Differential Thigmotropic Capacities Among Fusarial Isolates: Fusarium solani Species Complex (FSSC) Isolates Most Potent.

Members of Fusarium oxysporum species complex (FOSC) and Fusarium solani species complex (FSSC) are common etiologic agents of opportunistic mycoses. Their hyphae grow via apical extension followed by invasion of host epithelial surfaces, particularly corneal and epithelial tissues. Thigmotropism is the contact-sensing response of these growing hyphae to change the direction of their apical growth in response to changes in the topography of their contacted surfaces, such as clefts between adjacent host cells. Investigations of fungal thigmotropism among etiologic agents of opportunistic mycose

Role of the Fungal Serological Marker Galactomannan Antigen in the Diagnosis of Invasive Fungal Infection: A Three-Year Retrospective Study.

Background and objectives Invasive fungal infections (IFIs) are the most prevalent potentially fatal opportunistic mycoses in immunocompromised individuals. Early detection is essential for treating these patients. Biomarkers, measured in blood or other clinical samples, are effective supplements to conventional pathologic and microbiologic diagnostic methods for IFIs. We conducted this study to determine the positivity of serum galactomannan (GM) in clinically diagnosed IFI. Also, we assessed the diagnostic power of GM for predicting invasive aspergillosis (IA), the most common cause of IFIs.

Clinical spectrum, clinical associations, and treatment outcomes of scedosporiosis in South India: A retrospective cohort study.

Scedosporium is an emerging opportunistic filamentous fungus widely distributed in soil and stagnant water. Mortality remains substantial and management is often challenging due to intrinsic antifungal resistance and delays in diagnosis. In this retrospective cohort study, we describe the clinical spectrum, microbiological characteristics, and outcomes of microbiologically culture-confirmed scedosporiosis at a tertiary care centre in South India between May 2016 and August 2024. A total of 18 patients were included, with a median age of 54 years and a predominance of male patients. Contrary to

Mesenchymal Stem Cells in Fungal Infections: Immunomodulation, Direct Antifungal Activity, and the Promise of the Secretome.

Mesenchymal stem/stromal cells (MSCs) are widely recognized as potent modulators of inflammation and immune function in bacterial and viral infections. However, their roles in fungal disease remain comparatively under-defined despite the growing clinical burden of invasive and opportunistic mycoses. This Feature Review synthesizes emerging evidence that MSCs influence antifungal outcomes through two complementary axes: (i) host-directed effects, including modulation of immune responses, particularly macrophage responses, and tissue/barrier conditioning; and (ii) fungus-directed effects (direct

Fungal Infections in Disorders of Inborn Errors of Immunity.

Emerging infectious threats continually challenge the adaptability of the human immune system. Inborn errors of immunity (IEIs) provide a unique framework to dissect host-pathogen interactions, revealing how single-gene defects uncover critical pathways in antifungal defence. This review offers an overview of fungal immunity through the lens of IEIs, and integrates recent advances in innate fungal recognition, neutrophil and mononuclear phagocyte effector mechanisms, CARD9/NF-kB signalling, IL-17/IL-23 and IL-12/IFN-gamma cytokine axes, and transcription-factor defects that shape susceptibilit

Frequency of mycological diagnosis in a clinical laboratory in the city of Pelotas, south of Brazil, during the three-year period (2020-2023).

The increasing prevalence of immunosuppressive diseases, the increase in the elderly population, and globalization have led to the urgent need for research on the epidemiology and prevalence of fungi that cause mycoses in Brazil and worldwide. This is especially important, in the case of Brazil, for the South region, which currently lacks updated data on the subject. To this end, the objective was to investigate fungi diagnosed in mycoses by a laboratory in the city of Pelotas, South of Brazil. An observational and analytical survey was conducted for three years to analyze fungal diagnoses in

Fungal diagnostics capacity in Malaysia: A survey across 48 laboratories.

Fungal infections affect 2% of Malaysia's population, yet little is known about mycology laboratory practices in the country. This study surveyed 48 medical institutions, including 14 state hospitals, 26 major specialist hospitals, 4 minor specialist hospitals, 3 university hospitals, and 1 reference laboratory, to assess current practices and identify areas for improvement. Nearly all hospitals performed germ tube testing for Candida albicans (87.5%, 42/48). Yeast identification was conducted using API® (52%, 25/48) or VITEK® (52%, 25/48), with two institutions employing both methods. Advance

A novel case of feline Exophiala spinifera disease in Americas: review integrating diagnosis and clinical.

We report the first case of phaeohyphomycosis caused by the dematiaceous dimorphic fungus Exophiala spinifera in a free-roaming domestic cat from the Americas, specifically diagnosed in the Central-West Region of Brazil, which progressed to euthanasia due to severe clinical deterioration. A comprehensive diagnostic approach was employed, integrating epidemiological, clinical, laboratory, mycological, and molecular analyses to confirm the fungal infection. In addition to the fungal infection, the cat tested seropositive for feline immunodeficiency virus (FIV) and was suspected of having seromuc

New and emerging drugs for the treatment of invasive fungal infections.

