Regional alignment with sex-typical brain architecture, maternal health and environmental enrichment predict psychiatric symptoms in early life.

Robust sex differences in brain morphology and function exist from birth onwards. While traditionally conceptualized as categorical, they were recently shown to be best described along a female-to-male continuum. Here, we probed the under-explored implications of this proposal for understanding early life vulnerability to psychiatric conditions with sex-biased prevalence. We analyzed environmental, clinical and resting state functional neuroimaging data from 411 participants in Developing Human Connectome Project (dHCP) and 214 children from the Oregon ADHD 1000 study. In the dHCP, we identifi

Preterm Birth Causes Decreased Postnatal Neurogenesis: A Novel Mechanism Underlying Adverse Neurodevelopmental Outcomes.

Although the survival rates of preterm infants have markedly improved, their neurodevelopmental outcomes remain poor. Historically, brain injury in preterm infants has been recognized as intraventricular hemorrhage or white matter injury, often resulting in cerebral palsy. More recently, increasing attention has focused on the elevated risk of neurodevelopmental disorders among preterm infants across a wide range of gestational ages. Furthermore, preterm birth is also associated with an increased risk of psychiatric disorders in adulthood, including depression and schizophrenia. However, the u

Thyroid hormone excess targeted to oxytocin neurons disrupts oxytocin physiology without impacting behavior.

Deficiencies in oxytocin are associated with neurodevelopmental and behavioral disorders, and the oxytocin gene (Oxt) can be regulated by thyroid hormone. We have previously shown that a systemic excess of thyroid hormone during development causes deficits in oxytocin that are associated with major abnormalities in mood-related and social behaviors. To determine whether the disruption in oxytocin physiology is responsible for these aberrant behaviors we targeted thyroid hormone excess to oxytocin neurons using a mouse model of conditional deficiency of the type 3 deiodinase (DIO3), the neurona

Inhibition of bone morphogenetic protein 4 signaling promotes resilience to depressive-like behavior in the offsprings induced by prenatal stress.

The offspring has different responses to maternal prenatal stress, some susceptible ones exhibit depression or anxiety-like behavior, and some ones demonstrate resilience and do not go on to develop behavior disorders. However, the mechanism underlying the resilience in the offspring to maternal prenatal stress remains unclear. In this study, we aimed to explore whether BMP4 signaling was associated with resilience to depression in the offspring following maternal prenatal stress, and further to explore whether inhibition of BMP4 signaling could promote resilience to depression in the offsprin

Maternal psychological distress, breast milk extracellular vesicles, and infant immunity: A translational study.

Distress during early postnatal life has a potential impact on maternal mental health and related biological systems. However, the potential biological roles of these systems' modifications remain largely unknown. Bronchiolitis represents a life-threatening condition for infants and a possible stressful event for mothers with an impact on maternal breast milk composition and potentially on infants'health. In this framework purposes of the present study were: a) to evaluate the impact of infants' severe bronchiolitis on maternal mental health; b) to examine the correlation between maternal ment

Trajectories of maternal bonding over the perinatal period: Associations with demographic, obstetric, psychological, and infant variables.

Biopsychosocial perspectives suggest that maternal bonding is shaped by various factors. Moreover, bonding may vary among different groups of women. This study aimed to identify trajectories of maternal bonding from late pregnancy to 12 months postpartum and explored whether these were associated with demographic, obstetric, psychological, and infant variables. Questionnaires were completed by 1216 Dutch women (Mage = 31.50) and obstetric data were collected. To estimate longitudinal trajectories, latent class growth analysis was performed. As anticipated, three distinct bonding trajectories e

Risk factors and long-term outcomes of infantile colic: A nationwide population-based study.

Infantile colic is a common condition during early infancy with unclear etiology, likely involving multifactorial mechanisms and gut dysbiosis. This nationwide population-based study investigated perinatal risk factors and long-term health outcomes associated with infantile colic. Using Taiwan’s national birth cohort data from 2010 to 2015, we identified 19,191 infants with colic and 95,955 matched controls. The inclusion criteria for the infantile colic group were having two or more related ICD codes (ICD-9-CM 789.0x and 780.9) between 1 and 5 months of age. Conditional logistic regression wa

Investigating the influence of maternal prenatal BMI and perinatal depressive symptoms on neonatal brain network dynamics.

