Genomic Profiling of Anophthalmia/Microphthalmia-Associated CNVs Reveals Complex Genotype-Phenotype Correlations and Incomplete Penetrance.

BACKGROUND: Anophthalmia/microphthalmia (A/M) is a severe congenital ocular malformation characterized by the complete absence or small size of the eye bulb. Interpreting copy number variations (CNVs) in A/M is challenged by variable genotype-phenotype correlations and reduced penetrance. This study investigated the genetic etiology of A/M-associated CNVs. METHODS: Genomic profiling was performed on four unrelated families presenting with ocular anomalies or harboring A/M-susceptible CNVs. Variants were evaluated by integrating American College of Medical Genetics and Genomics (ACMG) guidelin

Case Report: Persistent fetal vasculature associated with lenticular coloboma.

BACKGROUND: Persistent fetal vasculature (PFV) is a rare congenital ocular anomaly, featuring typical fibrovascular stalks in the vitreous cavity. PFV may cause anterior and posterior segment anomalies such as leukocoria, microphthalmos, cataract, elongated ciliary processes, optic nerve hypoplasia, intralenticular hemorrhage, retinal dysplasia and strabismus. However, cases of PFV concurrent with lenticular coloboma are rarely reported. This report presents a rare case of anterior PFV associated with lenticular coloboma. CASE PRESENTATION: A 23-year-old female presented with poor vision sinc

Cataract and glaucoma surgery in microphthalmic, nanophthalmic, and high hyperopic eyes: a systematic review and meta-analysis.

TOPIC: This systematic review and meta-analysis evaluates refractive error (RE), visual acuity (VA), intraocular pressure (IOP), and anterior chamber depth (ACD) outcomes following surgical procedures in adults (≥ 18 years) with nanophthalmos, microphthalmos, and high hyperopia. CLINICAL RELEVANCE: Nanophthalmos, microphthalmos, and high hyperopia are a spectrum of anatomic variants characterized by short axial length and crowded anterior segment. These features create significant risks for angle-closure glaucoma and complex cataract surgery. As existing literature is fragmented and limited t

Characterization of Ocular Developmental Disorders in the Israeli Population: Genotype-Phenotype Correlations and Novel Candidate Genes.

Microphthalmia, anophthalmia and ocular coloboma (MAC) are rare developmental eye disorders. Although over 100 causative genes have been identified, the molecular spectrum and genotype-phenotype correlations remain incompletely understood, particularly in genetically diverse populations. We set out to molecularly characterize MAC in the Israeli population. Forty-seven MAC-affected individuals from 43 unrelated families were enrolled. DNA of all probands was subjected to whole exome sequencing. The most common phenotype was microphthalmia (64% of patients). Definite or possible molecular diagno

Lipemia retinalis in a neonate with Kaufman oculocerebrofacial syndrome: a case of co-occurrence.

We report the case of a term-born infant girl with notable ocular and facial anomalies who was found to have creamy white retinal vessels consistent with lipemia retinalis. Laboratory tests showed severe hypertriglyceridemia and high cholesterol levels. Neuroimaging revealed microphthalmia and optic nerve hypoplasia, and genetic testing identified a homozygous UBE3B mutation, confirming Kaufman oculocerebrofacial syndrome. The infant was treated with a fat-restricted diet and fenofibrate, which led to normal lipid levels and complete fundus recovery within one month. This case represents a cli

Nanophthalmos presenting with acute refractory angle-closure glaucoma: acute-phase histopathological evidence and bilateral comparative findings.

BACKGROUND: Nanophthalmos is a rare congenital ocular disorder characterized by a microphthalmic eye with abnormally thickened sclera. Although scleral histopathology has been described in elective surgical cases with normal intraocular pressure (IOP), scleral architecture during acute IOP elevation exceeding 60 mmHg remains unclear. This article reports unique histopathological findings obtained during an episode of refractory acute angle-closure glaucoma. CASE PRESENTATION: A 63-year-old man with lifelong high hyperopia (corrected with + 10.25 D spectacles) presented with acute severe ocula

Asymmetric uveal effusion in bilateral nanophthalmos presenting as unilateral secondary angle closure: a rare case report.

PURPOSE: To report a rare case of unilateral secondary angle closure in a young male with bilateral nanophthalmos caused by spontaneous supraciliary and choroidal effusion, highlighting the diagnostic role of multimodal imaging and the importance of individualized management. CASE REPORT: A 33-year-old male, a known case of hyperopia in both eyes, presented with gradual visual loss in both his eyes. Examination revealed asymmetric anterior chamber depth with shallowing in right eye (RE) > left eye (LE), bilateral short axial lengths, and a 360° choroidal detachment in the RE, while the fellow

Splicing variants in MYRF cause partial loss of function in the retinal pigment epithelium leading to nanophthalmos.

Improper light focus on the retina, refractive error, is primarily caused by eye size differences and is the leading cause of vision loss worldwide. C-terminal variants in the Myelin regulatory factor (MYRF) gene, a retinal pigment epithelium-derived (RPE-derived) transcription factor, lead to isolated nanophthalmos characterized by a small, though structurally sound eye. However, other MYRF loss-of-function variants cause syndromic disease. To address this discrepancy, in vitro and animal studies were performed on a pathogenic C-terminal variant dG-MYRF (p.Gly1126fs30*, c.3376-1G>A). Human RP

Genetic findings and clinical features of extreme refractive errors in Thai children using whole genome sequencing.

