PURPOSE: We aimed to broaden the understanding of autosomal recessive neurodevelopmental disorders caused by VARS1 by describing new clinical and molecular findings and assessing the predicted structural impact of identified variants.
METHODS: We clinically evaluated 13 affected individuals from 10 unrelated families presenting with a neurodevelopmental disorder. We used exome sequencing and cosegregation analyses to identify disease-causing variants, followed by three-dimensional in silico analyses and molecular dynamics simulations to assess the likely functional consequences of both previo

BACKGROUND: Congenital hypothyroidism (CH) is a preventable cause of growth and neurodevelopmental deficits. In India, lack of universal newborn screening (NBS) delays diagnosis. This study examines how treatment timing influences growth, nutrition, and body proportionality in children with CH.
METHODS: This cross-sectional study included 66 children with CH, aged under 5 years, receiving Levothyroxine(LT4) therapy for one year. Age and sex-matched 64 healthy children were also enrolled. Children with CH were stratified into early-treated (<3months) and late-treated (>3months) groups. Anthrop

RNF113A-related disorders are rare X-linked conditions characterized by neurodevelopmental impairment, endocrine abnormalities, and gonadal dysgenesis. To date, this condition has been documented in only a few individuals. We report 46,XY twin siblings with a hemizygous pathogenic RNF113A variant [c.901C > T, p.(Gln301*)], presenting with a remarkably consistent phenotype that includes microcephaly, corpus callosum dysgenesis, intractable epilepsy, subclinical hypothyroidism, microphallus, and bilateral testicular regression syndrome (TRS). Whole-exome sequencing identified a maternally inheri

YIPF5 is an ER-membrane protein implicated in ER-Golgi transport. Mutations in YIPF5 cause MEDS2 (microcephaly, epilepsy, and neonatal diabetes syndrome), a fatal disorder manifesting in early childhood. We demonstrate that YIPF5 is involved in ER export of a subset of proteins, including cargoes of the ER export receptor SURF4, with which it directly interacts. YIPF5 knockout cells display altered cell surface and secretome profiles, with reduced neuronal adhesion molecules and increased secretion of ER chaperones affecting migration. YIPF5 depletion enhances cell migration in a wound-healing

RTTN, encoding the centrosomal protein rotatin, has been reported in about 40 patients with mild to severe microcephaly, intellectual deficiency and facial dysmorphism. The most recurrent variant in this gene is c.2953A > G, identified at the homozygous state in 4 families. Skin fibroblasts were obtained from a 22-month-old female patient, and induced pluripotent stem cells (iPSC) were generated using episomal expression of Yamanaka's factors. This iPSC line presents classical morphology, pluripotency markers expression and can successfully differentiate into the three germ layers. This line r

Mutations in the ATRX gene are a primary cause of alpha-thalassemia intellectual disability X-linked (ATRX) syndrome, which is characterized by intellectual disability, autism, and a range of brain structural abnormalities, including microcephaly. We previously showed that mice with conditional ATRX ablation in forebrain excitatory neurons display deficits in fear memory and autism-related behaviors, with some effects exhibiting sexual dimorphism. In this study, we used high-resolution magnetic resonance imaging (MRI) to systematically characterize brain structural changes associated with thes

The construction of the human brain is dependent on neuronal progenitors that divide accurately generating billions of neurons. This neurogenic potential requires the efficient assembly of the mitotic spindle, which mediates the alignment of sister chromatids and their subsequent separation. The mitotic spindle is formed from microtubules, which are decorated with proteins that alter the functional repertoire of the cytoskeleton. Mutations in EB2, a member of the end binding protein family, have been identified in individuals with multiple congenital defects including microcephaly. Here, we co

The current outbreak of the emerging arthropod-transmitted Oropouche virus (OROV) in South America has been epidemiologically linked to vertical transmissions, microcephaly, and stillbirths. Nevertheless, the impact of OROV infection during pregnancy has not been experimentally evaluated. To address how OROV infection might impact pregnancy outcome, we performed experiments in human cells and mice. Studies in cell cultures showed that the human trophoblast cell lines BeWo and JEG-3 are permissive to OROV infection (strain BeAn19991) and develop type I interferon (IFN)-dependent antiviral respo

Neurodevelopmental disorders (NDDs) often remain unexplained due to limited assessment of non-coding genomic elements. Motivated by recent reports implicating RNU4-2, which encodes a spliceosomal small nuclear RNA (snRNA), we analyzed whole-genome sequencing data from 15 450 Korean individuals, including 2797 unrelated NDD probands. Rare pathogenic RNU4-2 variants were identified in 20 probands (0.72%), including 17 (85%) with a recurrent n.64_65insT variant. RNA secondary structure modeling and molecular dynamics simulations demonstrated that n.64_65insT disrupts the U4/U6 snRNA duplex and im

