A rare case of aphakia associated with anterior segment malformation in a cocker spaniel puppy.

This report describes a rare case of bilateral congenital aphakia with anterior segment malformation in a 12-week-old male cocker spaniel puppy. The puppy was initially suspected of having glaucoma and was subsequently diagnosed with anterior segment dysgenesis and aphakia resulting in blindness. Ophthalmic examination showed corneal opacity, absence of pupillary openings, and negative pupillary light reflexes. B-mode and high-frequency ultrasonography showed absence of the lens, poorly defined anterior chambers, and hyperechoic tissue adherent to the cornea, consistent with anterior segment d

A radiological case series of three siblings with osteogenesis imperfecta and shared paternal inheritance.

Источник пометил публикацию как отозванную

Osteogenesis imperfecta (OI), or brittle bone disease, is a genetically inherited connective tissue syndrome that manifests through autosomal dominant or recessive patterns. We present an unusual case involving three siblings with the same father but different mothers. The patients had extremely short stature, recurrent fractures, and varying degrees of severity of OI, in addition to distinctive features such as blue sclera and dentinogenesis imperfecta. The radiographic skeletal surveys revealed the characteristic features of OI in all three siblings. The patients exhibited Wormian bones, mul

Non-coding structural variants disrupting conserved PITX2 enhancer loci in Axenfeld-Rieger syndrome.

Axenfeld-Rieger Syndrome (ARS) is an autosomal dominant condition with both ocular and non-ocular manifestations. ARS is primarily caused by coding variants at the PITX2 or FOXC1 loci, yet many cases still remain undiagnosed. Here we used whole-genome sequencing to identify two families with non-coding structural variants associated with a typical presentation of PITX2-associated ARS: one family with a 450 kb deletion removing a series of conserved enhancer elements distal to PITX2, and the second family with a 12.54 Mb inversion displacing the PITX2 gene from these same enhancer elements. Severe Aortic Regurgitation in Osteogenesis Imperfecta Complicated by Mediastinal Hematoma.

BACKGROUND: Osteogenesis imperfecta (OI), a heritable type I collagen disorder, can cause aortic regurgitation (AR) and increase perioperative bleeding risk, even with normal routine coagulation tests. CASE SUMMARY: A man in his 40s presented with AR due to right coronary cusp prolapse from cusp bending. Blue sclera and childhood fractures suggested OI. After an attempted repair, surgical aortic valve replacement was performed; a large postoperative mediastinal hematoma with anemia was managed conservatively without reoperation. DISCUSSION: A prior report identified cusp bending in OI mainly

A new species of Pristimantis (Anura: Craugastoridae) from the western slopes of the Serranía del Baudó, Chocó, Colombia.

We describe a new species of Pristimantis based on morphological and acoustic evidence from the western slopes of the Serranía del Baudó, tentatively assigned to the Pristimantis ridens species group. The new species can be distinguished from congeneric species by having coppery or reddish bronze iris, with fine black reticulations and light blue sclera, upper eyelid bearing one to three subconical tubercles, posterior surfaces of thighs generally solid brown, with small cream specks present in a few cases and groin solid brown, with some individuals showing a cream or golden blotch. Moreover,

Putative congenital bilateral lens anomaly in a free-ranging American black bear cub from North Carolina, USA.

An ~6-mo-old, free-ranging, male, American black bear (Ursus americanus) cub that was observed stumbling, acting disoriented, and falling, was dispatched and sent for postmortem evaluation. Bilaterally, round concretions were present in both the anterior and posterior aspects of the lens, disrupting the lens epithelium. These concretions were continuous with and extended from the inner surface of the lens capsule, as seen with a periodic acid-Schiff-hematoxylin reaction. Immunohistochemistry for alpha A crystallin was inconsistent, with weakly positive immunolabeling indicative of lens fibers.

Gillespie syndrome caused by a novel ITPR1 gene variant: a phenotype-genotype correlation.

BACKGROUND: Gillespie syndrome (GS) is a rare congenital disorder with ocular and neurological defects. Classically, it has been noted to have a triad of cerebellar ataxia, variable absence of iris but always with a scalloped edge and intellectual disability. This scalloped iris edge is essential to make a diagnosis of GS. Both autosomal dominant and recessive inheritance have been reported with de novo pathogenic variants in the inositol 1,4,5-trisphosphate receptor type 1 gene (ITPR1). We report a novel variant of ITPR1 gene causing Gillespie syndrome in multiple members of a family. MATERI

ABL kinases regulate FGF signaling independent of CRK phosphorylation to prevent Peters anomaly type II.