INTRODUCTION: Extreme refractive errors in children may indicate underlying genetic or syndromic conditions. This study investigated the genetic basis of high refractive errors in Thai children and compared clinical characteristics between high myopia and high hyperopia, and identified clinical predictors of a positive genetic result. METHODS: Ninety-eight children (<18 years) with high refractive errors were recruited from the Queen Sirikit National Institute of Child Health. All underwent comprehensive ophthalmic examination; ocular biometry and optical coherence tomography (OCT) were perfo

Cutis Tricolor-Like Pigmentary Mosaicism in Mowat-Wilson Syndrome: Phenotypic Overlap With Ruggieri-Happle Syndrome.

Cutis tricolor (CT) is a rare pigmentary mosaicism characterized by the coexistence of hyperpigmented and hypopigmented areas on a background of normal skin; its syndromic form, Ruggieri-Happle syndrome (RHS), is associated with neurodevelopmental delay, facial dysmorphism, skeletal abnormalities, and other systemic defects. We report a 16-year-old girl with molecularly confirmed Mowat-Wilson syndrome caused by a de novo pathogenic ZEB2 frameshift variant, who presented with extensive CT-like pigmentary mosaicism and marked phenotypic overlap with RHS. Brain MRI disclosed bilateral microphthal

Translational animal model for genetic predisposition to anophthalmia/microphthalmia.

Anophthalmia and microphthalmia (A/M) are severe congenital malformations that are responsible for ∼11% of childhood blindness. Despite clinical advances, approximately 70-80% of human A/M cases lack a molecular diagnosis. This study aims to characterize a spontaneous, non-syndromic model of the microphthalmia, anophthalmia, and coloboma (MAC) spectrum in the laboratory opossum (Monodelphis domestica), providing a novel experimental opportunity for embryonic developmental and ocular pathology research, as well as translational research into preventions and treatments of these conditions. Maternal nutrition determines the variable expressivity of STRA6-associated eye malformations.

Mutations in STRA6, the high-affinity receptor for retinol-binding protein, cause Matthew-Wood syndrome, a congenital disorder characterized by striking phenotypic variability ranging from isolated ocular anomalies to severe multisystem malformations. The mechanisms underlying this variable expressivity and incomplete penetrance remain poorly understood. Here, we used mice to determine how maternal vitamin A status and embryonic Stra6 genotype interact to shape ocular development. Using Stra6-deficient mice bred under defined dietary conditions, we demonstrate that maternal vitamin A availabil

Congenital tarsal kink syndrome.

Congenital horizontal tarsal kink syndrome is a rare but well-defined form of congenital upper eyelid entropion, characterized by a horizontal tarsal fold causing inversion of the lid margin and lashes, with absence of a visible upper eyelid crease. Induced trichiasis may lead to corneal complications and a major risk of amblyopia if diagnosis and management are delayed. We report the case of a 4-week-old infant with right unilateral tarsal kink complicated by a central corneal ulcer and associated microphthalmia. This case was managed by an anterior approach with release of orbicularis adhesi

In silico insights into shared molecular mechanisms and drug repurposing for non‑small cell lung cancer and tuberculosis.

UNLABELLED: Universal trauma-informed screening is increasingly recommended across pediatric settings, yet it remains unclear which forms of adversity most accurately and equitably identify youth at risk for psychopathology. In response, the current study tested whether defining maltreatment by cumulative quality (i.e., number of subtypes) versus cumulative frequency (i.e., number of experiences regardless of subtypes), as well as potential differences within each subtype, yielded more accurate and fair predictions of trauma-related psychological distress. Participants included 839 adolescents

cGAS‑STING pathway in digestive tract cancer: From molecular mechanisms to therapeutic applications (Review).

BACKGROUND: Idiopathic lymphoplasmacytic rhinitis (LPR) is a common inflammatory disorder of the nasal cavity in dogs due to unknown etiology. It is characterised by non-specific clinical signs, including nasal discharge, epistaxis and breathing problems. Diagnosis is usually based on the histopathologic identification of infiltrating plasmocytes and lymphocytes in the nasal mucosa and the exclusion of other underlying diseases. Treatment strategies include glucocorticoids, non-steroidal anti-inflammatory drugs, antibiotics and antifungal medications. The aim of this study was to evaluate the

Understanding histotripsy ablation safety: a systematic review of pre-clinical and clinical adverse events.

BACKGROUND: Facet joint tropism is asymmetry in orientation of the bilateral facets. Some studies have shown that tropism may increase the risk of disc degeneration and herniations, as well as degenerative spondylolisthesis (DS). It remains controversial whether tropism is a pre-existing developmental phenomena or secondary to progressive remodeling of the joint structure due to degenerative changes. As such, the following study addressed the occurrence of tropism of the lower lumbar spine (i.e. L3-S1) in a degenerative spondylolisthesis patient model. METHODS: An international, multi-center

Mechanism of activation of an ancestral TEC kinase by PIP3.

BACKGROUND: The introduction of second-generation antipsychotics (SGAs) over the past 2 decades generated considerable optimism that better antipsychotic treatments for schizophrenia and bipolar disorder were possible. SGAs offer several tolerability benefits over first-generation antipsychotics (FGAs), particularly with respect to extrapyramidal symptoms. However, SGAs can induce serious metabolic dysregulations, especially in drug-naive, first-episode, and child and adolescent populations, with olanzapine and clozapine having the highest propensity to cause these abnormalities. Residential radon in Peru: challenges for health surveillance and lung cancer prevention.

The Journal of Genetics, started by William Bateson in 1910, played a distinguished role in the early years of genetics. However, it stopped publishing in 1978. The Indian Academy of Sciences revived it in 1985, and has published it regularly since then. To commemorate this landmark, I highlight one of the 17 articles published that year.
