Variants in ACTB gene encoding for cytoplasmic β-actin result in a group of rare disorders called non-muscle actinopathies (NMA). A subgroup of NMA patients with ACTB pLoF (predicted loss-of-function) disorder have various congenital malformations, such as microcephaly and intellectual disability. We investigated the cellular effects of an ACTB pLoF missense variant, G302A, and a four-amino-acid deletion, S338-I341 in cytoskeletal β-actin, in patient-derived fibroblast cells. We found that neither of the mutations affected the organization of actin or the width of the actin-filament bundles, w

Most infants born small for gestational age (SGA) achieve spontaneous catch-up growth within the first two years of life; however, a subset remains growth-restricted and warrants further evaluation. Persistent short stature accompanied by microcephaly should prompt consideration of an underlying monogenic disorder. We report a term male infant born with severe SGA (birth weight -3.8 SDS; birth length -2.2 SDS) and microcephaly. At 16 months of age, his height was -2.7 SDS and head circumference -3.1 SDS. Initial insulin-like growth factor 1 (IGF-1) concentration was mildly decreased (-1.0 SDS)

Some patients with hyperkinetic movement disorders may present with a severe and prolonged state of uncontrolled chorea and/or ballism, termed 'choreic storm', causing distress and prompting intervention. These events may be more frequent and severe in patients with childhood-onset movement disorders. Deep brain stimulation (DBS) can be effective for medically refractory dyskinesias including chorea, and the availability of event-based local field potential (LFP) sensing in clinically available devices present an opportunity to better understand the underlying neurophysiology of these episodic

BACKGROUND: Global developmental delay (GDD) and intellectual disability (ID) are neurodevelopmental conditions with significant phenotypic and genetic heterogeneity. EEF1D (OMIM* 130592) has recently been identified as a gene of interest in neurodevelopmental disorders. Neurodevelopmental disorder with thin corpus callosum, hypotonia, and absent language (NEDTCHAL) (OMIM #621150) is caused by homozygous or compound heterozygous disease-causing variants in the EEF1D.
METHODS: We performed whole exome sequencing (WES) on an Iranian sibling presenting with global developmental delay, intellectu

Meier-Gorlin syndrome (MGORS) is a rare primordial dwarfism syndrome characterized by the triad of pre- and postnatal growth restriction, microtia, and patellar aplasia or hypoplasia. Pathogenic variants in genes involved in DNA replication licensing and initiation have been implicated in MGORS, including the uncommon disease-associated gene CDC6, for which only two affected families have been reported to date. We report a male infant born at 37 weeks' gestation to a 40-year-old primigravida via cesarean delivery due to intrauterine growth restriction and fetal intolerance of labor. He present

Angelman syndrome is a severe neurodevelopmental disorder caused by the loss of UBE3A, a gene that encodes an E3 ubiquitin ligase1,2. The mechanisms that underlie Angelman syndrome are not well understood, and in particular, the link between UBE3A substrates and disease phenotypes remains uncertain3. Here, using gain- and loss-of-function mutations in UBE3A4,5 along with unbiased proteomics, we show that asparagine synthetase (ASNS) is a target of UBE3A. We find that ASNS protein abundance is increased in the brains of Ube3a-null mice and in human neurons6. Genetic correction of ASNS overabund

T-complex protein 1 (TCP1) encodes the TCP1α subunit, one of eight subunits of the chaperonin containing TCP1 ring complex (TRiC). Heterozygous TCP1 variants cause autosomal dominant neurodevelopmental disorder characterized by developmental delay or intellectual disability, seizures, pyramidal or cerebellar signs, and brain malformations, particularly polymicrogyria. However, the reported clinical spectrum remains limited. We report a 4-year-old girl with global developmental delay, speech and motor delay, behavioral dysregulation, microcephaly, hypotonia with core weakness, and a previously

Humans differ remarkably from great apes, with advanced cognitive abilities correlated to enlarged brains resulting from prolonged neurodevelopment. The SINE-VNTR-Alu (SVA) retrotransposons are candidate genetic drivers of these differences. Human genes with intronic SVAs are enriched for neurodevelopmental diseases; the microcephaly gene CDK5 regulatory subunit associated protein 2 (CDK5RAP2) and epilepsy/intellectual disability gene SCN8A coding for sodium channel subunit contain human-specific, intronic SVAs. Deleting gene ZNF91 coding for the SVA-binding zinc finger protein 91 or the intro

