Background/Objectives: New drugs are urgently needed for the treatment of neglected tropical diseases including leishmaniasis and eumycetoma, as well as globally occurring parasitic diseases such as toxoplasmosis. Fragrances, both natural and synthetic, were shown to be a rich source for the development of new anti-infectives and warrant deeper investigations. Exemplarily, we synthetically optimized the fragrance 4-(4,8-dimethyl-3,7-nonadienyl)-pyridine, a.k.a. Maritima, a pyridine derivative with marine odor. Methods: A new cationic N-cetyl-modified derivative of Maritima (dubbed Cetyl-Mariti

Mycetoma is a chronic granulomatous infection predominantly affecting young individuals, characterized by painless swelling and often misdiagnosed due to its resemblance to other conditions. This case report presents the first documented instance of eumycetoma located in the buttock of a 44-year-old male patient from Somalia, a region with limited literature on this disease. The patient exhibited a 10-year history of indurated swelling, which progressed to the discharge of dark granules, leading to the diagnosis of black grain eumycetoma. Comprehensive diagnostic methods, including imaging and

Background/Objectives: Eumycetoma, caused by Madurella mycetomatis, is a chronic fungal infection with limited treatment options and increasing drug resistance. CYP51, a key enzyme in ergosterol biosynthesis, is a well-established target for azole antifungals. However, existing azole drugs demonstrate limited efficacy in treating eumycetoma. Microbial-based natural products, with their structural diversity and bioactivity, offer a promising source for novel CYP51 inhibitors. This study aimed to identify potential Madurella mycetomatis CYP51 inhibitors from microbial natural products using mole

This study reports Madurella tropicana and Madurella pseudomycetomatis as pathogens causing eumycetoma in Senegal. The dominant species, Madurella mycetomatis, shares morphological features with both. Accurate species-level identification is crucial for treatment and epidemiology. Molecular diagnostic tools, especially ITS gene sequencing, enabled the identification of these species. Itraconazole treatment showed a favorable outcome in a patient infected with Madurella pseudomycetomatis, leading to sinus closure.

Nigrograna spp. have been reported as an emerging cause of black grain eumycetoma in immunocompromised patients. We report a unique case of Nigrograna chromolaenae, a plant pathogen previously unreported as a cause of cutaneous infection in a human host. A 71-year-old male with prior history of cardiac transplant and multiple comorbidities presented with a nodular hand lesion with continued progression. After initial antifungal agents were unsuccessful, the addition of posaconazole resulted in improvement of symptoms.

The WHO fungal priority pathogen list (FPPL), introduced in 2022, highlights 19 fungal species posing major public health risks. This systematic review evaluates the diversity and distribution of FPPL species in Indonesia, a tropical country favorable to fungal growth. Following Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, we searched PubMed, Scopus, Google Scholar, and Indonesian databases (2000-2025), identifying 79 eligible studies from 825 records. These covered clinical and environmental samples across 18 of 34 provinces. Medium-priority species

Invasive fungal infections (IFIs) are life-threatening affecting all age groups. Several studies on IFIs in Nigeria are documented, but reviews describing their occurrence in the paediatric population are lacking in the literature. We conducted a systematic review of the literature following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines. Using the PubMed database, we identified case reports, series, and observational studies reporting IFIs in the Nigerian paediatric population from inception to December 2024. We identified a total of 114 cases of IFIs from 3

INTRODUCTION: Mycetoma is a chronic disabling inflammatory disease characterized by the formation of granulomatous masses in subcutaneous tissue extending to the skin, deep tissues, and bones.
FINDINGS: An exhaustive and detailed exploration of medical literature unveiled only 11 published articles addressing mycetoma in Somalia since 1927. Among these, six were case reports involving 12 patients, and three were case series encompassing 163 patients, resulting in a total of 175 reported cases. Notably, two articles in French lacking abstracts were excluded from the review. Considering Somalia

Mycetoma is a chronic suppurative granulomatous infection of the deep dermis and subcutaneous tissue prevalent in tropical and subtropical regions and is caused by filamentous aerobic bacteria (actinomycetoma) or true fungi (eumycetoma), representing 60% and 40% of cases worldwide, respectively. The causative organism enters into the subcutaneous tissue, usually of the foot, from contaminated soil or vegetative material through inoculation from a thorn prick, or repeated trauma. It commonly affects men, farmers and field workers. Differentiating eumycetoma from actinomycetoma can be challengin

Mycetoma, or maduromycosis, is a chronic granulomatous infection involving the skin, subcutaneous tissues, and bones, most commonly affecting the lower extremities. Hand involvement is rare and often leads to diagnostic delays due to its atypical presentation. Mycetoma is classified as actinomycetoma (bacterial) or eumycetoma (fungal), each requiring distinct management approaches. We report a rare case of actinomycetoma affecting the right thumb in an 81-year-old man, initially misdiagnosed as a giant cell tumor. Magnetic resonance imaging revealed a well-defined subcutaneous mass without bon

Mycetoma is a chronic infection with a higher incidence in tropical and subtropical nations. We are presenting a rare case of mycetoma in a resident of Sudan, which is a temperate region, along with a short discussion. This case presentation discusses a 13-year-old male with a chronic foot discharge misdiagnosed as an abscess for two years. The condition, characterized by the development of a painless nodule leading to an open wound with purulent discharge, exemplifies the typical progression of actinomycetoma or eumycetoma. Following deep surgical debridement and antifungal therapy with itrac

Carcinoma cuniculatum (CC) is a rare and distinct clinicopathological variant of well-differentiated squamous cell carcinoma. It is a rare and slow-growing tumor with a peculiar infiltrative growth pattern resembling rabbit burrows (cuniculi). It usually occurs over the plantar aspect of the foot but can also occur at other sites like the oral cavity and genitals. The pathogenesis is unknown, with various hypotheses of trauma as proposed by different authors. It is essential to be aware of this entity as it commonly mimics benign and other low-grade squamous cell carcinoma (SCC). Diagnosis of

Eumycetoma, a chronic fungal infection caused by Madurella mycetomatis, is a neglected tropical disease characterized by tumor-like growths that can lead to permanent disability and deformities if untreated. Predominantly affecting regions in Africa, South America, and Asia, it imposes significant physical, social, and economic burdens. Current treatments, including antifungal drugs like itraconazole, often show variable efficacy, with severe cases necessitating surgical intervention or amputation. Drug discovery for eumycetoma faces challenges due to limited understanding of the disease's mol

Mycetoma is a chronic inflammatory disease characterized by progressive and devastating granulomatous subcutaneous tissue involvement. It primarily occurs in regions classified within the 'mycetoma belt', encompassing various countries. Notably, West African nations, including Burkina Faso (BF), are situated within this belt, yet there is a significant lack of data regarding mycetoma in these regions. With this background, this study set out to determine the mycetoma prevalence in BF. A comprehensive review was conducted to document the presence of mycetoma in BF. Searches were performed in Pu

OBJECTIVES: Eumycetoma is a neglected tropical fungal disease of the subcutaneous tissue that is currently not treatable with medication only. Standard itraconazole therapy is combined with surgical excision of the lesion. Recently, ibrexafungerp, a novel oral antifungal agent inhibiting 1,3-β-D-glucan synthesis, was approved by the United States Food and Drug Administration (FDA) for the treatment of vulvovaginal candidiasis. In this study we determined the in vitro activity and in vivo efficacy of ibrexafungerp in eumycetoma causative agents.
