The Ts65Dn mouse is the most widely used model of Down syndrome (DS), although, in addition to the triplication of 90 genes homologous to Human Chromosome 21 (Hsa21) genes, it bears the triplication of 46 extra genes. To clarify the latter's impact, the Ts66Yah model has been created from the Ts65Dn mouse by exploiting CRISPR/Cas9 technology for extra gene deletion. It has been found that, similar to the Ts65Dn model, the Ts66Yah model exhibits impairment in hippocampus-dependent learning and memory and age-related hippocampal deterioration, with no increased activity. We examine here the dend

Background/Objectives: Up to 75% of patients with major depressive disorder (MDD) exhibit persistent anhedonia symptoms related to abnormalities in the positive valence system. Cumulative evidence points to brain dysfunction in the reward system (RS), including in the ventral striatum, in patients with MDD with anhedonia. This study aims to evaluate the safety and efficacy of a novel neurofeedback (NF) device (termed Prism) which incorporates the EEG-FRI-Pattern biomarker of the reward system (RS-EFP) for use in self-neuromodulation training (RS-EFP-NF) for alleviating depression in patients w

NGLY1 is a key enzyme in the process of misfolded protein deglycosylation. Bi-allelic pathogenic variants in NGLY1 cause N-glycanase deficiency, also known as congenital disorder of deglycosylation (NGLY1-CDDG). This rare and multisystem autosomal recessive disorder is linked to a variable phenotype of global developmental delay, neuromuscular abnormalities, and alacrima, and it lacks effective treatment. We have studied the possible underlying mechanisms for the neuromuscular and ophthalmic phenotypes in an ngly1-deficient zebrafish model carrying a similar genetic variant that has also been

Nonketotic hyperglycemia (NKH) can present with acute neurological symptoms such as tremor, ballismus, seizures, hemianopia, and coma, and may be associated with abnormal signal intensities on brain MRI. We report the case of a woman in her 50s with poorly controlled diabetes mellitus (HbA1c 15.9%) who was admitted with a chief complaint of right homonymous hemianopia. No other neurological deficits were observed. At presentation, the patient's blood glucose level was markedly elevated (608 mg/dL), with a blood pH of 7.36, negative ketone bodies, and an increased serum osmolality of 301 mOsm/L

Lewy Bodies, proteinaceous inclusions containing α-Synuclein fibrillary aggregates, are defining neuropathological hallmarks of Parkinson's disease and Dementia with Lewy Bodies. Recently, zebrafish arose as a valuable model to study neurodegenerative diseases, but despite attempts to generate stable human α-Synuclein transgenic zebrafish lines recapitulating the main phenotypic characteristics of Lewy Body disorders, i.e., α-Synuclein fibrillary aggregate deposition, neurodegeneration and behavioral deficits, none of the models produced so far could develop all these features. Here, we descri

The TRAILBLAZER-ALZ 6 study (NCT05738486) evaluated the effect of different donanemab dosing regimens on amyloid-related imaging abnormalities with edema/sulcal effusions (ARIA-E). The modified titration arm met the primary outcome and significantly reduced ARIA-E frequency compared with the standard dosing while maintaining a similar pharmacodynamic effect on amyloid reduction at 24 weeks. Primary outcome and 52-week data were previously published. Completed study results at 76 weeks are reported here. ARIA-E frequencies were 15.6 % and 24.2 % in the modified titration and standard arms, resp

Loss-of-function mutations affecting the lysosomal protein progranulin are a leading cause of frontotemporal dementia. Progranulin mutations cause abnormalities in lysosomal lipid processing, particularly of sphingolipids, major components of neural cell membranes that play important signaling roles in the brain. Most work in this area has focused on two classes of sphingolipids, gangliosides and cerebrosides. Here, we examined enzymes involved in metabolism of another class of sphingolipids, the sphingomyelins, in both mouse models and patients with progranulin insufficiency. Acidic sphingomy

Mutations or deletions in the SHANK3 gene have been identified in up to 1% of autism spectrum disorder cases and are considered the primary cause of neuropsychiatric symptoms in Phelan McDermid syndrome (PMS). While synaptic dysfunctions have been extensively documented in the absence of Shank3, other mechanisms through which Shank3 may regulate neuronal functions remain unclear. In this study, we report that the ribosomal protein Rpl3 and overall protein synthesis are downregulated in the cortex and striatum of Shank3 knockout (KO) mice and in neurons differentiated from human-induced pluripo

UNLABELLED: Children with autism spectrum disorder (ASD) exhibit diverse language abilities, yet magnetic resonance imaging (MRI) studies have not systematically examined white matter microstructural differences based on language difficulties. This study aimed to use diffusional kurtosis imaging (DKI) and tract-based spatial statistics (TBSS) to investigate white matter variations in boys with ASD and explore their relationship with language abilities. The study included 61 boys with ASD and 30 typically developing (TD) peers. The ASD group was divided into mild (n = 28) and severe (n = 33) la