BACKGROUND: Elevated pre-pregnancy body mass index (BMI) and perinatal depressive symptoms have been linked to neonatal alterations in brain structure and function. This study examined associations between neonatal functional brain dynamics, maternal BMI, and perinatal depressive symptoms measured by the Edinburgh Postnatal Depression Scale (EPDS) in a community-based, largely low-risk cohort. METHODS: Funcitonal MRI and Leading Eigenvector Analysis (LEiDA) were applied in a neonatal cohort (N = 437; 236 males; mean gestational age 39.6 weeks) from the developing Human Connectome Project. We

Low-frequency TMS ameliorates neonatal hypoxia-ischemia injury by normalizing glutamatergic transmission in penumbra.

Perinatal hypoxic-ischemic encephalopathy (HIE) is a leading cause of morbidity and mortality in term neonates. The current standard of care, therapeutic hypothermia, provides only partial neuroprotection. This study investigates the potential of low-frequency transcranial magnetic stimulation (LF-TMS) as a novel non-pharmacological adjunct therapy by targeting a key pathological mechanism of HIE: a persistent, pathological increase in glutamatergic synaptic transmission, or hypoxic long-term potentiation. Using a neonatal mouse model of hypoxia-ischemia, we administered a single session of LF

Prenatal, but not neonatal, paracetamol exposure disrupts neuroendocrine regulation of stress and emotional behavior in adult male rats.

Paracetamol is a widely used analgesic and antipyretic, commonly administered during pregnancy and early postnatal life. Its mechanism of action involves inhibiting cyclooxygenase-2/3 (COX-2/3) enzymes, which play a critical role in normal brain development during the perinatal period. In this study, we investigated anxiety-like behavior, depression-like behavior, and corticosterone levels in adult male rats following either prenatal or neonatal paracetamol exposure. Pregnant dams received subcutaneous injections of paracetamol (60 mg/kg) or saline every 12 h from gestational day (GD) 16 to 20

Study on the effects of gestational arsenic exposure on the developmental toxicity of brain tissue in mice offspring using network toxicology and RNA-seq.

BACKGROUND: Long-term arsenic exposure is associated with health risks, including neurotoxicity, cardiovascular diseases, and cancer, with heightened vulnerability during early development. This study integrates RNA sequencing (RNA-seq) and network toxicology to construct a competing endogenous RNA (ceRNA) network and elucidate molecular mechanisms of prenatal arsenic-induced neurodevelopmental toxicity. METHODS: Pregnant ICR mice were exposed to sodium arsenite (NaAsO2, 0.5 ppm) through drinking water. Behavioral tests were performed on postnatal day 30 (PND 30). A total of 20 offspring mice

Neonatal inflammation induces ventral hippocampal microglial priming via epigenetic regulation of BAG3 to enhance adult depression susceptibility in mice.

BACKGROUND: Neonatal inflammation increases the risk of adult depression, but the mechanisms remain unclear. Under the two-hit hypothesis, early-life inflammation may enhance vulnerability to later-life stress. Whether neonatal inflammation induces depression susceptibility via microglial priming, and the molecular basis of this process, remain unknown. METHODS: Neonatal mice (postnatal day 4) received intraperitoneal injections of lipopolysaccharide (LPS; 10, 50, or 100 μg/kg) or saline. In adulthood, mice were exposed to a subthreshold 2-week chronic unpredictable mild stress (CUMS) paradig

Coenzyme Q10 Mitigates Anxiety- and Depression-Like Behaviors in a Fetal Alcohol Spectrum Disorder Model by Suppressing Hippocampal Astrocytic Reactivity, Oxidative Stress, and Neuronal Cell Death.

