Osteogenesis imperfecta (OI), or brittle bone disease, is a genetically inherited connective tissue syndrome that manifests through autosomal dominant or recessive patterns. We present an unusual case involving three siblings with the same father but different mothers. The patients had extremely short stature, recurrent fractures, and varying degrees of severity of OI, in addition to distinctive features such as blue sclera and dentinogenesis imperfecta. The radiographic skeletal surveys revealed the characteristic features of OI in all three siblings. The patients exhibited Wormian bones, mul

BACKGROUND: Osteogenesis imperfecta (OI), a heritable type I collagen disorder, can cause aortic regurgitation (AR) and increase perioperative bleeding risk, even with normal routine coagulation tests.
CASE SUMMARY: A man in his 40s presented with AR due to right coronary cusp prolapse from cusp bending. Blue sclera and childhood fractures suggested OI. After an attempted repair, surgical aortic valve replacement was performed; a large postoperative mediastinal hematoma with anemia was managed conservatively without reoperation.
DISCUSSION: A prior report identified cusp bending in OI mainly

We describe a new species of Pristimantis based on morphological and acoustic evidence from the western slopes of the Serranía del Baudó, tentatively assigned to the Pristimantis ridens species group. The new species can be distinguished from congeneric species by having coppery or reddish bronze iris, with fine black reticulations and light blue sclera, upper eyelid bearing one to three subconical tubercles, posterior surfaces of thighs generally solid brown, with small cream specks present in a few cases and groin solid brown, with some individuals showing a cream or golden blotch. Moreover,

Stüve-Wiedemann Syndrome (SWS) is a rare autosomal recessive skeletal dysplasia caused by variations in the leukemia inhibitory factor receptor (LIFR) gene. Due to its high rate of mortality in early life, current literature is largely limited to children with SWS. We present a case report of a 35-year-old female with SWS, one of the few documented adults with SWS. She was diagnosed at age 34 after clinical exome sequencing identified homozygous pathogenic variants in LIFR at c.756dup (p.Lys253*), a known pathogenic variant associated with SWS. Her past medical history consists of feeding and

Musculoskeletal disorders (MSDs) are primarily characterized by joint, bone, and connective tissue involvement, but they can also present with ocular manifestations that carry diagnostic and clinical significance. These findings such as uveitis, blue sclera, lens subluxation, and scleritis may serve as early markers of systemic disease or indicate ongoing inflammation that affects orthopaedic decision making. Despite their relevance, ocular symptoms are frequently underrecognized in orthopaedic settings, potentially delaying comprehensive care. This narrative review aims to explore the spectru

Osteogenesis imperfecta (OI) comprises a group of disorders that lead to bone fragility. The most common forms are caused by pathogenic variants in COL1A1 and COL1A2. There are many different animal models OI from zebrafish to dogs, with murine models being the most used model. In this brief study, we describe the successful generation of the first large animal porcine model of OI Type I carrying a clinically relevant pathogenic variant, c.757C>T (p.Arg253Ter). These pigs exhibited classic features of OI Type I including blue sclera, low bone mass, and bone fragility. This model is more geneti

Biallelic variants in PYROXD1 are associated with a life limiting muscle and connective tissue disorder characterised by generalised muscle weakness, breathing and feeding difficulties, distal laxity, hypernasal speech, blue sclera and osteopenia. PYROXD1 encodes an oxidoreductase implicated in mitochondrial function and tRNA ligase activity. Herein we present the first comprehensive suite of mouse models designed to elucidate PYROXD1 redox functions and the underlying pathogenetic basis for PYROXD1 disorders. A LacZ reporter strain reveals ubiquitous expression of Pyroxd1, particularly in the

PURPOSE: Ehlers-Danlos syndrome (EDS) is a group of inherited connective tissue disorders with systemic manifestations, but its ophthalmic implications are poorly characterized. Existing studies suggest an increased prevalence of many ophthalmic conditions as well as a higher rate of surgical complications, but a comprehensive understanding is lacking.
METHODS: An anonymous survey was distributed online through social media and patient advocacy organizations. Participants reported genetic and ophthalmic diagnoses, year of diagnosis, refractive error, and ophthalmic surgical history. Analyses

Osteogenesis imperfecta (OI) is a rare genetic disorder characterized by bone fragility. Its association with dentinogenesis imperfecta (DI) is well documented, but the concurrent presentation with nephrocalcinosis is uncommon and poorly understood. We documented the case of an 18-year-old male presenting with a triad of OI, DI, and nephrocalcinosis. The patient exhibited characteristic features including blue sclera, multiple fractures, dental abnormalities, bowing of long bones, a short stature, and biochemical evidence of altered calcium metabolism. Genetic testing revealed mutations in COL

