Sideroblastic anemias (SAs) represent a heterogeneous group of rare hematological disorders characterized by iron accumulation in mitochondria of erythroblasts with ineffective erythropoiesis. SAs are categorized into acquired and congenital forms. Acquired, secondary, and clonal, SA is rare in pediatric populations. Congenital SA (CSA) is classified into syndromic and non-syndromic forms. Herein, we describe three cases of pediatric patients with SA. The diagnosis of SA was based on the presence of type 3 sideroblasts on BM aspirate smear (greater than 15%) and genetic tests. In the first cas

Ionizing radiation exposure induces DNA damage and chromosome aberrations through both direct and indirect effect. The indirect effects are primarily mediated by the generation of hydroxyl radicals, a process attributed to radiation. Dimethyl sulfoxide (DMSO) and ascorbic acid (AA) are known as radical scavengers and have radioprotective effects. Radiation therapy is widely employed in the treatment of malignant tumors such as glioblastoma; however, its side effects, including cognitive impairments resulting from damage to healthy neurons, pose significant challenges. To ameliorate these effec

Sideroblastic anemias (SAs) represent a diverse group of congenital and acquired disorders, characterized by anemia and the presence of ring sideroblasts in the bone marrow. Congenital sideroblastic anemia (CSA) arises from genetic mutations that disrupt heme and iron metabolism within mitochondria. The most common form of CSA is X-linked sideroblastic anemia (XLSA), caused by mutations in the erythroid-specific aminolevulinate synthase 2 (ALAS2) gene, a key enzyme in the heme biosynthesis pathway in erythroid cells. On the other hand, the most common form of acquired SA is myelodysplastic syn

A man in his 60s with dry eye symptoms was noted to have angioid streaks under the peripapillary retina. Congenital dyserythropoietic anaemia was the major health issue throughout his life, requiring venesection for iron overload, but no transfusions for many years. Close inspection of the eyes with optical coherence tomography also detected small subclinical optic disc drusen, which have not been reported in association with congenital dyserythropoietic anaemia. Together, optic disc drusen and angioid streaks represent an ectopic calcification phenotype in the eye and can be seen in the more

A postpartum woman was diagnosed with congenital thrombotic thrombocytopenic purpura (TTP), triggered by pregnancy. She presented with severe thrombocytopenia, haemolysis, hypertension and neurological symptoms. Laboratory findings, including microangiopathic haemolytic anaemia, elevated lactate dehydrogenase (LDH) and a severely reduced ADAMTS13 activity, confirmed the diagnosis. Genetic testing identified a mutation in the ADAMTS13 gene. The patient was managed with plasma exchange (PLEX), fresh frozen plasma (FFP) and supportive care. After two sessions of PLEX, her platelet count increased

IntroductionMLASA (myopathy, lactic acidosis and sideroblastic anemia) is a rare, autosomal recessive mitochondrial disorder. Symptom onset typically occurs in childhood and differs considerably in disease severity. Congenital-onset disease is uncommon.MethodsWhole genome sequencing was performed which identified two heterozygous, rare YARS2 (NM_001040436.3) variants in trans, one of which was novel. The diagnosis was confirmed with muscle biopsy and mitochondrial enzyme activity testing. To compare the clinical phenotype of our patient to those previously described in the literature, we revie

Triosephosphate Isomerase deficiency (TPI-Df) is a devastating untreatable childhood metabolic disease resulting in anemia, severe locomotor impairment, and premature death. Numerous single amino acid substitutions in TPI are pathogenic and result in rapidly progressing multisystem disease. Importantly, all known pathogenic TPI-Df mutations result in a protein that retains function, and pathogenesis is known to result from decreased steady state levels of the functioning protein. There are no small molecule therapies for TPI-Df; current treatments are limited to symptomatic support and dietary

Background Pure red cell aplasia (PRCA) is an uncommon disorder characterized by severe normocytic normochromic anemia, reticulocytopenia, and an absence of erythroblasts from otherwise normal bone marrow. PRCA can present as either a congenital disorder or an acquired syndrome. Methodology A total of six cases of PRCA diagnosed in a tertiary care hospital from January 2016 to December 2019 were retrospectively studied. The diagnosis of PRCA was made based on bone marrow examinations showing erythroblastopenia with essentially normal myelopoiesis and megakaryopoiesis. Results Of the six patien