Nardilysin (NRDC) plays a role in multiple cellular functions in diverse cellular compartments, including ectodomain shedding in the plasma membrane, as well as chaperoning a key Krebs cycle enzyme in mitochondria. We had previously reported limited clinical information from two individuals with homozygous frameshift variants in NRDC. With inclusion of previously published individuals, here we report 14 individuals (10 females, four males) from nine unrelated families carrying homozygous NRDC pathogenic variants. Common clinical features include severe to profound developmental delay/intellect

INTRODUCTION: Claes-Jensen syndrome is a rare X-linked syndromic neurodevelopmental disorder by pathogenic variants in lysine specific demethylase 5C (KDM5C), a lysine-specific histone demethylase.
METHODS: In this study, clinical evaluations were conducted in affected individuals and carrier females. X-chromosome inactivation (XCI) assays were performed to assess genotype-phenotype correlations. Functional studies evaluated variant effects on RNA transcription, protein expression, and stability. Zebrafish models were used for in vivo validation. RNA sequencing with KEGG and GO analyses ident

Recurrent hypothermia is an uncommon but clinically significant problem in adults with prior traumatic brain injury (TBI), such as shaken baby syndrome (SBS). Central thermoregulatory dysfunction, due to hypothalamic or autonomic injury, can lead to episodic hypothermia that mimics endocrine pathology. We describe a 38-year-old woman with cerebral palsy (CP) and a remote history of SBS presenting with recurrent hypothermia despite normal thyroid and adrenal function. Her episodes were initially treated empirically with corticosteroids, given concern for adrenal involvement, but recurrent event

RATIONALE: Combined oxidative phosphorylation deficiency 35 (COXPD35) is an extremely rare mitochondrial disorder inherited in an autosomal recessive pattern. It results from pathogenic variants in the TRIT1 gene, leading to hypomodified cytosolic and mitochondrial tRNAs. We report the first identified case of COXPD35 in Palestine, resulting from 2 novel variants of the TRIT1 gene.
PATIENT CONCERNS: A 2-year-and-6-month-old female patient with seizures, neurodevelopmental delay, microcephaly, dysmorphic facial features, abnormal electroencephalogram (EEG), and thinning of the corpus callosum

Herbicide effectiveness depends on the adjuvants in the formulation, with surfactants being crucial for systemic ones such as glyphosate, but surfactants are often more toxic than the pesticide itself to nontarget organisms, such as amphibians. Previously, a novel experimental formulation (EF) of glyphosate has been developed with natural products (alkylglucosides and saponin) with great efficiency in field tests. The objective of this study was to compare the toxicity of the EF with a commercial formulation of glyphosate (Roundup Full II, CF) by standardized bioassays (AMPHITOX). For this, em

Inherited mutations in the spindle pole-associated scaffold protein WDR62 cause autosomal recessive primary microcephaly. Previous research has characterised the roles of WDR62 in the regulation of spindle dynamics, cell division, and brain development. Here, we identify a new function of this protein in regulating purine metabolism. WDR62 interacts directly with BAG2, a co-chaperone of HSP70/90. Under stress conditions, WDR62 and BAG2 re-localise to cytoplasmic granules enriched for enzymes involved in purine synthesis (PFAS) and salvage (HPRT). In WDR62-deficient cells, purine synthesis is i

Interstitial deletions involving 3q27.1 define a distinct microdeletion syndrome characterized by prenatal-onset growth restriction, postnatal microcephaly, hypotonia, intellectual disability, and distinctive craniofacial features. While AP2M1 haploinsufficiency has been proposed as the primary driver of this phenotype, the full spectrum of dosage-sensitive genes within the locus remains unclear. Here, we report a patient with a de novo heterozygous 3q27.1 microdeletion and delineate a refined minimal smallest region of overlap (SRO) of approximately 189 kb, representing the narrowest critical

Sotos syndrome is an overgrowth disorder caused by nuclear receptor binding SET domain protein 1 (NSD1) haploinsufficiency, whereas reciprocal 5q35.2q35.3 microduplication produces a reversed phenotype with growth retardation, microcephaly, delayed bone age, and neurodevelopmental delay. We describe a 43-month-old Korean girl born at 36 + 1 weeks with intrauterine growth restriction (birth weight 2200 g, length 46 cm, occipitofrontal circumference 42 cm). At presentation, all growth parameters were below the 10th percentile, with dysmorphic features including epicanthal folds, telecanthus, and
