Heterologous prime-boost Zika virus vaccination induces comprehensive humoral and cellular immunity in mouse models.

Zika virus (ZIKV) remained poorly studied until an outbreak in 2015 linked the virus to severe neurological disorders and congenital malformations. Currently, there are no antiviral drugs or vaccines available. We have previously demonstrated that a simian adenovirus vector vaccine (ChAdOx1 prMEΔTM) and a virus-like particle-based vaccine bearing E proteins locked in covalent dimers (VLP-cvD) are effective against ZIKV infection in animal challenge models. In this study, we further explored the efficacy of these vaccines, either individually or in combination, using a heterologous prime and bo

The differential burden of acute rhinovirus infections in children with underlying conditions.

INTRODUCTION: Rhinoviruses (RVs) are a well-known trigger of wheezing episodes in children with asthma. Their role in other pediatric chronic medical conditions is not fully known. METHODS: Patients ≤21 years hospitalized or evaluated as outpatients with symptomatic RV infection were identified from 2011-2013. Patients were categorized based on the type of underlying disease and differences in clinical parameters, RV loads (CT values), viral, bacterial and fungal co-infections and clinical outcomes were compared between groups. Multivariable analyses were performed to identify the comorbiditi

A VLPs based vaccine protects against Zika virus infection and prevents cerebral and testicular damage.

Still, Zika virus (ZIKV) infection poses a substantial public health risk, especially for pregnant women and their fetuses, as it can result in congenital abnormalities and fetal mortality during pregnancy. Despite significant advances in understanding and combating ZIKV, considerable challenges remain in the fight against this flavivirus. A crucial component of this effort is the development of vaccines, none of which have yet been licensed for human use. Here, we present a comprehensive study of a novel ZIKV vaccine candidate based on virus-like particles (VLPs), designed to provide broad im

Zika Virus: A Review of Biology, Clinical Impacts, and Coinfections.

The Zika virus (ZIKV) gained prominence as a significant global pathogen after the 2015-2016 outbreaks associated it with an increase in neurological complications in adults and congenital malformations. Different mechanisms have been proposed by which ZIKV may cross the blood-brain barrier and reach the central nervous system to cause neuroinflammation. Although ZIKV infection triggers a robust immune response, the virus has developed different strategies to escape it. Furthermore, although the virus is present in areas with cocirculation of other pathogenic agents, few studies have evaluated

Improving the quality of care of respiratory syncytial virus in the neonatal and pediatric populations globally.

Respiratory syncytial virus (RSV) is the leading cause of hospitalization for bronchiolitis in infants worldwide, and age (<6 months) and underlying comorbidities (e.g., prematurity, congenital heart disease) are risk factors for severe disease. However, some centers face challenges in identifying and implementing preventative measures, and best practices, for the care of at-risk infants. Therefore, this study aimed to identify best practice examples in RSV care for neonatal and pediatric populations in leading centers globally, and to understand how these practices can be widely implemented.

DYRK1A in blood and immune function: implications in leukemia, inflammatory disorders, infection and Down syndrome.

Down syndrome (DS) is the most frequent autosomal aneuploidy, and it arises due to an extra copy of human chromosome 21. Individuals with trisomy 21 (T21) exhibit an increased predisposition towards a wide number of developmental and physiological alterations, often referred to as DS co-occurring conditions, including congenital heart disease, leukemia, intellectual disability, neurodegenerative disorders or autoimmune diseases, among many others. The overexpression of several genes encoded on chromosome 21 have been linked to many of such T21-associated disorders, but we are still very far fr

Zika virus and the fetal-maternal interface: deciphering the mechanisms of placental infection and implications for pregnancy outcomes.

Zika virus (ZIKV) is an emerging flavivirus primarily transmitted by Aedes mosquitoes. It has gained significant attention due to its potential teratogenic effects during pregnancy, particularly the increased risk of severe congenital outcomes such as microcephaly and fetal mortality. ZIKV's ability to breach the fetal-maternal interface and compromise placental function distinguishes it from other arboviruses. This mini-review explores how ZIKV targets key placental cells, including cytotrophoblasts, extravillous trophoblasts, syncytiotrophoblasts, and Hofbauer cells, to establish infection i

Maternal inflammation disrupts neurogenesis and neocortical organization, affecting social and stress-related behavior.

