FOXL2 is a highly conserved transcription factor that is essential for ovarian development and female fertility. In humans, FOXL2 mutations or deletions cause blepharophimosis, ptosis, and epicanthus inversus syndrome (BPES) leading to eyelid defects and, in BPES type I, primary ovarian insufficiency. BPES is also associated with a wide range of less common phenotypes such as craniofacial features, cleft palate, or heart defects. In this study, we generated a new constitutive Foxl2-Cre mouse model using CRISPR/Cas9 editing. Crossing Foxl2-Cre mice with Rosa-tdTomato reporter confirmed robust a

Ankyloblepharon filiforme adnatum (AFA) is a rare, congenital eyelid anomaly characterized by partial fusion of the upper and lower eyelid margins by fine extensile tissue bands. We report a term female neonate with unilateral left AFA, intrauterine growth restriction, dysmorphic features, and congenital heart disease. Clinical evaluation raised suspicion of trisomy 18, which was confirmed by blood karyotyping. Echocardiography revealed a complex congenital heart defect suggestive of tetralogy of Fallot. The eyelid band was excised with scissors without sedation or local anesthesia, resulting

A 15-week-old Australian Labradoodle puppy was presented to The University of Queensland Small Animal Hospital for bilateral eyelid agenesis/coloboma and associated ocular complications. Enucleation of the left globe and repair of the right eyelid via a lip-to-lid transmucosal flap was performed. Given the subsequent development of prolapse of the gland of the nictitating membrane and a lack of vision of the right eye, enucleation of the right globe was then performed 2 months following the initial procedure. Both globes and a skin wedge from the right eye were submitted to The University of Q

BACKGROUND: Gelatinous drop-like dystrophy is a rare autosomal recessive disorder caused by pathogenic variants in the TACSTD2 gene and characterized by subepithelial amyloid deposits in the cornea. Alterations in FOXC2 are seen in lymphedema-distichiasis syndrome, in which patients classically exhibit lymphedema of the extremities and a double row of eyelashes. This report links alterations in the FOXC2 gene to a corneal phenotype that is histologically identical to that seen in gelatinous drop-like dystrophy. CASE PRESENTATION: A 14-year-old male with congenital distichiasis presented with c

PURPOSE: Marcus Gunn jaw-winking syndrome (MGJWS) is a rare congenital cranial dysinnervation disorder characterized by synkinetic eyelid elevation during jaw movement. Standard surgical management often involves levator excision with bilateral frontalis suspension, which abolishes levator function but carries morbidity and variable cosmetic outcomes. Posterior ptosis approaches such as Müller muscle-conjunctival resection (MMCR) are well established for phenylephrine-responsive ptosis, but their role in MGJWS remains unclear.
METHODS: A 12-year-old male with congenital left upper eyelid ptos

INTRODUCTION: Eyelid deformities can come from birth defects or other causes, and they often need full thickness reconstruction. The Cutler-Beard and Hughes techniques are still the best methods for repairing upper eyelid colobomas. However, there are not many choices available when the eyelid margin is still intact. Here, we report the clinical outcomes of a novel modification of the Cutler-Beard flap surgery in eyelid deformity for these difficult cases.

WNT10A mutations, a major genetic determinant of dental agenesis and ectodermal dysplasia, exert profound effects on craniofacial development. Although classified as rare disorders, these mutations account for more than half of oligodontia cases, reflecting their critical role. The associated phenotypes span a broad clinical spectrum, ranging from isolated tooth agenesis to complex multisystem disorders, including hypodontia, enamel and root defects, palmoplantar keratoderma, alopecia, nail dystrophy, skin xerosis, hypohidrosis, eyelid cysts, and skin cancer, skeletal abnormalities and impaire

BACKGROUND: Treacher Collins syndrome (TCS) is a rare craniofacial disorder characterised by variable expressivity. It is caused by pathogenic variants in the TCOF1, POLR1D, POLR1C, or POLR1B genes. Common clinical features include hypoplasia of the zygomatic complex and mandible, downward-slanting palpebral fissures, lower eyelid anomalies, microtia, and hearing loss. Owing to its phenotypic overlap with other craniofacial syndromes, molecular testing is essential for establishing an accurate diagnosis and guiding effective clinical management.