Introduction: Hereditary spherocytosis (HS) is a congenital hemolytic anemia, often under-recognized in neonates. Co-inheritance with other genetic disorders like Gilbert syndrome (GS) and beta-thalassemia trait (BTT) can complicate the diagnosis. Case Report: We report a neonate presenting with significant unconjugated hyperbilirubinemia and anemia. Genetic testing revealed a triple diagnosis- HS due to a heterozygous deletion in the SPTB gene, BTT with a splice-site variant in the HBB gene, and heterozygosity for UGT1A1 promoter polymorphism associated with GS. The father, previously diagnos

Hemoglobin A1c (HbA1c) is widely utilized for monitoring glycemic control in diabetes mellitus. Growth hormone (GH) therapy necessitates regular glucose monitoring due to an increased risk of diabetes. However, hemolytic anemia may result in spuriously low HbA1c levels due to a shortened erythrocyte lifespan, potentially leading to misinterpretation of glycemic status. We report an 11-year-old girl with GH deficiency undergoing GH therapy who presented with an abnormally low HbA1c level of 3.9% despite normal glucose tolerance. Laboratory tests revealed mild anemia (hemoglobin: 11.8 g/dL), ele

Triosephosphate isomerase (TPI) is a ubiquitously expressed enzyme encoded by the TPI1 gene. It catalyzes the interconversion of the triose phosphate isomers dihydroxyacetone phosphate and D-glyceraldehyde 3-phosphate in the fifth step of glycolysis. TPI deficiency (TPI Df; MIM# 615512) is an autosomal recessive disorder due to biallelic pathogenic variants in TPI1. In keeping with other glycolytic enzymopathies, severe hemolytic anemia is a common finding. Additionally, many individuals with TPI Df develop neuromuscular symptoms, which is unusual for a glycolytic enzymopathy. There appears to

Background/Objectives: Triosephosphate Isomerase (TPI) is a glycolytic enzyme known to be associated with TPI deficiency, a severe form of childhood-onset glycolytic enzymopathy associated with hemolytic anemia, neuromuscular impairment and early death. Most often the disease results from the common TPI1E105D mutation, which can be either homozygous or compound heterozygous with another allele. Methods: We purified TPIR5G protein, studied mutant protein biochemistry, established and characterized TPIR5G/f.s.patient cells, and investigated newly identified compounds for their efficacy in vitro

Pregnancy is a period characterized by extensive physiological changes in both the mother and fetus. During this period, the nutritional status of the mother has a profound and irreversible impact on her health and the growth and development of the fetus. The fetus depends exclusively on the mother and drives nutrients through the placenta. Therefore, mothers must be provided with a well-balanced diet that is adequate in both macro- and micronutrients. Most pregnant women generally manage to get adequate macronutrients; however, many women fail to get micronutrients up to the recommended dieta

BACKGROUND: Although healthcare providers (HCPs) know folic acid (FA), many are unaware of its various fetal-maternal-offspring beneficial effects.
METHODS: A cross-sectional study (September 2024) was conducted among HCPs using a pretested interviewer-based questionnaire. The total knowledge score was 30. The awareness score was good, average, and low for total scores >20, 11-20, and ≤10, respectively.
RESULTS: Out of 300 participants, 96 were excluded because of incomplete responses to the questions marked for outcome assessment. The participants were medical officers (42/206; 20.38%), nur

A total of 30 hydropic fetuses, including 25 cases from published reports and 5 from our own series, were reviewed, validated, and analyzed. This review yielded the following key findings: (1) Unlike most cases of nonimmune hydrops fetalis (NIHF), hydrops caused by syphilis is not only preventable but also treatable, with complete resolution possible when appropriately managed. (2) Syphilis-associated hydrops carries a poor prognosis if timely and appropriate treatment is not administered. (3) Based on limited data, intravenous penicillin G is probably more effective than intramuscular benzath

UNLABELLED: Patients with colon cancer can experience substantially different outcomes despite comparable anatomical stage, highlighting the need for molecular markers that complement conventional clinicopathological risk assessment. The interleukin-17 (IL-17) cytokine family has context-dependent roles in mucosal inflammation and tumour biology, but the prognostic relevance and cellular context of individual IL-17 receptors in colon cancer remain incompletely characterised. A prespecified family-wide analysis of 11 IL-17 ligands and receptors was performed in the GSE39582 Discovery cohort usi