It is imperative to acknowledge the potential impact of various environmental factors on the development of developmental disorders. Among these factors, the maternal immune response induced by the onset of infectious diseases during pregnancy emerges as a notable consideration. The study elucidates the behavioral and morphological abnormalities of the cerebral cortex caused by maternal immune response during pregnancy, emphasizing the significance of maternal immunity in this context. To this end, a mouse model of prenatal infection was established, inducing a congenital immune response analo

African strains of Zika virus resist ISG-mediated restriction.

Zika virus (ZIKV) is a neurotropic Orthoflavivirus transmitted by mosquito vectors, which has evolved into two lineages, namely African and Asian. ZIKV from the Asian lineage has been responsible for epidemics in the Pacific and the Americas, the largest of which occurred in Brazil in 2015 and was associated with severe neurological disorders, including cases of microcephaly and other congenital fetal malformations. Although never implicated in human epidemics, African strains exhibit faster replication, higher virus production, and greater virulence in animal models compared to their Asian co

Neuroendocrinology and the Genetics of Obesity.

The increase in the incidence of obesity has coincided with changes in lifestyle, diet, and environment. Comorbidities associated with obesity include cardiovascular disease, diabetes, musculoskeletal disorders, stroke, and thromboembolism, affecting public health. The effect of increased weight has recently become even more obvious, since obesity has been significantly associated with increased severity and higher mortality among COVID-19 patients. The need to decrease rates of obesity prompted a surge in the use of glucagon-like peptide-1 agonist medications. Twin studies, however, determine

Palivizumab for preventing severe respiratory syncytial virus (RSV) infection in children.

RATIONALE: Respiratory viruses are the leading cause of lower respiratory tract infection and hospitalisation in infants and young children. Respiratory syncytial virus (RSV) is the main infectious agent in this population. Palivizumab is administered intramuscularly every month for five months in the first RSV season to prevent serious RSV lower respiratory tract infection in children. Given its high cost, it is essential to know if palivizumab continues to be effective in preventing severe RSV disease in children. OBJECTIVES: To assess the effects of palivizumab in preventing severe RSV inf

Pediatric Zika virus: The virus and its neurologic impact.

The Zika virus (ZIKV) is a mosquito-borne flavivirus, transmitted by the genus Aedes, and is prevalent in tropical areas. It co-exists and shares a vector with other flaviviruses, such as the dengue virus. Its infection usually leads to a febrile illness undistinguishable of other viruses. Zika virus raised to prominence during an epidemic in the Americas during 2015 and 2016, as the increase in cases coincided with an increase in neonates born with severe microcephaly. This led to the description of congenital Zika syndrome and the establishment of the potential for ZIKV to cause congenital d

Schmallenberg virus epidemiology and regional control strategies: diagnostics, vaccines, and vector management.

Schmallenberg virus (SBV) is an emerging orthobunyavirus transmitted by Culicoides midges. It poses a serious global health threat to ruminants, especially during pregnancy, causing abortion, stillbirths, and congenital malformations. Since its first outbreak in 2011, SBV has spread across Europe and other regions. Its transmission has expanded due to global climate change and increased animal trade, resulting in recurrent outbreaks in endemic regions and a growing risk of introduction into non-endemic areas. This situation highlights the urgent need for improved control strategies. This revie

Dynamics of endemic virus re-emergence in children in the USA following the COVID-19 pandemic (2022-23): a prospective, multicentre, longitudinal, immunoepidemiological surveillance study.

BACKGROUND: The Pandemic Response Repository through Microbial and Immune Surveillance and Epidemiology (PREMISE) programme was established to translate knowledge gained from global immunoepidemiological surveillance into a better understanding of population-level dynamics of emerging and re-emerging infections, as well as into the discovery and development of biomedical countermeasures against potential pandemic threats. As proof of principle for this approach, we conducted a longitudinal immunoepidemiological study in children in the USA, focusing on enterovirus D68 (EV-D68) infection dynami