Heterozygous FOXL2 (non)coding sequence and structural variants (SVs) lead to blepharophimosis, ptosis and epicanthus inversus syndrome (BPES), a rare, autosomal dominant developmental disorder characterized by a completely penetrant eyelid malformation and incompletely penetrant primary ovarian insufficiency (POI). We collected variants from our in-house database, generated via clinical genetic testing and downstream research testing in the Center for Medical Genetics Ghent, Belgium (2001-2024) and via literature and other resources in the same period. All retrieved variants were categorized

UNLABELLED: Pseudomonas aeruginosa AG1 (PaeAG1) is a Costa Rican strain that was isolated in 2010 in a major Hospital. This strain has resistance to multiple antibiotics such as β-lactams (including carbapenems), aminoglycosides, and fluoroquinolones. PaeAG1 is considered critical (Priority 1) due to its resistance to carbapenems, and it was the first report of a P. aeruginosa isolate carrying both VIM-2 and IMP-18 genes encoding for metallo-β-lactamases (MBL) enzymes (both with carbapenemase activity). Owing to these traits, we have studied this model for 10 years using diverse approaches inc

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

BACKGROUND: In Europe, the prevalence of congenital ocular anomaly (COA) is estimated at 3.7 per 10,000 births. While certain COAs have a genetic origin, the cause for most patients remains unknown. The role of medications administered during pregnancy in COA genesis in humans is unclear.
OBJECTIVE: To investigate any association between fetal exposure in the first trimester of pregnancy to medications and the occurrence of COA.
METHODS: We conducted a case-malformed-control study using data on 298,351 cases registered as having congenital anomalies (CA) from 19 registries and one healthcare

BACKGROUND: Treacher Collins syndrome (TCS, MIM #154500), a severe congenital disorder, predominantly involves dysplasia of craniofacial bones and is characterized by features such as downslanting palpebral fissures, lower eyelid colobomas, microtia, and other craniofacial anomalies. Despite its clinical importance, the underlying pathogenic mutations in TCS remain largely uncharacterized, representing a critical knowledge gap for researchers in the field.
RESULTS: To address this, we performed mutation screening on a familial TCS case (trio) and 11 sporadic cases from a Chinese population. W

Optic cup morphogenesis is a complex process involving cellular behaviors such as epithelial folding, cell shape changes, proliferation, and tissue fusion. Disruptions to these processes can lead to an ocular coloboma, a congenital defect where the optic fissure fails to close. This study investigates the role of Shroom3, a protein implicated in epithelial morphogenesis, in mouse embryos during optic cup development. It was observed that Shroom3 is apically localized in the neural retina and retinal pigmented epithelium, and its deficiency leads to a both a conventional coloboma phenotype char

Miller syndrome is an extremely rare condition in the group of facial dysostosis syndromes. These syndromes have great phenotypic overlap and variability, even within families. To facilitate the differentiation of Miller syndrome from related facial dysostosis syndromes, such as Treacher Collins and Nager syndrome, this study aims to provide an overview of the phenotypic spectrum of the syndrome. A systematic literature search of Embase, MEDLINE/PubMed, Web of Science, and CINAHL was conducted until November 2024. Periorbital defects resulting from oncologic resections, trauma, or congenital malformations pose a complex reconstructive challenge, due to the need to simultaneously restore eyelid function and facial esthetics. We present the case of a male patient in his seventh decade of life with a right orbitomalar squamous cell carcinoma, who underwent a wide oncologic resection involving the upper and lower eyelids, as well as the malar and infraorbital regions. Reconstruction was performed using a radial free forearm flap from the left arm, including the palmaris longus tendon, which was strategicall
