Inherent Lipid Composition Abnormalities in Astrocytes Associated with Late-Onset Alzheimer's Disease (LOAD).

Lipid abnormalities have been observed in brain, cerebrospinal fluid (CSF), and blood in association with late-onset Alzheimer's disease (LOAD). It is unknown which of these abnormalities are precursors to LOAD and which are concomitants of illness or its treatment. Inherent abnormalities can be identified in induced pluripotent stem cell (iPSC)-derived brain cells. These cells lack markers associated with aging and environmental exposures. The iPSC lines of patients with LOAD or healthy individuals were differentiated to astrocytes. Astrocytes are crucial to neural activity and health, and al

Characterization of sensory nerve conduction abnormalities at the time of ALS diagnosis.

OBJECTIVES: Although sensory nerve abnormalities have been reported in amyotrophic lateral sclerosis (ALS), their distribution at diagnosis, relative to motor involvement and clinical onset phenotype remains incompletely characterized. We aimed to systematically compare sensory and motor nerve conduction abnormalities at ALS diagnosis and determine whether sensory involvement follows an onset-dependent pattern similar to motor dysfunction. METHODS: In this prospective cross-sectional study, 40 newly diagnosed ALS patients enrolled in the Iran University ALS Registry (March 2022-March 2023) un

Transdiagnostic mapping of brain structural abnormalities and network-constrained gray matter atrophy patterns across affective and psychotic disorders.

Major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia (SZ) are severe psychiatric disorders with distinct and overlapping clinical and neurobiological features. Despite extensive evidence of brain structural abnormalities, the transdiagnostic neuropathological mechanisms remain poorly understood. A comprehensive literature search was performed for voxel-based morphometry (VBM) studies reporting altered gray matter volume (GMV) in MDD, BD or SZ. A transdiagnostic meta-analysis was conducted to identify common and disorder-specific GMV alterations using the Seed-based d Mappi

ENT1 inhibition links oligodendrocyte lipid metabolism to connectivity in tauopathy.

INTRODUCTION: Metabolic dysfunction, altered adenosine signaling, and white matter abnormalities are implicated in tauopathies, but their relationship to network disconnection remains unclear. Myelinating oligodendrocytes may represent a metabolically vulnerable hub linking these processes to circuit dysfunction. METHODS: We assessed human hippocampal tissue by immunofluorescence and profiled THY-Tau22 mice using diffusion tensor imaging, metabolomics, lipidomics, and single-nucleus RNA sequencing. Symptomatic mice were treated with the equilibrative nucleoside transporter 1 (ENT1) inhibitor

Rebastinib inhibits cerebral cavernous malformation in a chronic mouse model.

BACKGROUND: Cerebral cavernous malformations (CCMs) are vascular lesions caused by the loss-of-function mutations in one of three CCM genes, CCM1 (KRIT1), CCM2, and CCM3 (PDCD10), or gain-of-function mutation in MAP3K3, which encodes MEKK3. Loss of function in CCM genes leads to pathological activation of MEKK3 signaling. Genetic reduction of MAP3K3 gene dosage or pharmacological inhibition of MEKK3 suppresses CCM formation in mouse models, establishing MEKK3 as a promising therapeutic target for CCM. METHODS: In silico screening was performed to identify candidate MEKK3 inhibitors. Therapeut

Structural morphometry distinguishes vagus nerve stimulation outcomes in individuals with epilepsy.

OBJECTIVE: Vagus nerve stimulation (VNS) is a commonly used treatment for drug-resistant epilepsy. However, response to VNS is variable, while understanding of this response variability is limited. The vagus afferent network (VagAN), the neural circuits thought to be involved in response to vagal afferents, has been implicated in response prediction in modalities such as diffusion-weighted imaging, functional imaging, and neurophysiology. Structural abnormality in the VagAN from T1w MRI has not yet been studied. METHODS: We assessed if VagAN abnormalities, derived from pre-implantation T1w MR

PLCγ2 deficiency compromises systemic immune tolerance and erodes myelin homeostasis while enhancing oxidative metabolism in the mouse brain.

BACKGROUND: Phospholipase C gamma-2 (PLCγ2) catalyzes the hydrolysis of the membrane phosphatidylinositol-4,5-bisphosphate (PIP2) to form diacylglycerol (DAG) and inositol trisphosphate (IP3), feeding into diverse downstream signaling pathways. PLCG2 polymorphisms have been associated with reduced and/or increased risk of Alzheimer's disease (AD) and related dementias, longevity, autoinflammation, and immune disorders. In the brain, PLCγ2 is expressed in microglia, and other neuroimmune and vascular interface populations, yet its role in brain homeostasis remains incompletely defined. METHODS

Bi-allelic variants in neuronal adhesion molecule astrotactin 1 gene ASTN1 cause diverse neurodevelopmental disorders.

ASTN1 encodes astrotactin 1, a neuronal-glial ligand in the developing brain that promotes neuronal migration along radial glia in brain structures with laminar organization, such as the cerebral cortex, hippocampus, and cerebellum. In mouse models, disruption of Astn1 results in neuronal migration deficits, a mild reduction in cerebellar volume, and balance and coordination deficits. In humans, bi-allelic ASTN1 variants have been identified in nine individuals with neurodevelopmental disorders (NDDs) with or without brain malformations. ASTN1 additionally interacts with astrotactin 2 (ASTN2)

Structural targeting of a familial variant of neuroserpin (Ser52Arg) by epigallocatechin gallate binding that reduces polymerization and enhances the tissue plasminogen activator inhibition.