Excessive telomere shortening contributes to pulmonary fibrosis (PF), with at least 30% of familial cases attributed to telomere biology disorders (TBDs). However, TBD-associated PF remains underdiagnosed because of unidentified disease-causing mutations and limited referral for telomere and genetic testing. Here, we combined telomeric restriction fragment analysis, telomere analysis by a long-read Nanopore sequencing method (NanoTelSeq), exome sequencing (ES), and optical genome mapping (OGM) to investigate the underlying cause of disease in a family affected by PF, microcephaly, and prematur

Triple A syndrome (AAAS) is a rare autosomal recessive neuroendocrine disorder caused by pathogenic variants in the AAAS gene and characterized by marked clinical heterogeneity. In this multicenter retrospective study, we analyzed 23 patients from 18 families diagnosed across nine centers in Türkiye to characterize the molecular and clinical features of AAAS. Six distinct vari-ants in the AAAS gene were identified in this cohort, with truncating variants predominating. The most frequent variant was c.1066_1067delCT (p.Leu356Valfs*8), detected in 7 of 18 families, followed by the nonsense varia

Triphenyltin hydroxide (TPTH) is a widely used agricultural fungicide and environmental pollutant well known for its potent toxicological potential. Despite this, its embryotoxic effects remain unexplored in avian models. This study presents a novel evaluation of TPTH's acute toxicity, genotoxicity, cytotoxicity, teratogenic potential, and enzymatic and tissue architectural potential using the in ovo chick embryo model. Fertilized chicken eggs were exposed to varying TPTH concentrations that further revealed a time-dependent median lethal dose (LD50) decline from 547.37 µg/egg at 24 h to 395.7

The PIKfyve-FIG4-VAC14 lipid kinase complex generates phosphoinositide PI(3,5)P2. Pathogenic variants in VAC14 and FIG4 cause neurodevelopmental disorders and PIKFYVE has been linked to autosomal dominant fleck corneal dystrophy. We searched for the underlyging, genetic cause of a neurodevelopmental disorder in five affected individuals from three families using exome- or genome sequencing. Cranial MRI images and clinical phenotypes were analyzed. Size, number and acidity of cellular vacuoles were investigated in patient-derived fibroblasts from family 1 using LysoSensor acidophilic probes. Le

BACKGROUND: Canopy FGF signalling regulator 3 (CNPY3) is an endoplasmic reticulum (ER)-resident chaperone essential for toll-like receptor (TLR) folding, trafficking and inflammasome activation. In rare paediatric cases, biallelic CNPY3 variants have been associated with developmental and epileptic encephalopathy (DEE), but available data remain sparse.
METHODS: 50 individuals from 38 unrelated families with biallelic CNPY3 variants and DEE were evaluated for exome and genome findings, standardised clinical phenotype descriptions, EEG results, and neuroimaging data. A selection of variants un

UNLABELLED: ANKLE2 is a scaffolding protein with crucial roles in neuroprogenitor cell division and embryonic brain development. Pathogenic variants in ANKLE2 cause primary microcephaly, a congenital disorder characterized by impaired neurodevelopment. Nonetheless, the underlying dysfunction of ANKLE2 is not understood. Here, we define the ANKLE2 protein interaction landscape, which includes cell division proteins and novel microcephaly candidates in the APC (anaphase-promoting complex). Analysis of six pathogenic variants reveals changes in this interactome that likely result in microcephaly.

BACKGROUND: Dimenhydrinate (DH) is the first generation medicine used as antiemetic. It is neurotoxic in rat and chick embryos. Both green cabbage (GCbg) and red cabbage (RCbg) have many health benefits and red cabbage is also full of anthocyanins and may minimize oxidative stress in chick embryo.
MATERIAL AND METHOD: The study was designed to check the teratogenic impacts of dimenhydrinate and curative abilities of green and red cabbage extracts against dimenhydrinate in chick embryo through morphometry, histopathology and micrometry. Fertilized eggs were divided into six groups (n = 50): (1