INTRODUCTION: The prevalence of invasive fungal infections (IFI) is increasing globally, with approximately 6,500,000 cases of IFI per year and 3,800,000 deaths. Most IFI deaths are attributable to opportunistic mycoses, such as Aspergillus and Candida. However, changes in climate and travel are leading to rising rates of endemic mycoses. Compounding this burden is the rise of antifungal drug resistance, requiring the development of new agents. AREAS COVERED: Data were identified using PubMed searches for the terms 'antifungal,' 'olorofim,' 'SUBA-itraconazole,' 'ibrexafungerp,' 'rezafungin,'

Silver nanoparticles as antifungal: from formulation to therapy in mycoses.

Silver nanoparticles (AgNPs) have gained prominence in the scientific literature as potential antifungal therapeutic agents, especially those obtained through biological synthesis, due to their superior physicochemical properties. This review examines formulations and patents incorporating AgNPs with antifungal activity, focusing on their applicability in the treatment of mycoses, particularly in vivo. Comparative data on the mechanism of action of AgNPs in planktonic cells and biofilms are presented, as well as transcriptional analyses in fungi exposed to these nanoparticles. The results demo

A case of panophthalmitis with orbital infection of polymicrobial origin.

BACKGROUND: Infectious endophthalmitis following penetrating ocular trauma represents an acute and often blinding disease with a generally poor prognosis. The causative pathogens involved are diverse, among which bacterial organisms are the most prevalent. In contrast, infections caused by fungi of the order Mucorales are rare, constituting acute and fulminant opportunistic mycoses. These fungal infections are characterized by an insidious onset and are associated with an exceedingly high mortality rate, particularly in patients with comorbid diabetes mellitus. CASE PRESENTATION: A 60-year-old

Human Mycobiota and Its Role in Cancer Progression, Diagnostics and Therapeutics: A Link Lesser-Known.

Although not as well studied as the bacterial component of the human microbiota, the commensal fungi or mycobiota play important roles in maintaining our health by augmenting our immune system. This mycobiota is also associated with various fatal diseases like opportunistic mycoses, and even cancer, with different cancers having respective type-specific mycobiota. The different fungal species which comprise these different intratumoral mycobiota play important roles in cancer progression. The aim of this review paper is to decipher the association between mycobiota and cancer, and shed light o

Mucormycosis in Liver Allograft Following Transplant for Secondary Biliary Cirrhosis.

Mucormycosis, a group of opportunistic mycoses caused by Mucorales, present a significant threat to immunocompromised patients. In this report, we present the case of a 57-year-old male patient who underwent liver transplant for secondary biliary cirrhosis following inadvertent bile duct injury. Despite initial satisfactory postoperative evolution, the patient developed fever, and imaging revealed a suspicious lesion. Preliminary culture growth suggested a filamentous fungus, leading to initiation of liposomal amphotericin B. However, the lesion progressed, and a surgical debridement was neces

Misleading subcutaneous mycosis: a case report of subsequent clinical mycetoma-like and histological chromoblastomycosis-like lesions.

Hyalohyphomycosis and phaeohyphomycosis are groups of mycoses caused by several agents and show different clinical manifestations. We report a case of an immunocompromised patient who presented rare manifestations of opportunistic mycoses: mycetoma-like hyalohyphomycosis on his right foot caused by Colletotrichum gloeosporioides, followed by cutaneous phaeohyphomycosis on his right forearm caused by Exophiala oligosperma. Further to the rarity of this case, the patient's lesion on the foot shows that the clinical aspects of mycetomas could falsely appear in other fungal infections similar to h

Extracellular vesicles derived from Talaromyces marneffei contain immunogenic compounds and modulate THP-1 macrophage responses.

Pathogenic eukaryotes including fungi release extracellular vesicles (EVs) which are composed of a variety of bioactive components, including peptides, nucleic acids, polysaccharides, and membrane lipids. EVs contain virulence-associated molecules suggesting a crucial role of these structures in disease pathogenesis. EVs derived from the pathogenic yeast phase of Talaromyces (Penicillium) marneffei, a causative agent of systemic opportunistic mycoses "talaromycosis," were studied for their immunogenic components and immunomodulatory properties. Some important virulence factors in EVs including

ABSTRACT: One in every two humans is having Helicobacter pylori (H. pylori) in stomach causing gastric ulcer. Emergence of several drugs in eliminating H. pylori has paved way for emergence of multidrug resistance in them. This resistance is thriving and thereby necessitating the need of a potent drug. Identifying a potential target for medication is crucial. Bacterial 5'-methylthioadenosine/S-enosyl homocysteine nucleosidase (MTAN) is a multifunctional enzyme that controls seven essential metabolic pathways. It functions as a catalyst in the hydrolysis of the N-ribosidic bond of adenosine-bas
