Gallbladder Cancer: Epidemiology and Prevention.

Gallbladder cancer (GBC) is an uncommon but highly lethal malignancy, characterized by striking geographic clustering, late diagnosis, and a prevention profile that differs from other gastrointestinal malignancies. This narrative review synthesizes epidemiologic, mechanistic, clinical, and public-health evidence on GBC determinants with a prevention-oriented lens. Rather than merely cataloguing risk factors, we propose a pragmatic framework that combines epidemiologic credibility, population-attributable burden, baseline risk, biologic plausibility, feasibility, and the benefit-risk balance of

A rare case of aphakia associated with anterior segment malformation in a cocker spaniel puppy.

This report describes a rare case of bilateral congenital aphakia with anterior segment malformation in a 12-week-old male cocker spaniel puppy. The puppy was initially suspected of having glaucoma and was subsequently diagnosed with anterior segment dysgenesis and aphakia resulting in blindness. Ophthalmic examination showed corneal opacity, absence of pupillary openings, and negative pupillary light reflexes. B-mode and high-frequency ultrasonography showed absence of the lens, poorly defined anterior chambers, and hyperechoic tissue adherent to the cornea, consistent with anterior segment d

Characterization of a spontaneous microphthalmia-like mutant mouse with isolated ocular defects.

Microphthalmia is a significant eye defect owing to its profound effects on visual acuity. Microphthalmia accounts for 3.2%-11.2% of blind children. To date, there has been no cure for this disease. In this study, we aimed to identify microphthalmia-like mutant mouse and study its growth and development. In this study, we identified mutant mice exhibiting eye abnormalities using a forward genetics approach in a C57BL/6J cohort. To identify ocular characteristics of the mutant mouse, we conducted systematic evaluations including basic measurements (body length, body weight, and palpebral fissur

Putative congenital bilateral lens anomaly in a free-ranging American black bear cub from North Carolina, USA.

An ~6-mo-old, free-ranging, male, American black bear (Ursus americanus) cub that was observed stumbling, acting disoriented, and falling, was dispatched and sent for postmortem evaluation. Bilaterally, round concretions were present in both the anterior and posterior aspects of the lens, disrupting the lens epithelium. These concretions were continuous with and extended from the inner surface of the lens capsule, as seen with a periodic acid-Schiff-hematoxylin reaction. Immunohistochemistry for alpha A crystallin was inconsistent, with weakly positive immunolabeling indicative of lens fibers.

Zebrafish as a model for crystallin-associated congenital cataracts in humans.

Congenital cataracts are a leading cause of vision loss in children and can be an isolated finding or associated with systemic abnormalities. Isolated congenital cataracts are most commonly associated with pathogenic variants in one of the Crystallin genes. The α-Crystallins are small heat shock proteins that act as chaperones in the lens and other organs throughout the body to prevent protein aggregation and maintain tissue function. In contrast, the ß- and γ-Crystallins are structural proteins that are predominantly expressed in the mature lens and regulate its refractive index. However, the

Risk of Congenital Ocular Anomaly After Prenatal Exposure to Medications: A EUROmediCAT Study.

BACKGROUND: In Europe, the prevalence of congenital ocular anomaly (COA) is estimated at 3.7 per 10,000 births. While certain COAs have a genetic origin, the cause for most patients remains unknown. The role of medications administered during pregnancy in COA genesis in humans is unclear. OBJECTIVE: To investigate any association between fetal exposure in the first trimester of pregnancy to medications and the occurrence of COA. METHODS: We conducted a case-malformed-control study using data on 298,351 cases registered as having congenital anomalies (CA) from 19 registries and one healthcare

Shroom3 facilitates optic fissure closure via tissue alignment and reestablishment of apical-basal polarity during epithelial fusion.

Optic cup morphogenesis is a complex process involving cellular behaviors such as epithelial folding, cell shape changes, proliferation, and tissue fusion. Disruptions to these processes can lead to an ocular coloboma, a congenital defect where the optic fissure fails to close. This study investigates the role of Shroom3, a protein implicated in epithelial morphogenesis, in mouse embryos during optic cup development. It was observed that Shroom3 is apically localized in the neural retina and retinal pigmented epithelium, and its deficiency leads to a both a conventional coloboma phenotype char

A de novo FBN1 variant likely causes congenital bilateral ectopia lentis in a crossbred horse.

Although several inherited ocular disorders have been extensively studied in horses, few reports of equine ectopia lentis exist and no genetic investigations have been reported. Ectopia lentis in humans and other species is reported to be caused by trauma, genetic variants, and systemic diseases. The most commonly reported genetic causes are dominant alleles in FBN1. Here we examined a 3-day old Oldenburg x Thoroughbred colt due to concerns over bilateral ocular anomalies and hypothesized that either a recessively inherited allele or a dominant de novo allele was the genetic cause. Examination

Primary Angle Closure in a 7-Year-Old Child: Evaluation and Findings With Conventional Ultrasound and Ultrasound Biomicroscope.

Primary angle closure (PAC) is rare in pediatric patients, typically associated with persistent hyperplastic vitreous, congenital cataracts, ocular surgery, retinopathy of prematurity, topical or oral medications, and ocular albinism. This case report describes the youngest documented case of unilateral subacute PAC in a seven-year-old female without these underlying conditions. The patient underwent evaluation using A-scan ultrasound and ultrasound biomicroscopy (UBM), revealing a shallow anterior chamber and a short axial length (20.45 ± 0.02 mm in the left eye). The angle closure was caused

In vivo quasi-elastic light scattering detects molecular changes in the lenses of adolescents with Down syndrome.

Down syndrome (DS) is the most common chromosomal disorder in humans. DS is associated with increased prevalence of several ocular sequelae, including characteristic blue-dot cerulean cataract. DS is accompanied by age-dependent accumulation of Alzheimer's disease (AD) amyloid-β (Aβ) peptides and amyloid pathology in the brain and comorbid early-onset Aβ amyloidopathy and colocalizing cataracts in the lens. Quasi-elastic light scattering (QLS) is an established optical technique that noninvasively measures changes in protein size distributions in the human lens in vivo. In this cross-sectional

Advances in epileptic network findings of hypothalamic hamartomas.

Hypothalamic hamartomas (HHs) are congenital developmental malformations located in the hypothalamus. They are associated with a characteristic clinical manifestation known as gelastic seizures (GS). However, the traditional understanding of HHs has been limited, resulting in insufficient treatment options and high recurrence rates of seizures after surgery. This is consistent with the network hypothesis of focal epilepsy that the epileptogenic zone is not only limited to HH but may also involve the distant cerebral cortex external to the HH mass. The epilepsy network theory, on the other hand

Analysis of long-range chromatin contacts, compartments and looping between mouse embryonic stem cells, lens epithelium and lens fibers.

BACKGROUND: Nuclear organization of interphase chromosomes involves individual chromosome territories, "open" and "closed" chromatin compartments, topologically associated domains (TADs) and chromatin loops. The DNA- and RNA-binding transcription factor CTCF together with the cohesin complex serve as major organizers of chromatin architecture. Cellular differentiation is driven by temporally and spatially coordinated gene expression that requires chromatin changes of individual loci of various complexities. Lens differentiation represents an advantageous system to probe transcriptional mechani
