Relationship Between External Ear Severity and Temporal Bone Malformation in Microtia: A Computed Tomography Study.

Background/Objectives: Microtia is frequently associated with temporal bone malformations, but the extent to which external ear phenotype reflects underlying CT-defined anatomy remains uncertain. This study aimed to compare the relationships of Marx severity grading and Nagata morphological classification with temporal bone anatomy in patients with microtia. Methods: This retrospective study included 26 patients with 27 affected ears who had both preoperative external ear photographs and temporal bone computed tomography (CT) examinations. Each affected ear was classified according to the Marx

Virtual outpatient assessment in microtia reconstruction: A sustainable and equitable model for quaternary care.

BACKGROUND: Centralisation of highly specialised services improves outcomes for complex congenital conditions, but it often requires families to travel substantial distances for outpatient review and multidisciplinary assessment. Microtia reconstruction in the United Kingdom is delivered through a small number of specialist centres, creating potential financial, logistical and environmental burdens for patients. We evaluated the impact of a fully virtual supra-regional microtia clinic at Great Ormond Street Hospital (GOSH). METHODS: A retrospective review was undertaken of all patients who at

Surgical Technique for Deep Concha, a Pseudomeatus, and High Projection in Congenital Auricular Reconstruction.

BACKGROUND: Despite successful cartilage framework construction and auricular elevation in 2-stage reconstruction, achieving adequate conchal cavity depth and auricular projection remains challenging. This study introduces a systematic third-stage refinement technique incorporating cartilage flap support, conchal excavation, and split-thickness skin grafting to address these aesthetic limitations. METHODS: Between January 2020 and December 2025, 127 patients with congenital microtia underwent 3-stage ear reconstruction. The technique comprised supraperiosteal tissue excavation to deepen the c

[Research advance on the clinical phenotypes and molecular genetic mechanisms of Microcephalic primordial dwarfism].

Primordial dwarfism (PD) refers to a group of monogenic genetic disorders characterized by intrauterine growth restriction (IUGR) and severe, persistent postnatal growth retardation. These diseases have been associated with variants of multiple genes whose products are mainly involved in critical cellular biological processes such as maintenance of genomic stability, DNA damage repair, mRNA splicing regulation, and centrosome function. Variants of such genes can directly impair cell proliferation and developmental potential. With the widespread application of molecular genetic technologies suc

Genetic variation near ROBO1 is associated with craniofacial microsomia and related phenotypes in the Finnish population.

Craniofacial microsomia (CFM) encompasses a phenotypic continuum of congenital anomalies ranging from isolated microtia to more complex manifestations within the oculo-auriculo-vertebral spectrum, including Goldenhar syndrome, reflecting abnormal development of first and second pharyngeal arch-derived structures. While several genetic susceptibility loci have been reported, population-based evidence in individuals of European ancestry remains limited. Using nationwide data from FinnGen in the Finnish founder population, we identified a genome-wide significant association at a conserved interge

Clusters in craniofacial microsomia and microtia according to facial morphology and craniofacial anomalies.

UNLABELLED: Associated craniofacial malformations such as ocular anomalies and macrostomia are frequently seen in craniofacial microsomia (CFM). This study assessed associations between objectively measured facial morphology and clinically observed craniofacial malformations in patients with CFM in order to identify clinically significant patient clusters based on these characteristics for preferential screening purposes. In this multicenter cross-sectional cohort study, facial morphology was assessed from three-dimensional (3D) photographs using asymmetry index (ASI) and facial signature (FS)

Clinical and zebrafish studies of truncating SF3B2-variants in craniofacial microsomia.

Craniofacial microsomia (CFM) exhibits significant phenotypic variability and degree of severity. While loss-of-function variants in SF3B2 have recently emerged as a genetic etiology, the molecular basis underlying this clinical heterogeneity remains poorly understood. Here, we report two probands harboring novel truncating SF3B2 variants and presenting with distinct clinical phenotypes. Proband 1, with a heterozygous p.(Gln60*) variant, exhibited characteristic CFM features, including mandibular hypoplasia, cleft palate, bilateral tragal abnormalities, microtia, external auditory canal stenos

Analysis of Eya1 and Tbx1 mutants highlights interactions between the muscle and developing cartilage during external ear formation.

Microtia is a common feature of several human syndromes affecting the external ear (pinna), yet the cellular and molecular mechanisms remain poorly understood. Using human embryos and mouse models of branchio-oto-renal (BOR) and 22q11.2 deletion syndromes, we show that the syndromic genes Eya1 and Tbx1 are expressed in mesoderm-derived auricular muscle. In Eya1 mutant mice, auricular muscles failed to form and pinna morphogenesis was disrupted, with comparable defects observed in mesoderm-specific Tbx1 mutants. Both mutant pinnae exhibited impaired cartilage differentiation, suggesting that au

Early cleft lip repair in an infant with cleft lip-palate and ipsilateral microtia identified through a community outreach program.

Cleft lip and palate are common craniofacial anomalies that may affect feeding, speech, and hearing. When associated with external ear malformations such as microtia, careful evaluation for auditory impairment and possible syndromic conditions is required. We report a 3-month-old male infant with a left complete unilateral cleft lip and palate and ipsilateral Weerda grade II microtia who underwent early cleft lip repair through an outreach cleft care program. The patient presented with feeding difficulty but no additional craniofacial or systemic anomalies. Surgical repair using a rotation-adv

Functional and Genetic Analyses Unveil the Implication of hoxa4a in Zebrafish Craniofacial Development.

Microtia-atresia is a rare craniofacial malformation primarily affecting the first and second pharyngeal arches, leading to the deformity of the auricle and atresia of the external ear canal. Its etiology is heterogenous and largely unknown, including both genetic and environmental factors. The HOXA4 gene has been identified as potentially pathogenetic for microtia-atresia in three twin families. A hoxa4a mosaic knockdown zebrafish model was constructed using CRISPR/Cas9. hoxa4a was expressed in the mandible during early development in zebrafish, while the F0 mosaic knockdowns exhibited cranio

Comprehensive Prosthodontic Rehabilitation of a Completely Edentulous Adolescent With Cleft Lip and Palate and Multiple Facial Defects: A Case Report.

Cleft lip and palate (CLP) is the most prevalent congenital craniofacial anomaly and is frequently associated with dental anomalies and compromised maxillary anatomy. Complete edentulism in adolescents with CLP is exceedingly uncommon, and its concurrent presentation with bilateral auricular agenesis and nasal hypoplasia represents a rare clinical scenario. A 17-year-old male patient with surgically corrected bilateral CLP, congenital bilateral auricular agenesis, and a congenital nasal defect presented with complete edentulism of both arches. Comprehensive prosthodontic rehabilitation was per
