The brain and spinal cord originate from a neural tube that is preceded by a flat structure known as the neural plate during early embryogenesis. In humans, failure of the neural plate to convert into a tube by the fourth week of pregnancy leads to neural tube defects (NTDs), birth defects with serious neurological consequences. The signaling mechanisms governing the process of neural tube morphogenesis are unclear. Here we show that in Xenopus laevis embryos, glutamate is released during neural plate folding in a Ca2+ and vesicular glutamate transporter-1 (VGluT1)-dependent manner. Vesicular

BACKGROUND: Intracranial dural arteriovenous fistula (DAVF) disrupts cerebral hemodynamics and can lead to widespread alterations in brain network connectivity and cognitive function. This study aimed to evaluate spontaneous brain activity and cognitive changes in DAVF patients using resting-state functional MRI (rsfMRI) and neuropsychological assessment, with evaluations conducted at baseline, 1 month, and 1 year postembolization to capture dynamic recovery-related changes in brain function and cognition.
METHODS: Fifty DAVF patients and 50 age and sex-matched healthy controls underwent rsfM

BACKGROUND: While several theories describe the pathophysiology of dural arteriovenous fistulas (DAVF), their true causes and development remain debated. There are a few documented cases of DAVF formation following venous sinus stenting (VSS) in the literature. Here, we present a case series of DAVF formation after VSS and provide a literature review to further emphasize their association.
METHODS: We conducted a retrospective analysis of patient data with DAVFs following VSS at our department and performed a review of the relevant literature. Baseline characteristics of the patients and DAVF

OBJECTIVES: Animal models suggest that combining mesenchymal stromal cells (MSCs) with prenatal myelomeningocele (MMC) repair may help preserve spinal cord function. This study assessed the feasibility of administration human umbilical cord-derived MSCs during prenatal repair of MMC in a sheep model.
METHODS: MMC (L1-L5) were surgically created in 11 fetal lambs at 75 days. Seven underwent repair 25 days later - five with and two without MSCs. A high-dose, ready-to-use MSCs product (Desacell®), derived from healthy human neonatal umbilical cord tissue was administered in five cases. Each rece

Cortical gyrification is a key marker of fetal brain development and is typically assessed qualitatively on ultrasound or MRI. While previous quantitative approaches have characterized gestational trajectories in typically developing (TD) fetuses, only a few studies have investigated cortical malformations such as lissencephaly and polymicrogyria. Spectral analysis, which characterizes signals by their frequency content, has been successfully applied to study gyrification in neonates and adults but has not yet been explored prenatally. This study aimed to apply spectral analysis to routine fet

A variety of neuropsychiatric symptoms have been reported in tuberous sclerosis. Although psychosis may occur, little has been written about this topic. This systematic review sought to characterize the nature of tuberous sclerosis-associated psychosis, including with respect to treatment response. A literature search was completed in June 2025 using PubMed and Scopus. Only articles that mentioned psychotic symptoms occurring in tuberous sclerosis in a patient-specific manner were eligible. Twenty-six papers describing 27 individuals were included in the main analyses, all of which are case re

BACKGROUND AND OBJECTIVES: The growing use and improved accessibility of noninvasive imaging modalities have increased the detection of incidental intracranial findings, such as unruptured brain arteriovenous malformations (AVMs). At the same time, existing literature presents conflicting findings regarding whether the incidence of ruptured AVMs has increased or decreased. Considering these developments, we hypothesized that the detection rate of unruptured AVMs has increased, and the detection rate of ruptured AVMs has changed over the period of 18 years, potentially reflecting shifts in unde

The mitochondrial outer membrane (OMM) plays a crucial role in maintaining cellular homeostasis by regulating mitochondrial dynamics, organelle interactions, and stress responses. In peripheral neurons-cells with high metabolic demands and long axons-the OMM acts as a vital platform for coordinating bioenergetics, calcium signaling, and redox balance. Ganglioside-induced differentiation-associated protein 1 (GDAP1), an OMM-anchored protein, has emerged as a key regulator of mitochondrial fission and transport, redox homeostasis, and mitochondrial membrane contact sites (MCSs). Genetic variants

