Three hydrophobic surface binding a proteins link surface hydrophobicity to sporulation, biofilm formation, and host interaction in Mucor lusitanicus.

INTRODUCTION: Mucor lusitanicus is a model organism for studying fungal development and physiology, as well as pathogenicity of Mucorales fungi. Hydrophobic surface-binding proteins (HsbA family) have been described in filamentous fungi as interface-associated factors involved in adhesion, enzymatic recruitment, and surface interactions; however, their functional diversification in Mucorales remains poorly understood. METHODS: Here, we present a comprehensive characterization of three HsbA proteins in M. lusitanicus. RESULTS: All examined HsbA proteins share a conserved α-helical fold with a

Confronting the "lethal duo" in the ICU: early identification of Aspergillus-Mucorales co-infection using a clinical-immuno-inflammatory signature.

BACKGROUND: Co-infection with Aspergillus and Mucorales in the intensive care unit (ICU) represents a devastating syndrome with high mortality that is frequently clinically occult. Clinically distinguishing this co-infection from invasive pulmonary aspergillosis (IPA) is challenging but critical for tailoring precise antifungal strategies. METHODS: We conducted a single-center, retrospective observational study involving 93 critically ill patients (75 with Aspergillus infection and 18 with co-infection) admitted between 2017 and 2025. We compared clinical characteristics, inflammatory markers

Management of invasive fungal infections in the intensive care unit.

INTRODUCTION: Invasive fungal infections (IFI) are becoming more and more frequent in the intensive care unit (ICU), affecting not only immunocompromised patients. Invasive pulmonary aspergillosis, invasive candidiasis, and mucormycosis are associated with substantial mortality. Management of these patients remains challenging because the histopathological confirmation is often unfeasible. Consequently, clinicians must rely on a probabilistic approach integrating host factors, clinical and radiological features, and mycological evidence. AREAS COVERED: This narrative review summarizes current

Treatment of pulmonary mucormycosis: current concepts.

INTRODUCTION: Pulmonary mucormycosis (PM) is a rapidly progressive angioinvasive fungal infection with high mortality. Despite therapeutic advances, optimal management remains uncertain. AREAS COVERED: We review the evidence on managing PM. We emphasize aggressive host factor optimization (glycemic control, immunosuppression reduction) and outline the evidence-based antifungal strategy: liposomal amphotericin B (L-AMB) for induction, followed by oral maintenance (posaconazole or isavuconazole). We discuss critical nuances, including LAMB dosing, timing, and duration of therapy, drug interacti

Pulmonary mucormycosis with bacterial coinfection in an adolescent with poorly controlled type 1 diabetes: a case report.

BACKGROUND: Pulmonary Mucormycosis (PM), a severe fungal infection affecting mainly immunocompromised individuals, is often caused by fungi like Rhizopus and Mucor. This report details a 12-year-old diabetic girl with pulmonary mucormycosis from an unusual Rhizopus species. Successful treatment involved stabilizing her blood glucose and managing multiple co-infections. This case provides important insights into diagnosing and treating rare fungal infections in diabetic children. CASE SUMMARY: A 12-year-old girl with a two-year history of type 1 diabetes, inconsistently monitored, was hospital

Liposomal amphotericin B in the treatment of pediatric gastrointestinal basidiobolomycosis.

BACKGROUND: Gastrointestinal basidiobolomycosis is a rare fungal infection caused by Basidiobolus ranarum, increasingly recognized as an intestinal pathogen in children. Its clinical features closely mimic malignancy or inflammatory bowel disease, leading to frequent diagnostic delays. Although endemic in regions such as Saudi Arabia, Iran, and Oman, sporadic cases occur worldwide. Amphotericin B-liposomal remains a cornerstone therapy for invasive fungal disease, but no standardized treatment protocol for pediatric gastrointestinal basidiobolomycosis has been established. CASE PRESENTATION:

Angioinvasive pulmonary mucormycosis presenting with massive haemoptysis secondary to pulmonary artery pseudoaneurysm.

Pulmonary artery pseudoaneurysm (PAP) is a rare but potentially fatal entity caused by a variety of underlying conditions. They may be congenital in origin or occur due to infective complications or occasionally secondary to pulmonary arterial hypertension. Among infective processes, tuberculosis and bacterial infections are the most common causative agents. There are few isolated reports of angio-invasive mucormycosis causing PAP; many of them were detected postmortem. Diagnosis of specific aetiology is often overlooked due to lack of clinical suspicion, as a result of incomplete investigatio

Transcriptomic and proteomic profiling of pulmonary mucormycosis reveals a failed activation of host immune response.

INTRODUCTION: Mucormycosis is one of the most life-threatening fungal infections with delayed diagnosis and limited antifungal treatments. The transcriptome and proteome of pulmonary mucormycosis have not been fully investigated. METHODS: We obtained lung tissues and paired controls from five pulmonary mucormycosis patients and utilized transcriptomic and proteomic approaches to explore host immune response during pulmonary mucormycosis. RESULTS: Our transcriptomic analysis found a number of up-regulated genes and pathways associated with immune defense. These genes were related to iron meta

Mucormycosis.

Mucormycosis is a rapidly progressive, invasive fungal infection that affects patients who are severely immunocompromised, as well as patients with diabetes and persons with immunocompetence who have major trauma. Mucormycosis manifests in several clinical forms, including sino-orbital, rhinocerebral, sinopulmonary, gastrointestinal, cutaneous, musculoskeletal, osteoarticular, and disseminated mucormycosis, as well as single-organ disease. Although mucormycosis is often lethal, early intervention reduces mortality. Successful treatment depends on early detection and staging of the disease, tim

[Comparative bronchoscopic manifestations in pulmonary mucormycosis and invasive pulmonary aspergillosis].

Pulmonary mucormycosis (PM) and invasive pulmonary aspergillosis (IPA) are life-threatening, opportunistic infections that are commonly seen in immunocompromised patients. Both are characterized by their tendency to invade the airways. This study retrospectively analyzed 46 PM patients and 41 IPA patients diagnosed at Beijing Tsinghua Changgung Hospital between January 2018 and February 2025, systematically evaluating their bronchoscopic features and proposing the morphological concepts of "mucormycelium", "aspermycelium", and "bronchial pulmonary cavity fistula". The results showed that mucor

Three consecutive cases of rhinocerebral mucormycosis in calves caused by Lichtheimia ramosa infection on a beef cattle farm.

Three Japanese Black calves died within a two-month period on a beef cattle farm. Necropsy consistently revealed liquefactive necrosis of the ventral brain (extending from the olfactory bulb to the midbrain) and thymic atrophy. Histopathologically, necrosuppurative encephalitis characterized by abundant fungal hyphae was observed in all three cases. Fungal lesions also involved the olfactory mucosa, olfactory nerves, and olfactory bulb. Fungi isolated from the cerebrum of all three calves were identified as Lichtheimia ramosa. These cases were diagnosed as rhinocerebral mucormycosis, originati
