BACKGROUND: De novo mutations in transcriptional regulators are emerging as key risk factors contributing to the etiology of neurodevelopmental disorders. Human genetic studies have recently identified ZMIZ1 and its de novo mutations as a cause of a neurodevelopmental syndrome strongly associated with intellectual disability, autism, attention-deficit/hyperactivity disorder, microcephaly, and other developmental anomalies. However, the role of ZMIZ1 in brain development or how ZMIZ1 mutations cause neurological phenotypes is unknown.
METHODS: We generated forebrain-specific Zmiz1 mutant mice

PURPOSE: COPB1 encodes the coatomer subunit beta protein, which is essential for brain development and intracellular protein trafficking. Homozygous mutations cause Baralle-Macken syndrome that characterized by global developmental delay, severe intellectual disability, and early-onset cataracts. Although immunodeficiency has been observed in patients with COPB1 deficiency, the immunological phenotype remains incompletely characterized. Here, we comprehensively describe the clinical features and delineate the immunological phenotype associated with COPB1 mutations.
METHODS: We performed detai

Pathological CGG expansions in the FMR1 gene, encompassing premutations (55–200 repeats) and full range mutations (> 200 repeats), cause a spectrum of complex and incurable disorders. Premutation carriers (PMC), particularly men, face the risk of developing fragile X-associated tremor/ataxia syndrome (FXTAS). Other conditions associated with premutations in FMR1 include both Fragile X-associated primary ovarian insufficiency (FXPOI), and Fragile X-associated neuropsychiatric disorders (FXAND). Hyperintensities in the middle cerebellar peduncle (MCP), known as the MCP sign, have been considered

Aicardi syndrome (AIC) is a congenital condition involving characteristic neurological and ocular abnormalities. We describe a 30-year-old pregnant woman referred at 32 weeks of gestation after an ultrasound at 29 weeks showed corpus callosum agenesis, an interhemispheric arachnoid cyst, and colpocephaly. These findings raised prenatal suspicion of AIC. Postnatal evaluation confirmed the diagnosis with the identification of chorioretinal lacunae on fundoscopic examination. In this case, the prenatal imaging findings were key to recognizing a pattern suggestive of AIC, while postnatal assessmen

To date, only two families with variants in the IQSEC1 gene associated with intellectual developmental disorder with short stature and behavioral abnormalities (IDDSSBA) have been described. Here, we report an 8-year-old boy with short stature, speech delay, dysmorphic facial features, hypotonia, and behavioral disorders, as well as corpus callosum dysgenesis associated with compound heterozygous variants Pro1095ArgfsTer97 and Thr485Met in the IQSEC1 gene. To our knowledge this is the first report of brain anomalies associated with IQSEC1 variants, highlighting the need for MRI in affected pat

Here, we propose a new concept linked to neuroplasticity and cognitive reserve: 'Connectomic reserve'. It is suggested and discussed as a hypothesis, aiming to stimulate experimental testing, taking as an example the exuberant circuits formed during development by commissural neurons and their projections. Upon reviewing the developmental strategies of the formation of callosal circuits, from axonal emission by cortical cell bodies to the choice of trajectories within the ipsi and contralateral hemispheres, we identified the formation of many ectopic and heterotopic projections. Ectopic projec

The corpus callosum is the largest white matter tract in the brain, connecting the left and right cerebral hemispheres. Callosal agenesis is a condition where this tract fails to form during neurodevelopment. Despite this, individuals with callosal agenesis can present as cognitively and behaviorally asymptomatic, retaining some alternate form of interhemispheric communication. Afferent and efferent pathways linking the cortex to the contralateral cerebellum, such as the corticopontocerebellar tract (CPCT) and dentatorubrothalamic tract (DRTT), could serve as compensatory routes in individuals

INTRODUCTION: Spina bifida aperta (SBA) is frequently associated with a wide range of structural anomalies in the brain, some of these thought to be related to epilepsy. However, the relationship between these is poorly studied. This study aims to assess the prevalence of brain anomalies and epilepsy in SBA patients and explore the nature and magnitude of their association.
METHODS: We conducted a retrospective cross-sectional cohort study including all consecutive postnatally treated SBA patients, managed at Erasmus MC Sophia Children's Hospital, Rotterdam from January 1st 2000 up to June 1s

INTRODUCTION: Congenital arachnoid cysts (ACs) are cerebrospinal fluid collections resulting from abnormal splitting of the arachnoid membrane during embryogenesis. They may occur as isolated lesions or in association with other central nervous system (CNS) malformations. Congenital cytomegalovirus (cCMV) infection is a leading infectious cause of neurodevelopmental impairment and may contribute to complex CNS anomalies.
PRESENTATION OF CASE: A term male infant was diagnosed prenatally at 30 weeks with midline cystic lesions, ventriculomegaly, and absent corpus callosum. Postnatal magnetic re

PTEN hamartoma tumour syndrome (PHTS), a rare disease caused by germline heterozygous PTEN variants, is associated with multi-organ/tissue overgrowth, autism spectrum disorder and increased cancer risk. Phenotypic variability in PHTS is partly due to diverse PTEN variants and the protein's multifaceted functions. PTEN is primarily a phosphatidylinositol(3,4,5)trisphosphate (PIP3) phosphatase regulating PI3K/AKT signalling but also maintains chromosomal stability through nuclear functions such as double-stranded (ds)DNA damage repair. Here, we show that PTEN-R173C, a pathogenic variant frequent

OBJECTIVES: Our aim is to define the microstructural organization of the corpus callosum (CC) in people with congenital mirror movements (CMM). We hypothesised that smaller fractional anisotropy (FA) values will be found in our CMM sample compared to the control group.
METHODS: Diffusion tractography (DTI) was applied to quantify CC microstructure in 46 neurologically-normal controls and four individuals with CMM characterised by pathogenic variants known to interact with netrin-1 during embryonic development of axonal pathfinding processes. FA was quantified across 80 nodes within each of 8

Monoallelic pathogenic variants consistent with Mendelian inheritance patterns causing congenital heart disease (CHD) have been increasingly identified as genetic testing, including genome sequencing (GS), has become more widely available within the clinical space. Here, we focus on a newly described pathogenic variant in CDH2 resulting in Agenesis of Corpus Callosum (ACC), Cardiac, Ocular, and Genital Syndrome (ACOGS). While previous studies document variable expressivity of the CDH2 variants in unrelated individuals, no cases have described such variable expressivity in twins with the same C

RATIONALE: Opioid addiction, including morphine use, is a major public health crisis in the U.S. It has been associated with brain volume changes in reward-related regions, neuronal loss, and neuroinflammation. While these alterations have been studied separately, it remains unclear whether structural changes co-occur with microglial adaptations at early stages of morphine use.
OBJECTIVE: This study aimed to examine region-specific brain volume changes, cellular counts, and the emergence of distinct microglial phenotypes in addiction-related regions, using a model of morphine self-administrat