BACKGROUND/OBJECTIVES: Human induced pluripotent stem cell (hiPSC) models provide a unique platform for testing the effect of genomic variants identified in patients with inherited diseases. In Alström syndrome, a rare multisystem disorder mainly caused by nonsense mutations in the ALMS1 gene, patients often present with infantile cardiomyopathy, retinal dystrophy, type 2 diabetes, and hearing loss in addition to obesity. These diverse clinical manifestations highlight the pleiotropic functions of ALMS1 in cellular processes such as ciliary signalling, cell cycle regulation, and tissue homeost

PURPOSE: Brain micro-arteriovenous malformations (BMAVMs) treatment is often challenging and considered safe for microsurgical resection. However, the role of endovascular treatment (EVT) remains to be clarified in this indication. The purpose of our study was to assess the safety and effectiveness of endovascular treatment as the first-line therapy for BMAVMs.
METHODS: From our local database, we retrospectively retrieved patients with BMAVMs treated by embolization as first-line therapy in our Department between January 1999 and January 2022. The primary endpoint was the total obliteration

The LAMA2 gene encodes the alpha-2 chain of laminin-2, a key component of the basement membrane that maintains muscle fiber stability and integrity. Mutations in this gene are linked with LAMA2-related muscular dystrophies, which includes congenital muscular dystrophy type 1 A (MDC1A) and limb-girdle muscular dystrophy autosomal recessive 23 (LGMDR23). Here, we present two siblings from one of 200 unrelated Iranian Charcot-Marie-Tooth (CMT) families, exhibiting axonal sensorimotor polyneuropathy, without any clinical signs of muscular dystrophy, carrying a homozygous variant in the LAMA2 gene.

Myelomeningocele (MMC) is the most common congenital defect of the central nervous system, which results in devastating and lifelong disability. In MMC, the initial failure of neural tube closure early in gestation is followed by a progressive prenatal injury to the exposed spinal cord, contributing to the deterioration of neurological function in fetuses. Amniotic fluid, collected during intrauterine correction of MMC, has been investigated as a new cell source for stem cells in the development of future cell-based transplantation. Human amniotic fluid stem cells (hAFSCs) adhered on decellula

Computational fluid dynamics (CFD) has been widely used to study cerebrospinal fluid (CSF) flow in Chiari Malformation Type I (CM-I). However, most approaches rely on limited patient-specific detail, and it remains unclear whether such minimal input is sufficient to yield physiologically realistic flow predictions. In this study, we construct a series of MRI-based models of the craniocervical CSF space in a CM-I patient, complemented with representations of microanatomical features derived from ex vivo measurements and patient MRI data, to assess how CFD predictions are influenced by the choic

Transarterial embolization (TAE) of dural arteriovenous fistulas (DAVFs) has evolved significantly following insurance approval of liquid embolic agents such as OnyxTM and Histoacryl® (n-butyl-2-cyanoacrylate[NBCA]). Onyx allows prolonged injection and controlled penetration into the venous side, whereas NBCA offers rapid polymerization and thrombogenicity. The success of TAE depends on the selection of appropriate delivery devices. The ultra-fine microcatheter, DeFrictor® nano, provides superior distal access to small feeders near the shunt point, thereby facilitating effective embolization.

Primary cilia play a pivotal role in cellular signaling and development. Human primary microcephaly is strongly associated with pathogenic variants in primary cilia genes. Here, we examine the role of Ttc21b, a component of the intraflagellar transport-A complex, during mouse forebrain development by using a Ttc21balien null allele. Our findings reveal that significant microcephaly in homozygous mutants is caused by disrupted neural progenitor proliferation and differentiation. Histological and immunohistochemical analyses show an enlarged ventricular zone and reduced cortical plate thickness

This review aims to provide a comprehensive pictorial review of low-flow vascular malformations (LFVMs) of the central nervous system (CNS) without arteriovenous shunting, focusing on their epidemiology, pathophysiology, imaging features, and associations with other vascular anomalies. LFVMs - developmental venous anomalies (DVAs), cavernous malformations (CMs), brain capillary telangiectasias (BCTs), and sinus pericranii (SP) - are typically benign and incidental but may cause symptoms or hemorrhage. Differentiating LFVMs from neoplastic, inflammatory, or high-flow vascular lesions is critica
