Mapping Shroom3 expression across the adult mouse.

Shroom3 is an actin binding protein integral to apical constriction and apical-basal elongation during mammalian morphogenesis. Shroom3 function has been demonstrated in the development of the heart, neural tube, gut tube, eye, thyroid bud, and kidneys, with mutations linked to human congenital defects including anencephaly, spina bifida, cleft lip and palate, and ventricular septal defects. Genome-wide association studies implicate Shroom3 in human conditions such as chronic kidney disease and heterotaxy. While this suggests that Shroom3 expression continues postnatally, the extent of adult e

Anencephaly in Brazil: an ecological study on sociodemographic factors and prenatal care.

BACKGROUND: Anencephaly is a serious and fatal congenital anomaly. It can be detected in the first trimester of pregnancy through an ultrasound performed during prenatal care. This study aimed to identify maternal and prenatal care factors that predict anencephaly in Brazil. METHODS: This was an ecological observational study using secondary data on live births in Brazil between 2017 and 2022. The study variables were grouped into maternal sociodemographic factors (Brazilian regions, maternal age, marital status, schooling and race/color) and prenatal care (consultations performed, gestationa

Epidemiological insights into neural tube and orofacial malformations in Chile using data from the National Registry of Congenital Anomalies (RENACH).

BACKGROUND: Congenital anomalies affect approximately 6% of global births annually and contribute significantly to infant mortality and long-term disability. Low folate status has been linked to malformations such as neural tube defects (NTDs) and orofacial clefts (OFCs). This work aimed to estimate the national prevalence of these two groups of malformations in Chile, a country subjected to mandatory folic acid fortification of flour. We also analyzed the sex dimorphism of the NTD and OFC and described the main associated comorbidities. METHODS: In this cross-sectional descriptive study, we

Association between maternal occupation as a cleaner/maid/janitor during early pregnancy and selected birth defects in the National Birth Defects Prevention Study.

OBJECTIVES: Cleaning work involves potential exposures to physical, chemical, biological and psychosocial hazards, which might increase the risk of birth defects. Yet, there is limited research focused on maternal occupation in cleaning jobs and the risk of birth defects. We investigated the association between maternal occupation in cleaning-related jobs during early pregnancy and selected birth defects in the National Birth Defects Prevention Study, a case-control study from 1997 to 2011. METHODS: Mothers self-reported job details, which were classified by occupational epidemiologists into

Mechanosensor YAP orchestrates human neural rosette morphogenesis via TEAD4-LEF1 transcriptional nexus.

Mechanical signaling plays a crucial yet poorly understood role in human neural tube morphogenesis. In this study, we elucidate how the Hippo pathway mechanosensor YAP converts apical tension into transcriptional programs to guide this process. Using human cortical organoids, we demonstrated that YAP accumulates and translocates to the nucleus within high-tension apical domains of neural rosettes. YAP depletion disrupted apicobasal epithelial polarity, manifested as disorganized cytoskeleton, compromised tight junctions, and impaired ciliogenesis, which ultimately resulted in defective rosette

Trends and disparities in anencephaly-related early childhood mortality in the United States between 1999 and 2020: A nationwide CDC WONDER analysis.

Anencephaly is a severe neural tube defect that affects approximately 1 in every 5250 live births in the United States. This fatal congenital condition is linked to modifiable risk factors, including folic acid deficiency, maternal diabetes, and exposure to teratogenic medications such as certain anti-epileptics. Despite public health efforts to promote folic acid supplementation, disparities in mortality persist. This study aimed to evaluate national trends in anencephaly-related early childhood mortality from 1999 to 2020 using the CDC Wide-Ranging Online Data for Epidemiologic Research (CDC

Incidental Prenatal Discovery of Anencephaly via Portable Ultrasounds in a Remote Medical Care Setting.

