Microbiome disruption is a proposed mechanism for the observed differences in child health outcomes by maternal HIV status, but the early neonatal microbiome of HIV-exposed (HE) newborns is not well-studied. We used 16S ribosomal ribonucleic acid sequencing to analyze the microbiome composition of nasal, skin, and rectal samples collected ≤72 h after birth from 57 hospitalized neonates in Botswana, 33% of whom were HE. Beta diversity differed by anatomic compartment (P = .001) and days since birth; however, interindividual differences were greater than those by anatomic site (P = .001). There

Streptococcus agalactiae (group B Streptococcus; GBS) is a leading cause of neonatal sepsis and meningitis. Despite advances in molecular microbiology, GBS genome engineering remains laborious due to inefficient mutagenesis protocols. Here, we report a versatile and rapid Cas12a-based toolkit for GBS genetic manipulation. We developed two shuttle plasmids-pGBSedit for genome editing and pGBScrispri for inducible CRISPR interference-derived from an Enterococcus faecium system and optimized for GBS. Using these tools, we achieved targeted gene insertions, markerless deletions, and efficient, tem

AIM: The aim of this study was to clarify whether the detection rate of group B Streptococcus (GBS) in screening tests for pregnant women at 35-37 weeks of gestation increases using the selective enrichment media available in Japan.
METHODS: Vaginal-rectal swabs were collected from pregnant women at 35-37 weeks of gestation from nine obstetric medical institutions in the Nagasaki Prefecture. The collected swabs were inoculated into selective enrichment media and directly plated onto blood agar to investigate the differences in the isolation frequencies of GBS.
RESULTS: We collected vaginal-r

UNLABELLED: Streptococcus agalactiae or Group B Streptococcus (GBS) is an opportunistic pathogen that asymptomatically colonizes the vaginal tract of up to 30% of healthy individuals. However, during pregnancy it is associated with adverse pregnancy outcomes, and GBS can be transmitted to the fetus in utero , or the newborn during vaginal birth, resulting in invasive neonatal disease. Previously we identified that Akkermansia muciniphila alters the dynamics of GBS vaginal colonization. However, the global effect of A. muciniphila in the female genital tract remains largely unknown, as it has p

Background/Objectives: Streptococcus agalactiae (Group B Streptococci, GBS) is Gram-positive, beta-hemolytic coccus known to be transmitted by vertical transmission in neonates during birth with neonatal sepsis, pneumonia, and meningitis. In adults, particularly the elderly and those with diabetes mellitus, GBS can also cause pneumonia and sepsis. Penicillin is the drug of choice, and GBS is generally susceptible to this antibiotic. This study investigates trends in GBS isolation rates and penicillin non-susceptibility over time at a university hospital. Methods: We retrospectively analyzed 24

BACKGROUND: Group B Streptococcus (GBS) commonly colonizes the female reproductive tract and rectum. An estimated 19.7 million pregnant women worldwide are colonized with GBS. Infection during late pregnancy can lead to adverse pregnancy outcomes. Pregnant women infected with GBS face a significantly increased risk of transmitting the bacteria to their newborns during delivery, potentially resulting in serious conditions such as sepsis, pneumonia, and meningitis major contributors to neonatal morbidity and mortality. While previous studies have primarily focused on the prevalence of GBS coloni

BACKGROUND: There has been little decline in neonatal mortality rates over recent decades, and this is now further challenged by the rising prevalence of antimicrobial resistance. In Australia, the incidence of neonatal sepsis is low on a global scale, yet there are increasingly frequent outbreaks of multidrug-resistant (MDR) infections in neonatal intensive care units, alongside rising rates of colonisation with MDR bacteria.
METHODS: We analysed positive blood and cerebrospinal fluid cultures collected from infants (aged 0 to ≤180 days) across five clinical sites in Australia between 2010 a

Streptococcus agalactiae also known as Group B Streptococcus (GBS) is a Gram-positive, encapsulated, pathogenic bacterium. GBS causes severe perinatal infections that lead to chorioamnionitis, funisitis, premature rupture of membranes, preterm birth, maternal sepsis, neonatal sepsis, stillbirth, and maternal demise. Epidemiological data indicate that the nutrient selenium provides protection against infection and adverse disease outcomes and is a critical nutrient for development of a healthy pregnancy. We hypothesized that selenium could have antimicrobial activity against GBS. To test this,

BACKGROUND: Streptococcus agalactiae is responsible for sepsis and meningitis, and the major cause of neonatal morbidity and mortality. However, how S. agalactiae disrupts endothelial barriers is poorly understood.
OBJECTIVES: Analyse the influence of endothelial cell (HUVECs) growth under static and shear stress conditions during infection with S. agalactiae, and the role of pilus PI-2b during endothelial barrier disruption and increased endothelial permeability.
METHODS: HUVECs under static and shear conditions were infected by S. agalactiae (GBS90356 and GBS90356Δpilus2b) strains in the p