Background: Osteogenesis imperfecta (OI) is marked by clinical and genetic heterogeneity, and the genotype-phenotype correlation remains not very clear. We conducted a clinical and genetic study in a Chinese OI cohort to determine the spectra of phenotypes and pathogenic variants. Methods: In this study, 298 Chinese families were recruited from 2019 to 2024. Clinical phenotypes including fractures, short stature, skeletal deformities, blue sclera, dentinogenesis imperfecta, and hearing loss were recorded and analyzed. Next-generation sequencing combined with PCR-based techniques was used to de

Variants in the Col1a1 gene typically lead to a connective tissue disorder called osteogenesis imperfecta (OI), which is characterized by increased bone fragility that may be associated with blue sclera, dentinogenesis imperfecta and hearing loss. However, the coexistence of pes equinovarus and OI is rare, and to date, no genetic basis has been described. We report a female patient who was admitted with short stature, and growth hormone replacement treatment was initiated following a diagnosis of growth hormone deficiency. She also had blue sclera, a bulbous nose, flexion contractures in both

UNLABELLED: In pediatric populations, unexplained fractures, particularly in non-ambulatory children, raise immediate concern for nonaccidental trauma (NAT). Osteogenesis imperfecta (OI) is a rare genetic disorder marked by bone fragility, frequent fractures, and features like blue sclera. Distinguishing between these two entities remains a significant diagnostic challenge. Misdiagnosis of OI as NAT carries substantial medical, legal, and psychosocial consequences for families and underscores the need for accurate, timely diagnosis supported by multidisciplinary evaluation. We present a case o

Osteogenesis imperfecta (OI) is a congenital skeletal disorder characterized by varying degrees of bone fragility and deformities. Extraskeletal manifestations, such as blue sclera, dentinogenesis imperfecta, growth disturbance, hearing impairment, and muscle weakness, occasionally accompany OI. Many genes have been identified as causative of OI, such as the type I collagen gene and genes involved in the folding, processing, and crosslinking of type I collagen molecules, osteoblast differentiation, and bone mineralization. According to the discovery of the causative gene of OI, nosology and cl

RATIONALE: Osteogenesis imperfecta (OI) is a genetic disorder of mesenchymal hypoplasia and collagen defects. Whether abnormal type I collagen predisposes OI patients to multilevel lumbar disc herniation remains unclear.
PATIENT CONCERNS: An 18-year-old male with childhood-diagnosed type I OI (fragility fractures, blue sclera, dentinogenesis imperfecta, severe osteoporosis) developed progressive low-back pain and bilateral radiculopathy.
DIAGNOSES: Magnetic resonance imaging revealed multilevel lumbar disc herniation with relatively mild nucleus pulposus degeneration. Whole-exome sequencing

Background: Factor X (FX) deficiency is a rare autosomal recessive inherited bleeding disorder, with an estimated prevalence of approximately 1 in 1,000,000 individuals. According to the most recent data published by the World Federation of Hemophilia, no cases of FX deficiency have been reported in Cambodia to date. Case Report: A 14-year- and 7-month-old Cambodian boy presented with recurrent gum bleeding. His medical history was notable for multiple hematomas, joint ankylosis, and blue sclera. He was born to second-degree consanguineous parents, with no known family history of bleeding diso

INTRODUCTION: Osteogenesis imperfecta (OI) is a rare disorder causing multiple fractures throughout life. No treatment has been shown to reduce the risk of fractures in OI. Here, we present the baseline characteristics of participants in the Treatment of Osteogenesis Imperfecta with Parathyroid Hormone and Zoledronic Acid (TOPaZ) trial. The aim of the trial is to determine whether teriparatide and zoledronic acid are superior to standard care in reducing the risk of clinical fractures.
METHODS: We summarised data on the baseline characteristics of TOPaZ participants, including demographics, g

PURPOSE: Blue sclera is a characteristic and common clinical sign of Osteogenesis Imperfecta (OI). However, there is currently no widely accepted, objective method for assessing and grading blue sclera in individuals with OI. To address this medical need, this study is aimed to design and validate a new method called 'BLUES' (BLUe Eye Sclera) to objectively identify and quantify the blue color in the sclera of patients affected by OI.
METHODS: Sixty-two patients affected by OI and 35 healthy controls were enrolled in the present prospective study, for a total of 194 eyes analyzed. In the 'BLU