Peters anomaly is an anterior segment dysgenesis and a leading cause of congenital corneal opacity. Here, we show that loss of ABL kinases restores lens induction in the absence of FGF signaling but induces Peters anomaly type II independently of ERK signaling, a phenotype also observed with elevated FGF-Ras activity. This defect is rescued by allelic deletion of the ABL substrates CRK and CRKL. Contrary to prevailing models, ABL kinases do not act through direct phosphorylation of CRK proteins; instead, they phosphorylate PTPN12, suppressing p130CAS phosphorylation and CRK recruitment require

Clinical Outcomes of Pars Plana Vitrectomy in Boston Keratoprosthesis Type I in Pediatric Patients.

PURPOSE: Keratoprosthesis offers certain pediatric patients with congenital corneal opacity an alternative for improving vision. Pars plana vitrectomy (PPV) combined with Boston keratoprosthesis type 1 (KPro) has demonstrated success in reducing postoperative complications such as retinal detachment (RD) in adults. However, its role and outcomes have yet to be investigated in pediatric patients. This study investigates the outcomes of PPV in KPro pediatric patients, with a focus on prosthesis retention, visual acuity (VA), and postoperative complications. METHODS: We performed a retrospective

Pre-operative High-Resolution CT and MRI Evaluation in Pediatric Cochlear Implant Candidates: Correlation With Surgical Findings and Outcomes.

Purpose Congenital sensorineural hearing loss (SNHL) is a major childhood disability, and early cochlear implantation offers optimal auditory and language outcomes. High-resolution computed tomography (HRCT) and magnetic resonance imaging (MRI) are essential in evaluating candidacy, identifying inner ear malformations (IEMs), assessing cochlear nerve integrity, and predicting surgical challenges. This study evaluates the role of HRCT and MRI in pediatric cochlear implant candidates and correlates imaging findings with intraoperative events and postoperative outcomes. Methods This retrospective

Universal Newborn Eye Screening in São Paulo, Brazil.

This brief report describes the incidence and characteristics of ocular abnormalities in healthy term newborn infants (HTNI) in 5,000 consecutive eye examinations from a database of 32,000 infants from 17 hospitals in São Paulo, Brazil. Imaging consisted of five views of the retina (optic nerve center, superior, inferior, nasal, temporal) with 130° wide-angle lens and one view of the anterior segment in each eye. These images were curated on four separate occasions for the presence or absence of ocular abnormality by a pediatric retina specialist. All images were obtained within 72 hours of bi

Adult Case of Stüve-Wiedemann Syndrome.

Stüve-Wiedemann Syndrome (SWS) is a rare autosomal recessive skeletal dysplasia caused by variations in the leukemia inhibitory factor receptor (LIFR) gene. Due to its high rate of mortality in early life, current literature is largely limited to children with SWS. We present a case report of a 35-year-old female with SWS, one of the few documented adults with SWS. She was diagnosed at age 34 after clinical exome sequencing identified homozygous pathogenic variants in LIFR at c.756dup (p.Lys253*), a known pathogenic variant associated with SWS. Her past medical history consists of feeding and

Association of FOXC1 Duplications With Juvenile Open-Angle Glaucoma.

IMPORTANCE: While FOXC1 single-nucleotide variants and deletions are well-established causes of Axenfeld-Rieger syndrome, few FOXC1 duplications have been reported. This study investigated families with duplications encompassing the FOXC1 gene to refine the associated phenotypic spectrum and contribution to glaucoma. OBJECTIVE: To investigate the prevalence and phenotype of FOXC1 duplications in 2 large glaucoma registries. DESIGN, SETTING, AND PARTICIPANTS: This retrospective observational genetic cohort study included participants recruited from the Australian & New Zealand Registry of Adv

Gene Panel Analysis Reveals Overlapping Genetic Causes of Inherited Cataracts and Other Ocular Phenotypes in Bulgarian Patients.

BACKGROUND: Inherited forms of cataract are a clinically important and genetically heterogeneous cause of visual impairment, usually present at an early age. Both syndromic and non-syndromic congenital cataracts exhibit all modes of inheritance and may occur in association with various ocular or systemic abnormalities. Early diagnosis and treatment are crucial for the visual prognosis. METHODS: We applied next-generation sequencing (NGS) of clinical exome for identification of the disease-causing variants in a group of seven Bulgarian patients with syndromic and non-syndromic cataracts, accom