OBJECTIVE: Fetal alcohol spectrum disorder (FASD) results from prenatal alcohol exposure and is associated with long-term neurobehavioral deficits, including anxiety and depression. Ethanol-induced oxidative stress, neuroinflammation, and hippocampal damage are key contributors to these outcomes. Coenzyme Q10 (CoQ10), a mitochondrial antioxidant, may offer neuroprotection against such ethanol-induced damage. This study aimed to investigate the potential therapeutic effects of CoQ10 on anxiety- and depression-like behaviors and hippocampal neurotoxicity in a neonatal rat model of FASD. METHOD:

The effects of early life pain and juvenile fear conditioning on CRF-receptor expression in the amygdala and hypothalamus of the juvenile rat.

Early life pain and stress have lasting consequences on nervous system development that can interact with later stress or trauma to create a susceptibility to fear, anxiety, depression and chronic pain among other psychological disorders. Recent work has identified changes in corticotropin releasing factor signaling in limbic system structures, such as the amygdala and hypothalamus, as a key mechanism behind these changes - albeit in a sex-dependent manner. CRF has two major receptors, CRFR1 and CRFR2 which have also been shown to play key roles in fear and pain expression. The current work ex

Maternal depression with and without a history of childhood maltreatment and newborn white matter microstructure.

Maternal depressive symptoms during pregnancy have consequences for offspring brain development, likely mediated via biological signals. However, gestational biological correlates of maternal depression may differ depending on childhood maltreatment (CM) history. We investigated the association of maternal depressive symptoms in pregnancy and CM history with newborn global white matter microstructure. In a sample of N = 90 mother-infant dyads from two cohorts, maternal depressive symptoms were assessed with the Edinburgh Postnatal Depression Scale. CM was assessed with the Childhood Trauma Que

Predictors of antepartum and postpartum suicidal ideation at a diverse urban obstetric setting.

BACKGROUND: Perinatal suicidality is a leading contributor to maternal mortality, and is associated with adverse outcomes for birthing individuals and children. Further research is needed to establish risk factors for perinatal suicidal ideation (SI), particularly among diverse populations. This study explored risk factors for perinatal SI within a diverse urban obstetric setting. METHODS: Data were derived from the electronic medical records of 3,983 perinatal individuals (41.8% Black/African American, 34.6% utilizing public health insurance) who completed the Edinburgh Postnatal Depression S

Repeated neonatal sevoflurane exposure causes adolescent anxiety/depression via microglial P2Y12R-Wnt/β-catenin pathway-mediated neurogenesis deficits.

Sevoflurane is the most commonly used inhaled anesthetic in pediatric and obstetric practice. However, early-life multiple sevoflurane exposure may increase long-term neurodevelopmental risks, though the underlying mechanisms remain incompletely understood. This study aimed to elucidate the role of the microglia-specific purinergic receptor P2Y12R in early-life multiple sevoflurane-induced developmental neurotoxicity. Using a neonatal mouse model of repeated sevoflurane exposure combined with behavioral tests, immunofluorescence staining, Western blot, qRT‑PCR, and microglia-specific condition

Quercetin mitigates CUMS-induced depression-like behaviors in rats by regulating argininosuccinic acid-induced astrocyte pyroptosis in the hippocampus.

Depression, or major depressive disorder, is a prevalent psychological disorder, but its underlying mechanisms remain understood, hindering effective prevention and treatment. This study investigated the protective effects of quercetin on chronic unpredictable mild stress (CUMS)-induced depressive-like behaviors and explored the underlying mechanisms using in vivo and in vitro experiments. Metabonomic analysis revealed that CUMS significantly elevated serum levels of argininosuccinic acid (ASA), which were strongly correlated with depression-like behaviors in rats, suggesting a potential role

The Important Role of miR-96 in Depression: New Research Progress and Future Prospects.

Depression is a debilitating mental illness recognized as one of the leading causes of disability worldwide, and its exact pathogenesis remains poorly understood. MicroRNAs (miRNAs) are endogenous small RNAs approximately 22 nucleotides in length that are involved in a range of important pathophysiological processes, including cell proliferation, apoptosis, differentiation, organ development, neurological diseases, and cancer. Given that several microRNAs are specifically expressed or enriched in the brain and central nervous system, accumulating evidence indicates that miRNAs are useful bioma