Cognitive, social behavior, speech, and motor skills are known challenges for people with trisomy 21/Down syndrome (DS), but the precise mechanisms that lead to these impactful changes have not yet been described. Data from human and mouse model fetal brains indicate that alterations in prenatal neurogenesis might account for the neurological phenotypes that manifest after birth. Here, we evaluated key features of cortical neurogenesis in the humanized mouse model of DS (TcMAC21 of undetermined sex) to test whether and how the presence of the human HSA21q transchromosome impacts cortical devel

A relationship between epileptic activities and neuropsychological dysfunction has been indicated in epileptic children. Epileptic activities including frequent/prolonged seizures and severe EEG abnormalities could be associated with cognitive decline and behavioral disturbances. Frequent, prolonged seizures could induce growth disturbances in the prefrontal cortex, leading to behavioral disturbance. Meanwhile, frequent epileptic seizures or EEG abnormalities, including interictal epileptiform discharges (IEDs), are evident in attention deficit/hyperactivity disorder (ADHD). Subclinical IEDs c

PURPOSE: Approximately one-third of patients with epilepsy become refractory to traditional anti-seizure medications. Evidence from animals and in humans suggests a bidirectional relationship between epilepsy and neuroinflammation, offering potential as a novel therapeutic pathway. This study investigates region-specific relationships between neuroinflammation and medication-refractory focal epilepsy using [18F]FEPPA, a next-generation translocator protein radioligand and biomarker for microglial density with favorable binding kinetics and signal-to-noise ratio compared to [11C]PK11195.

BACKGROUND: Vascular anomalies are commonly associated with pulsatile tinnitus, but their potential role in triggering non-pulsatile tinnitus through neuroinflammation is not well established. Furthermore, few studies have investigated non-pulsatile tinnitus associated with altered cerebrovascular supply and whether anti-neuroinflammatory therapy might alleviate symptoms.
MATERIALS AND METHODS: We describe two patients presenting with non-pulsatile tinnitus and sigmoid sinus stenosis confirmed by imaging, with other causes systematically excluded.
RESULTS: Magnetic resonance venography demon

BACKGROUND: Diabetes mellitus (DM) is a known risk factor for cognitive decline and Alzheimer's disease (AD), possibly due to insulin resistance and impaired cerebral glucose metabolism. Fluorodeoxyglucose positron emission tomography-computed tomography (FDG PET-CT) imaging has shown AD-like hypometabolism patterns in diabetic individuals in various global studies. However, such data is lacking in the Indian population. This study investigated the presence of AD-like hypometabolism in Indian diabetic patients compared to non-diabetic controls.
AIM AND OBJECTIVE: This study aimed to evaluate

Noonan syndrome is the most common RASopathy and is associated with high rates of neurodevelopmental disorders. Prior neuroimaging studies in children with Noonan syndrome have identified structural effects on subcortical regions, though most focus on volumetric differences, overlooking finer morphological changes. These studies also tend to examine common genetic variants, excluding rarer forms within the Noonan syndrome spectrum. Shape analysis offers a sensitive approach to detecting subtle alterations, and when applied across variants, may reveal distinct neuroanatomical signatures. We acq

INTRODUCTION: Lipid abnormalities have been observed in brain, CSF, and blood in association with late-onset Alzheimer's disease (LOAD). It is unknown which abnormalities are precursors to LOAD and which are concomitants of illness or its treatment. Inherent abnormalities can be identified in induced pluripotent stem cell (iPSC)-derived neural lines.
METHODS: iPSC lines of patients with LOAD or healthy individuals were differentiated to astrocytes. Lipidomics analyses were performed on whole cell and mitochondrial extracts.
RESULTS: Large reductions in cholesterol esters (CE) and imbalances

Schizophrenia is a complex psychiatric disorder, driven by genetic and environmental factors. While individual risk genes have limited impact, polygenic susceptibility increases the likelihood of schizophrenia and heightens sensitivity to environmental stressors, such as prenatal immune activation. Yet, preclinical studies often focused on single-gene mutations, leaving polygenic influences largely unexplored. Using the apomorphine-susceptible (APO-SUS) rat model, which exhibits schizophrenia-like features, we investigated how polygenic susceptibility influences early neurodevelopment, synapti

Angelman syndrome (AS) is a debilitating neurodevelopmental disorder triggered by impaired function of the maternal UBE3A gene, which codes a protein that functions as a ubiquitin ligase and transcriptional coactivator. Ube3a maternal deficient mice replicate many key behavioral deficits associated with AS; however, the underlying molecular mechanisms remain poorly understood. Using an RT2 Profiler PCR array that summarizes the expression of 84 genes regulating synaptic plasticity, we identified a number of dysregulated genes in the visual cortex of AS mice brains at postnatal day 25 (P25) com
