Distinct STRIPAK subunits drive conserved and subunit-specific signaling programs in Cryptococcus neoformans.

The striatin-interacting phosphatase and kinase (STRIPAK) complex is a conserved protein phosphatase 2A (PP2A)-associated signaling hub that integrates kinase-phosphatase networks, yet its roles in human fungal pathogens remain poorly defined. Here, we dissected STRIPAK functions in the opportunistic pathogen Cryptococcus neoformans by combining genetic, genomic, virulence, and phosphoproteomic analyses across mutants lacking individual STRIPAK subunits. Loss of the core STRIPAK components via PPH22, FAR8, FAR9, or FAR11 mutations caused severe defects in growth, stress adaptation, cell cycle

Monoclonal Antibody-Based Anti-Cryptococcosis Therapy.

Cryptococcosis remains one of the most severe invasive fungal diseases worldwide, causing high morbidity and mortality, particularly among immunocompromised individuals. Despite the availability of antifungal drugs, current therapeutic regimens are limited by drug toxicity, prolonged treatment, antifungal resistance, biofilm-associated tolerance, and frequent disease relapse, highlighting the need for novel therapeutic approaches. Monoclonal antibodies (mAbs) have emerged as promising immunotherapeutic agents because of their ability to specifically target fungal components while modulating ho

Temporal shifts in adaptive immunity drive HIV-associated cryptococcal IRIS: mechanisms, models, and therapeutic opportunities.

Cryptococcal meningoencephalitis (CME) remains a leading cause of AIDS-related mortality, and up to one-quarter of persons living with HIV who survive CME develop cryptococcal immune reconstitution inflammatory syndrome (CME-IRIS) after antiretroviral therapy (ART) initiation. CME-IRIS arises when recovering immunity encounters a high residual fungal antigen burden, yet the temporal adaptive immune dynamics that distinguish protective reconstitution from damaging neuroinflammation are not fully defined. This review synthesizes clinical and experimental data to propose a temporal framework cent

Investigating the therapeutic potential of the antibacterial drug nitroxoline in combating cryptococcal infection.

Cryptococcosis is a major health threat among immunocompromised individuals, resulting in life-threatening consequences secondary to brain invasion. The limited treatment options and the increased incidence of treatment failure and relapses urge the need for more effective therapies. In this study, we investigated the therapeutic potential of nitroxoline (NTX) in treating cryptococcal infection. NTX exhibited potent fungicidal activity against Cryptococcus neoformans/gattii clinical isolates, with an MIC90 of 1 µg/mL. In addition, NTX substantially inhibits the main virulence traits of cryptoc

Cerebral Cryptococcoma Masquerading as High-Grade Glioma in an Immunocompetent Traveler from South America: A Diagnostic Challenge and Systematic Review.

INTRODUCTION: Cerebral cryptococcoma is a rare focal form of central nervous system (CNS) cryptococcosis, typically described in immunocompromised hosts. In immunocompetent individuals, it may present as a solitary mass lesion, closely mimicking a high-grade glioma and posing a significant diagnostic challenge, especially in returning travelers from endemic regions. CASE PRESENTATION: A 36-year-old previously healthy man from a rural region of South America presented with progressive left hemiparesis, headache, behavioral changes, and a generalized tonic-clonic seizure. Neuroimaging revealed

Immune Modulation and Evasion by Cryptococcus spp.: An Integrative Review of Molecular Mechanisms and Experimental Evidence.

The Cryptococcus species complex are major human pathogenic fungi capable of causing cryptococcosis, a severe pneumonia and meningitidis in both immunocompromised and, in some cases, immunocompetent patients. Their pathogenic success depends not only on classical virulence factors, but also on a coordinated capacity to evade, modulate, and exploit host immune responses across multiple stages of infection. In this review, we synthesize experimental evidence on the principal mechanisms that support cryptococcal immune evasion, integrating findings from molecular, cellular, and host-pathogen inte

A protective hybrid protein vaccine informed by mapping major histocompatibility class II epitopes in Cryptococcus neoformans chitin deacetylases that stimulate CD4 + T cell responses.

UNLABELLED: Infections due to Cryptococcus species are estimated to cause over 100,000 deaths a year. No cryptococcal vaccines are approved for human use. We have shown that vaccine formulations consisting of C. neoformans chitin deacetylase (Cda)2 and Cda1 adjuvanted with Cationic Adjuvant Formulation 01 (CAF01) protect mice against experimental cryptococcosis by a mechanism dependent upon CD4 + T cells. Here, using overlapping peptide libraries, we mapped epitopes in Cda2 and Cda1 that stimulated interferon-gamma (IFNγ) release from splenocytes of C57BL/6 and DR4 mice immunized with these pr

Cryptococcosis in Colombia: Analysis of Data from Laboratory-Based Surveillance 2017-2024.

Since 1997, a laboratory-based survey on cryptococcosis has been conducted in Colombia. We present the results for the period 2017-2024. A total of 891 surveys were received. The overall incidence was 0.22 cases per 100,000 people. Among those living with HIV, the incidence was 38, and among HIV-negative people, it was 0.08. Cryptococcosis demonstrated a higher prevalence among men than women (3.2:1). Among patients living with Human Immunodeficiency Virus (HIV), the condition primarily affected younger adults (26-40 years). In contrast, among HIV-negative people, it was mostly observed in old

[Disseminated cryptococcosis resulting in fungal septic shock: a case report].

Fungal sepsis and septic shock are syndromes involving acute organ dysfunction that result from a dysregulated host response to invasive fungal infections. These syndromes are characterized by diagnostic challenges, rapid progression, complex management, high medical costs, and high mortality, particularly among immunocompromised patients. This article mainly presents a case of fungal septic shock caused by disseminated Cryptococcosis, which resulted in death within a short period. It emphasizes the importance of the principle of "early warning and rapid intervention" in clinical practice for

18 F-FDG PET/CT in Disseminated Cryptococcosis Mimicking Malignancy With Rib Osteolysis and Chest Wall Invasion.

Disseminated cryptococcosis is a rare systemic fungal infection that can mimic thoracic malignancies. We report a 56-year-old man with known cryptococcosis on antifungal therapy who underwent 18 F-FDG PET/CT for response evaluation. Imaging revealed a heterogeneously FDG-avid pleural-based right upper lobe mass with lytic destruction of the first and second ribs and extension into chest wall musculature, with pulmonary nodules and nodal disease. Interval imaging demonstrated metabolic regression of adrenal and pulmonary lesions. This case highlights the diagnostic challenge of cryptococcosis p

Successful Long-Term Use of Isavuconazole in a Tacrolimus-Treated Japanese Kidney Transplant Recipient With Disseminated Cryptococcosis and Probable Invasive Aspergillosis.

Disseminated cryptococcosis, particularly cryptococcal meningitis, remains a severe fungal infection with high mortality in immunocompromised patients and usually requires prolonged azole therapy. In kidney transplant recipients receiving tacrolimus (TAC), careful management of drug-drug interactions via CYP3A is essential to maintain stable blood concentrations. We report the case of a 44-year-old man with a history of focal segmental glomerulosclerosis who had undergone a living-donor kidney transplant three years earlier. He was receiving immunosuppressive therapy with TAC, mycophenolate mo
