WZB117-Induced Glucose Hypometabolism Triggers Mitochondrial Dysfunction and Amyloidogenic Processing in an In Vitro Primary Forebrain Neuron Model.

Background Alzheimer's disease (AD) is a progressive neurodegenerative disorder in which cerebral glucose hypometabolism represents one of the earliest and most consistent pathological abnormalities, often preceding classical amyloid and tau pathology. Despite strong clinical evidence, the causal contribution of impaired neuronal glucose utilization to AD-related cellular alterations remains incompletely understood. Objective This study aimed to determine whether selective inhibition of neuronal glucose transport induces AD-relevant cellular stress responses, including mitochondrial dysfunctio

A Stepwise Approach to Macrocephaly: Clinical Clues to the Rare Diagnosis of PTEN Hamartoma Tumor Syndrome.

Macrocephaly, defined as a large head size, has a very broad differential and sometimes can be challenging to differentiate as benign versus indicative of pathology. In this review, we outline a stepwise approach to improving diagnosis of neurogenetic disorders versus other causes using the example of PTEN (phosphatase and TENsin homolog) Hamartoma Tumor Syndrome (PHTS). PHTS is a multiple hamartoma syndrome with medical management implications including the need for tumor surveillance, but often cases are not diagnosed until later in adulthood. Our review emphasizes the utility of using head

Genotypic and phenotypic heterogeneity in tubulinopathies: insights from a Turkish multicenter cohort.

BACKGROUND: Tubulinopathies encompass a clinically and neuroradiologically diverse group of neurodevelopmental disorders caused by mutations in genes encoding tubulins and microtubule-associated proteins. This study aims to delineate the extensive phenotypic and genotypic spectrum within a pediatric cohort. METHODS: We conducted a multicenter retrospective analysis of 15 pediatric patients from 12 unrelated families with genetically confirmed tubulinopathies across five tertiary centers in Turkey. Clinical data, seizure profiles, and neuroradiological findings were systematically evaluated.

Maternal Fish Intake in the Year Prior to Conception and Birth Defects, National Birth Defects Prevention Study, 1997-2011.

BACKGROUND: Epidemiologic data on the association between maternal fish intake and birth defects are sparse. Our objective was to assess associations between maternal fish intake and 52 different birth defects, most of which have not been assessed previously. METHODS: Using logistic regression, data was analyzed for 29,242 mothers of infants with birth defects and 10,973 mothers of control infants who participated in the National Birth Defects Prevention Study (NBDPS) and delivered between 1997 and 2011. We focused on associations between mothers who reported high fish intake of 2 or more ser

Postmortem MRI reveals distinct structural features in sudden unexpected death in epilepsy.

Magnetic resonance imaging (MRI) abnormalities have been reported in individuals who later die from sudden unexpected death in epilepsy (SUDEP), but their specificity and predictive value remain uncertain. Postmortem MRI (PM-MRI) offers a unique opportunity to distinguish structural features associated with SUDEP from changes related to epilepsy, comorbid illness, or the postmortem interval. We performed PM-MRI in nine individuals: five with suspected SUDEP, two with epilepsy who died from non-seizure-related causes, and two without epilepsy who died from sudden cardiac death. TTC14 dysfunction contributing to microcephaly and lissencephaly spectrum features through protein mislocalization and impaired RNA processing.

Congenital microcephaly and lissencephaly spectrum disorders are characterized by disrupted neuronal proliferation and migration, often driven by underlying genetic variants. Here, we identified and characterized of a novel gene (Tetratricopeptide Repeat Domain 14, TTC14) and its homozygous missense variant (c.89 A > G; p.His30Arg (p.H30R)) in a female proband presenting with microcephaly, epileptic spasms, global developmental delay, and neuroimaging features of simplified gyral pattern, focal pachygyria, and corpus callosum thinning. Trio-based whole exome sequencing revealed this variant to

Malformations of Cortical Development.

Malformations of cortical development are complex and diverse. They are an etiology for drug-resistant epilepsy, and their imaging presentation may be striking or quite subtle. There are multiple schemes to classify this pathology group. One of the most well known is to group disorders based on the type of embryologic error that leads to each malformation. They can also be grouped based on imaging appearance, with the morphology of the involved cerebral cortex acting as the first major distinction.

Developmental Delay and Macrocephaly Unraveling a Leukodystrophy: A Case Report.

An uncommon inherited autosomal recessive disorder, Van der Knaap disease is also referred to as megalencephalic leukoencephalopathy with subcortical cysts (MLC). Megalencephaly, which either develops at birth or during infancy, along with seizures and a mild motor development delay, is its defining characteristic. Leukodystrophy and subcortical cystic degeneration are the two MRI abnormalities that are indicative of the illness and typically provide the key to diagnosis. The disease is more common in certain ethnicities, like the Aggarwal community in India, where marriages within the communi

A rare case of hereditary spastic paraplegia with homozygous mutation in the ZFYVE26 gene.

Hereditary spastic paraplegia (HSP) refers to a group of rare, progressive neurodegenerative disorders characterized by spasticity and weakness predominantly affecting the lower limbs, with significant clinical and genetic heterogeneity. Clinically, HSP is classified into "pure" and "complicated" forms. SPG15 (ZFYVE26) is a rare autosomal recessive subtype typically presenting in childhood with cognitive decline, followed by the development of gait disturbances and spasticity, often accompanied by thinning of the corpus callosum. Here, we report the case of a 19-year-old female patient, born t

The hedgehog co-receptors cdon and boc function redundantly to regulate zebrafish craniofacial development.

BACKGROUND: Craniofacial development is a complex developmental process that involves formation of the skeleton, muscle, and tendons. Defects in craniofacial development result in common hereditary disorders. Among genetic factors regulating craniofacial development, dysregulated hedgehog signaling is associated with craniofacial skeletal defects such as orofacial clefting and holoprosencephaly. Here, we characterize craniofacial phenotypes associated with two hedgehog signaling co-receptors, cdon and boc, in zebrafish. Genetic pedigree analyses have previously linked both cdon and boc to micr

Microglia depletion alleviates the disruptions in circadian rhythms and anxiety caused by bmal1 deficiency.

Microglia, the intrinsic immune cells of the brain, play a crucial role in maintaining neuronal health and circadian rhythms. BMAL1 is a core transcriptional regulator of the molecular circadian clock that coordinates daily rhythms in physiology and behavior. In this study, we investigated the impact of microglial modulation on circadian disruptions and behavioral abnormalities observed in bmal-/- mice. Our results revealed disruptions in circadian rhythms and microglial activation in bmal-/- mice, characterized by altered activity patterns and increased microglia numbers. Inhibiting microglia

Semilobar holoprosencephaly associated with maternal diabetes in a toddler: A case report.

Holoprosencephaly is a rare congenital malformation of the forebrain caused by incomplete division of the prosencephalon during early embryonic development, and semilobar holoprosencephaly represents the intermediate form of the spectrum. We report the case of a 3-year-old child presenting with developmental delay, seizures, feeding difficulties, and midline facial abnormalities. The child was born to a mother with poorly controlled maternal diabetes mellitus. The MRI scan of the brain showed partial fusion of the frontal lobes, incomplete separation of the hemispheres of the cerebrum, presenc