OBJECTIVE: To evaluate whether expectant management of late preterm prelabor rupture of membranes (PPROM) in patients colonized with Group B streptococcus (GBS) is associated with an increased risk of adverse maternal and neonatal outcomes.
STUDY DESIGN: This cohort study analyzed data from singleton pregnancies complicated by late PPROM between 34+0 and 36+6 weeks of pregnancy, where a documented GBS rectovaginal culture result was available, and who underwent expectant management until 37+0 weeks. After rectovaginal GBS swabs were taken, patients received an ampicillin-based regimen: this w

INTRODUCTION: Group B Streptococcus (GBS) is a leading etiological agent of neonatal sepsis and meningitis, as well as invasive infections that predominately affect adults over 65 and people with comorbidities. The Centers for Disease Control and Prevention's (CDC) nationwide surveillance has identified rising clindamycin and erythromycin resistance as a concerning level threat for invasive GBS infections. West Virginia (WV) is a rural state outside the CDC catchment area, therefore, we investigated 65 WV-GBS isolates identified in clinical specimens from various sources for serotype distribut

OBJECTIVE: Early-onset neonatal sepsis due to ascending infection is a potentially preventable complication of preterm premature rupture of membranes. Our objective was to determine whether the analysis of bacteria from vaginal swab samples is predictive of the risk of early-onset neonatal sepsis in preterm premature rupture of membranes.
STUDY DESIGN: In a prospective 3-center observational cohort, patients with preterm premature rupture of membranes were enrolled between 22 and 36 weeks of gestation + 6 days. Vaginal swab samples at delivery were analyzed using 2 different approaches, class

Neonatal invasive Group B Streptococcus (GBS) infection remains a leading cause of mortality and long-term neurodevelopmental impairment. Effective treatment for neonatal GBS sepsis is critically hampered by a dysregulated immune response that fails to clear the infection and induces neuroinflammation. The P2X7 receptor (P2X7R) plays a key role in inflammatory responses and neuroinflammation during infection. Here, we investigated the effects of systemic P2X7R modulation on immune and neuronal dysfunction in a neonatal GBS infection model. Neonatal mice born to GBS-colonized dams were treated

OBJECTIVES: To examine the association between prenatal antibiotic exposure and Group B Streptococcus (GBS) disease within 4 weeks postpartum.
METHODS: We conducted a population-based cohort study including all singleton live births in Sweden from 2006 to 2016, using national registers. Neonates were classified by prenatal antibiotic exposure status, and GBS disease was ascertained within four weeks postpartum. Adjusted odds ratios (aOR) were estimated using multivariable logistic regression with a robust variance estimator. Effect heterogeneity by GBS risk factors was evaluated, and potentia

Streptococcus enterococci group D is a rare cause of neonatal sepsis, and it is associated with significant morbidity and a very high fatality rate. The infection is usually acquired intra-partum, from the colonization of the maternal genital tract. Most affected neonates have an early-onset presentation of symptoms, usually within the first 48 hours after birth, which is similar to other causes of neonatal sepsis such as Streptococcus agalactiae or Escherichia coli. We describe an early preterm infant with a fatal, early-onset sepsis caused by Streptococcus D.

Group B Streptococcus (GBS) is a component of the natural human microbiota, colonizing the genitourinary tract and the distal gastrointestinal tract. Due to its production of numerous virulence factors, GBS can cause infections in pregnant women, newborns, and immunocompromised individuals. In newborns, GBS infection may present as severe pneumonia, meningitis, or sepsis. Screening for maternal GBS colonization, combined with intrapartum antibiotic prophylaxis for colonized women, is currently regarded as the most effective strategy for preventing neonatal GBS infections. However, growing conc

Strongly encourage pregnant women with the history of a penicillin allergy to undergo prenatal allergy testing as first-line intrapartum antibiotic prophylaxis for group B strep (GBS) is superior to second-line or no antibiotics, which have similar rates of early onset sepsis (EOS).

Neonates born prematurely are highly vulnerable to life-threatening conditions such as bacterial sepsis. Streptococcus agalactiae, also known as group B Streptococcus (GBS) and Escherichia coli are frequent causative pathogens of neonatal sepsis, however, it remains unclear if distinct sepsis pathogens induce differential adaptive immune responses. In the present study, we find that γδ T cells in neonatal mice rapidly respond to single-organism GBS and E. coli bloodstream infections and that these pathogens induce distinct activation and cytokine production from IFN-γ and IL-17 producing γδ T
