The transplantation of stem cells and viruses in utero has tremendous potential for treating congenital disorders in the human fetus. For example, in utero transplantation (IUT) of hematopoietic stem cells has been used to successfully treat patients with severe combined immunodeficiency. In several other conditions, however, IUT has been attempted without success. Given these mixed results, the availability of an efficient non-human model to study the biological sequelae of stem cell transplantation and gene therapy is critical to advance this field. We and others have used the mouse model of

Paired box 2 (PAX2)-related disorder, also known as renal coloboma syndrome, is a variably penetrant autosomal dominant condition, associated with renal and ophthalmological abnormalities. We report a child with PAX2-related disorder who presented atypically with acute ataxia on a background of stage 3 chronic kidney disease. Extensive biochemical, radiological and gene agnostic rapid trio exome sequencing was non-diagnostic. Identification of bilateral optic disc colobomas in the proband and his father raised the suspicion of an inherited PAX2-related disorder. No causative variants were iden

Optic disc pit is a congenital anomaly of the optic disc which is usually asymptomatic, but some patients can develop optic disc pit maculopathy at some stage of life. Choroidal neovascularization (CNV) is an abnormal growth of blood vessels into the neurosensory retina from choroidal vessels. Although CNV is not commonly seen in the pediatric age group, there are certain optic nerve head anomalies, like optic disc pit, optic nerve drusen, optic nerve head cavity abnormalities, etc., which are associated with CNV in children. Here, we present a 13-year-old girl who was diagnosed with CNV in as

Morning glory syndrome (MGS) is a rare congenital optic disc malformation characterized by a distinctive funnel-shaped optic disc resembling a blooming morning glory flower, typically diagnosed in childhood or adolescence and primarily presenting unilaterally, although bilateral cases have been reported. The anomaly features an enlarged optic disc, radiating blood vessels, and surrounding excavation, leading to visual impairment that can range from mild to profound. The etiology of MGS is not fully understood but has been associated with genetic factors, particularly mutations in the PAX6 and

PURPOSE OF REVIEW: Optic disc pit maculopathy (ODPM) is a rare yet serious condition that can lead to visual impairment. While pars plana vitrectomy (PPV) remains the gold standard for ODPM that requires treatment, there is ongoing debate regarding adjunctive surgical techniques. This review explores the latest surgical advancements and innovations in the management of ODPM from the past few years.
RECENT FINDINGS: Vitrectomy remains an effective procedure for ODPM management, achieving high success rates. Adjunctive techniques, including internal limiting membrane (ILM) peeling, gas tamponad

INTRODUCTION: Optic disc pit maculopathy (ODP-M) describes the variable intra- (IRF) and/or subretinal fluid (SRF) accumulation complicating a congenital optic disc anomaly that is primarily observed in young adults. This study aimed to explore the morphological variance in ODP-M, in order to measure associations between demographic and functional characteristics and investigate the natural course of the disease.
METHODS: A single-centre, retrospective, observational study was performed. Subjects with ODP-M were identified through electronic notes review. Demographic characteristics, visual a

Observation with optical coherence tomography (OCT) is a reasonable management approach for optic pit maculopathy presenting with macular fluid. We describe two patients with optic pit maculopathy who exhibited preserved vision and fluctuating macular fluid managed with serial OCTs. A 49-year-old woman presented with blurred vision in the left eye. Best-corrected visual acuity (BCVA) was 20/40 in the left eye. Examination revealed an inferotemporal gray depression in the optic disc; OCT demonstrated submacular fluid. Serial OCTs over 19 years showed fluctuating fluid with stable vision. BCVA w

A 33-year-old male presented with bilateral, painless diminution of vision. During a previous ophthalmic consultation elsewhere, he was diagnosed with papilledema. Neuroimaging revealed an Arnold Chiari Type 1 malformation, and an urgent neurosurgical intervention was performed. Currently, ophthalmic examination revealed an anterior chamber reaction, vitritis, optic disc edema (ODE), and exudative retinal detachment in both eyes. He was clinically diagnosed with Vogt-Koyanagi-Harada disease and treated with high-dose intravenous methylprednisolone, followed by immunosuppression, resulting in a

Persistent fetal vasculature (PFV) refers to a spectrum of developmental ocular anomalies due to incomplete regression of embryonic vasculature. PFV may present with a wide range of manifestations and visual consequences. We report a rare manifestation of PFV wherein an exudative choroidal neovascular membrane formed in association with the posterior origin of the fibrovascular stalk. While most PFV stalks arise from the optic disc, the stalk in this case originated away from the disc in the inferonasal peripapillary area and had not undergone surgical transection. The patient was treated with
