophylaxis among children in Hong Kong.

Hong Kong has a high burden of hospitalisations associated with respiratory syncytial virus (RSV) infection in young children. Most international guidelines concerning RSV prophylaxis are based on studies conducted in temperate climates and may not fully apply to subtropical locations such as Hong Kong. In July 2022, a group of nine experts in neonatology, paediatric intensive care, paediatric respiratory medicine, and paediatric cardiology in Hong Kong convened to formulate recommendations for RSV prophylaxis. The recommendations were based on literature review and expert discussion. Each exp

Disruption of functional network development in children with prenatal Zika virus exposure revealed by resting-state EEG.

Children born to mothers infected by Zika virus (ZIKV) during pregnancy are at increased risk of adverse neurodevelopmental outcomes including microcephaly, epilepsy, and neurocognitive deficits, collectively known as Congenital Zika Virus Syndrome. To study the impact of ZIKV on infant brain development, we collected resting-state electroencephalography (EEG) recordings from 28 normocephalic ZIKV-exposed children and 16 socio-demographically similar but unexposed children at 23-27 months of age. We assessed group differences in frequency band power and brain synchrony, as well as the relation

Role of non-human primate models in accelerating research and developing countermeasures against Zika virus infection.

Zika virus, a mosquito-transmitted orthoflavivirus, has become a pathogen of global health concern ever since the virus caused an epidemic in Brazil in 2015 associated with approximately 700 000 laboratory-confirmed cases of congenital microcephaly. The subsequent spread of the epidemic in 2016 resulted in a wide spectrum of congenital neurological, ophthalmological, and developmental abnormalities across the Americas, Africa, and Asia. In this context, non-human primate models have become essential tools for Zika virus research to understand the pathogenesis of congenital brain injury and per

Zika virus: advancing a priority research agenda for preparedness and response.

The 2015-16 Zika virus epidemic emerged in the Americas and rapidly spread throughout the region and beyond, showing the epidemic potential of this mosquito-borne Orthoflavivirus and its capacity to cause severe congenital malformations and neurological sequelae. WHO declared the Zika virus epidemic a public health emergency of international concern in 2016. Despite this declaration, there are no licensed Zika virus vaccines, therapeutics, or diagnostic tests appropriate for routine antenatal screening. To address this absence of essential tools to detect and mitigate the threat of future Zika

Cell-penetrating peptide-grafted AAV2 capsids for improved retinal delivery via intravitreal injection.

Recombinant adeno-associated virus (rAAV) is a leading vector for retinal gene therapy due to its favorable safety profile demonstrated by the FDA-approved Luxturna for Leber congenital amaurosis. However, challenges with low transduction efficiency and immunogenicity, coupled with the invasiveness of subretinal injections, have driven efforts to engineer AAV capsids for minimally invasive intravitreal delivery. Intravitreal injections face the barrier of the inner limiting membrane (ILM), particularly with AAV2-based vectors. In this study, we displayed cell-penetrating peptides (CPPs) on AAV

Congenital Zika virus infection in laboratory animals: a comparative review highlights translational studies on the maternal-foetal interface.

The 2015-16 Zika virus (ZIKV) epidemic has posed unprecedented concern for maternal-infant health, mainly due to the substantial risk of microcephaly and other neurological birth abnormalities associated with congenital ZIKV syndrome (CZS). As licenced vaccines and effective antivirals are still unavailable, attention has been focused on post-delivery in vitro or translational in vivo studies to understand the impact of maternal ZIKV infection on placentation and neurodevelopmental consequences for the foetus. Here, we review clinical and translational studies highlighting ZIKV-induced materna

Age-associated differences in mucosal and systemic host responses to SARS-CoV-2 infection.

Age is among the strongest risk factors for severe outcomes from SARS-CoV-2 infection. Here we describe upper respiratory tract (URT) and peripheral blood transcriptomes of 202 participants (age range of 1 week to 83 years), including 137 non-hospitalized individuals with mild SARS-CoV-2 infection and 65 healthy individuals. Among healthy children and adolescents, younger age is associated with higher URT expression of innate and adaptive immune pathways. SARS-CoV-2 infection induces broad upregulation of URT innate and adaptive immune responses among children and adolescents. Peripheral blood

Co-detection of various viruses in SARS-CoV-2 in children with respiratory infections.

The co-detection of SARS-CoV-2 variant with other respiratory virus has been extensively investigated. However, conclusive evidence remains elusive and conflicted. This study investigated the source- and age-dependent prevalence, incidence, and co-detection of multiple viral infections in children younger than 18 years old who presented with clinical symptoms indicative of respiratory infections during SARS-CoV-2 pandemic. We retrospectively obtained and analyzed pediatric patients admitted to the institution and underwent Film Array Respiratory Panel (BIOFIRE® RP2.1) testing between January 2

COVID-19 Vaccination During Pregnancy and Major Structural Birth Defects.

BACKGROUND AND OBJECTIVES: COVID-19 vaccination is recommended during pregnancy; however, evidence on the prevalence of major structural birth defects born to people vaccinated early in pregnancy (≤20 weeks of gestation) is limited. We compared the prevalence of major structural birth defects by COVID-19 vaccination status and key strata: insurance provider, clinically diagnosed SARS-CoV-2 infection during pregnancy, and concomitant administration of other maternal vaccines. We also compared, head-to-head, the prevalence of birth defects by brand (Moderna mRNA-1273 vs Pfizer-BioNTech BNT162b2)

Retinal gene therapy for Stargardt disease with dual AAV intein vectors is both safe and effective in large animal models.

Retinal gene therapy using dual adeno-associated viral (AAV) intein vectors can be applied to genetic forms of blindness caused by mutations in genes with coding sequences that exceed single AAV cargo capacity, such as Stargardt disease (STGD1), the most common inherited macular dystrophy. In view of clinical translation of dual AAV intein vectors, here we set to evaluate both the efficiency and safety of their subretinal administration in relevant large animal models. Accordingly, we have developed the first pig model of STGD1, which we found to accumulate lipofuscin similarly to patients. Th

A novel approach using IFNAR1 KO mice for assessing Akabane virus pathogenicity and vaccine efficacy.

Akabane virus (AKAV) is a Simbu serogroup virus that can cause congenital abnormalities in ruminants. In 2010, an AKAV-7 strain exhibiting different characteristics and belonging to a distinctive genogroup compared to previous AKAVs was isolated in South Korea. Although this novel pathogenic AKAV-7 has been discovered, in vivo studies on AKAV-7 are currently insufficient due to limitations of using large animals and suckling mice. Therefore, the development of a novel small animal model for AKAV studies is necessary. Type I interferon receptor knock out (IFNAR1 KO) mice are widely employed as

Assessing Schmallenberg Virus Disease in Sardinia (Italy) After the First Epidemic Episode in 2012.

Schmallenberg virus (SBV), an Orthobunyavirus transmitted by Culicoides, causes congenital malformations and mild symptoms, such as fever, reduced appetite, decreased milk production, and occasional diarrhea, in ruminants. First detected in Central Europe in 2011, SBV spread across the continent, reaching Sardinia (Italy) in late 2012. This study evaluates the occurrence of SBV infections in Sardinian sheep from 2013 to 2024 by anatomo-pathological, virological, serological, and entomological data. The results suggest the presence of SBV infections in a continuous enzootic status over the year