Neuroserpin (NS) inactivation of tissue plasminogen activator (tPA) in brain reduces neurotoxicity. However several familial point mutations in its gene are linked with a neurodegenerative disease termed Familial Encephalopathy with Neuroserpin Inclusion Bodies (FENIB) that results in epilepsy/dementia. Variable expression level of the NS has been linked to several other neurological pathologies like Alzheimer's, glaucoma and ischemic stroke. A familial variant of NS that leads to mortality, Ser52Arg (S52R) was made using site directed mutagenesis. A circular dichroism study indicated close co

Effects of Anodal Left DLPFC tDCS on Panic Severity in Panic Disorder Patients With and Without Comorbid Rhinosinusitis and Nasal Septum Deviation.

PURPOSE: Panic disorder (PD) is characterized by recurrent, unexpected panic attacks and impaired top-down modulation of the fear circuitry, notably left dorsolateral prefrontal cortex (DLPFC) hypoactivity and dysregulation of respiratory-panic pathways. Transcranial direct current stimulation (tDCS) is a non-invasive brain stimulation technique that modulates cortical excitability by delivering low-intensity electrical currents through scalp electrodes. Chronic rhinosinusitis (CRS) and nasal septum deviation (NSD) commonly disrupt nasal breathing and sleep while increasing anxiety vulnerabili

White matter abnormalities in bipolar II and unipolar depression - Evidence from fixel-based analysis.

BACKGROUND: Challenges in correctly identifying bipolar disorder II (BD-II) during depressive states have led to poor clinical outcomes. BD-II-specific diffusion-weighted imaging studies are lacking. This study addresses current knowledge gaps by comparing white matter (WM) integrity in BD-II and unipolar depression (UD) using fixel-based analysis (FBA). METHOD: Primary analyses compared fibre density (FD) among 33 individuals with BD-II, 50 with UD, and 51 healthy controls (HC) in 11 a priori tracts. Sensitivity analyses among unmedicated participants examined whether medication status affec

Activity-dependent neuroprotective protein (ADNP) derived peptide NAP prevents musculoskeletal abnormalities in ovariectomized mice.

Estrogen is an essential hormone that critically impacts bodily and brain functions, supporting learning, memory, and motor activities. A decrease in estrogen levels is associated with cognitive decline and motor dysfunction, such as muscle weakness. While conventional hormone replacement treatments (HRT) exist, those have limitations and potentially severe side effects. NAP (davunetide) is the smallest neuroprotective peptide site of activity-dependent neuroprotective protein (ADNP), a master regulator of cognition, essential for brain formation. It is known that NAP restores ADNP activity in

Retinal Capillary Density Reduction Contributes to Dysthyroid Optic Neuropathy via an L-Arginine-NO Pathway: A Metabonomics and Clinical Trial Study.

Dysthyroid optic neuropathy (DON) is the most severe complication of thyroid-associated ophthalmopathy (TAO). Although recent evidence indicates that reduced retinal capillary density (RCD) may increase DON risk independently of orbital apex crowding, the underlying mechanisms and associated metabolic reprogramming remain unclear. In a retrospective analysis of TAO patients with and without DON, those with DON demonstrated elevated pulse pressure (PP), decreased RCD, and higher incidences of dyslipidemia, hyperglycemia, and internal carotid artery calcification. To explore the metabolic basis

Differences in white matter detected by ex vivo 9.4 T MRI are associated with axonal changes in the R6/1 model of Huntington's disease.

White matter volume loss has been reported as one of the first indicators in Huntington's disease (HD) patients, but the cellular basis of this deficit remains to be elucidated. To address this, we assessed white matter microstructure in the transgenic R6/1 mouse model of HD with ex vivo MRI. Specifically, a tractometry approach was employed to inspect region-specific differences across the corpus callosum (CC), while voxel-based morphometry (VBM) and tract-based spatial statistics (TBSS) were used to identify brain-wise white matter macro- and microstructure abnormalities. Alterations in the

Building a pediatric spasticity surgery program in a low-awareness setting: capacity creation, multidisciplinary integration, and early outcomes.

PURPOSE: Selective dorsal rhizotomy (SDR) and other spasticity-relieving procedures remain markedly underutilized in many regions of the world, not because of technical infeasibility but because of low awareness among referring specialties, the absence of structured referral pathways, and limited multidisciplinary integration. We describe the stepwise establishment of a pediatric spasticity surgery program in such a setting, with emphasis on capacity creation, stakeholder engagement, individualized treatment selection, and early clinical outcomes. METHODS: This program was developed at a tert

TFAP2E is implicated in central nervous system, orofacial and maxillofacial anomalies.

BACKGROUND: Previous studies in mouse, Xenopus and zebrafish embryos show strong tfap2e expression in progenitor cells of neuronal and neural crest tissues suggesting its involvement in neural crest specification. However, the role of human transcription factor activator protein 2 (TFAP2E) in human embryonic central nervous system (CNS), orofacial and maxillofacial development is unknown. METHODS: Through a collaborative work, exome survey was performed in families with congenital CNS, orofacial and maxillofacial anomalies.