Anencephaly is a fatal neural tube defect often linked to folic acid deficiency and undetected in resource-limited settings. We report a case in rural Uganda where portable ultrasound revealed anencephaly with polyhydramnios in a patient who has been pregnant six times, with one term delivery, four preterm deliveries, and no abortions, and currently has one living child (G6P1401). Early detection of this anomaly enabled surgical planning and maternal counseling before stillbirth. This case highlights the impact of malnutrition and the importance of portable ultrasound use to improve prenatal c

Risk factors of the appearance of anencephaly in Tunisia.

INTRODUCTION: Anencephaly is a serious developmental defect of the central nervous system in which the brain and cranial vault are grossly malformed. The cerebrum and cerebellum are reduced or absent, but the hindbrain is present. Anencephaly is a part of the neural tube defect spectrum. This defect results when the neural tube fails to close during the third to fourth weeks of development, leading to fetal loss, stillbirth, or neonatal death. AIM: To find out probable principal risk factors for the appearance of anencephaly. METHODS: This study was conducted to compare between pregnancies a

GPR161-GLI3 repressor signaling at cilia directs apical constriction and cell fate during cranial neural tube closure.

Failure to close the cranial neural tube, known as exencephaly/anencephaly, is a lethal congenital defect. However, the mechanisms driving patterning and reshaping of the broad cranial neural folds are poorly understood. Loss of the primary cilium-localized G protein-coupled receptor GPR161 causes ectopic, excessive hedgehog signaling in the mouse neural tube and fully penetrant exencephaly. GPR161 promotes GLI3 transcriptional repressor (GLI3R) formation while preventing GLI2 transcriptional activator formation. Here, we studied the mechanisms underlying cranial closure in mice using a Gpr161

Craniorachischisis Totalis: A Detailed Case Report.

Neural tube defects (NTDs) are severe congenital anomalies resulting from improper neural tube closure. Craniorachischisis totalis, the most extreme form, involves failure of neural tube formation along the entire cranio-spinal axis. This rare condition is fatal, with limited reported cases globally. We report a case of a 35-year-old G3P1L1A1 woman admitted at 20 weeks and 4 days gestation for medical termination of pregnancy following second-trimester ultrasound findings of anencephaly and spinal dysraphism. The patient began folic acid supplementation only after pregnancy confirmation. The f

Interstellar encounter: Postmortem imaging and virtopsy on a preserved anencephalic Indo-Pacific bottlenose dolphin specimen after 30 years.

BACKGROUND: Anencephaly is a deadly type of cephalic axial skeletal and neural disorder with a multifactorial aetiology that causes the failure of the rostral neuropore closure, compromising the formation of the neural folds, basicranium, and neurocranium. In cetaceans, there is only one report of this fetal abnormality, dated in late 1991, in a male stillborn Indo-Pacific bottlenose dolphin (Tursiops aduncus) and diagnosed using transabdominal ultrasonography on its mother that was kept under human care in the Ocean Park Corporation. After birth, physical and general radiographic examination

Critical appraisal on neural tube defects and their complexities.

Neural tube defects (NTDs), as a group of diseases, are congenital disabilities due to incomplete closure of the neural tube along its length, which otherwise forms the fully developed brain and spinal cord. An amalgamation of genetic, nutritional, and environmental factors plays a role in causing NTDs. They develop relatively early, within the first month of pregnancy-the time of neurulation, which could indicate that the pathogenesis of these diseases could stem from even pre-pregnancy causes like folic acid deficiency. This article provides an overview of the various etiology of NTDs and ho

Food Fortification Ineffective in Preventing Neural Tube Defects in India due to Regulation Promoting Inadequate Levels of Folic Acid and Vitamin B12.

BACKGROUND: There is a high prevalence of neural tube defects (NTDs) and nutritional anemias that increase the risk of birth defects in India. The current staple food fortification regulations for folic acid and vitamin B12 are ineffective in the country. We provide an evidence-based viewpoint urging for WHO-recommended levels of fortificants (i.e., 1300 mcg of folic acid and 10 mcg of vitamin B12 per kilogram of wheat flour or rice) to be included in the regulation. METHODS: Micronutrient fortified foods are being distributed through various governmental safety-net programs under the Nationa
