Splenic macrophage-B cell axis drives systemic autoimmune-like pathology in Cerebral Malaria.

Cerebral malaria (CM) is a severe complication of Plasmodium infection, classically attributed to parasite sequestration and neuroinflammation. Here, we uncover a spleen-centered humoral autoimmune circuit that drives CM pathology. Proteomic analyses identify CD36 as a dominant host-derived antigen enriched in infected red blood cells (iRBCs), triggering anti-CD36 autoantibody production in patients with falciparum malaria. Although contributing to iRBCs clearance, these autoantibodies also target other CD36-expressing cells, thereby driving thrombocytopenia, endothelial injury, and macrophage

Evaluation of HLA-G and Hematobiochemical levels and their association with malaria and miscarriage among pregnant women: A Ghanaian case-control study.

BACKGROUND: HLA-G (Human Leukocyte Antigen-G) is a non-classical molecule belonging to the major histocompatibility complex (MHC) class I family, located on chromosome 6 within the MHC region, which is pivotal in promoting immune tolerance. The immune-suppressive ability of Human Leukocyte Antigen (HLA-G) and its binding to fetal maternal interface predispose pregnant women to Plasmodium falciparum (Pf) infection and pregnancy loss, respectively. Alterations in hematological parameters may accelerate the severity of malaria infection. Limited data is available about the effects of malaria infe

Inflammation and Routine Laboratory Associations in Imported Falciparum Malaria Assessed by Partial Correlation Analysis.

PURPOSE: To examine whether associations among routine admission laboratory measurements in imported Plasmodium falciparum malaria persist after accounting for other measured variables, including C-reactive protein (CRP). METHODS: This retrospective single-centre study included all adults recorded in a departmental malaria database as hospitalised with microscopically confirmed P. falciparum monoinfection between 2010 and 2025. A rank-based precision matrix yielded 28 partial correlations among seven laboratory measurements and age, each adjusted for the other six variables. Bonferroni correc

Sepsis secondary to co-infection with malaria, dengue and COVID-19: a case report.

The co-infection of malaria, dengue fever, and COVID-19 is rarely described in Sub-Saharan Africa, yet the overlapping clinical and biological features of these diseases pose a significant diagnostic challenge in endemic areas. We report the case of a 36-year-old woman requiring admission to the intensive care unit in Burkina Faso with acute respiratory distress. She presented with a triple co-infection of Plasmodium falciparum malaria, dengue fever (DEN3 serotype), and COVID-19, complicated by sepsis and multiple organ dysfunction. The patient exhibited severe hypoxemia, hepatic dysfunction (

A Study on Prevalence of Malarial Retinopathy among Malaria Cases and to Determine Its Presence as a Marker of Severe Malaria: Observations from Eastern India.

BACKGROUND: Malarial retinopathy refers to a constellation of changes seen in severe or complicated malaria cases. These include: retinal whitening, vessel changes-whitening, tramlining, retinal hemorrhages, and papilledema. There are very few Indian studies on this entity. Since retina can be easily visualized by direct ophthalmoscopy, this study was done to determine prevalence of malarial retinopathy among malaria cases and to determine relationship between malarial retinopathy and severity of the disease. MATERIALS AND METHODS: The study was done at Indoor and Outdoor Departments of Tropi

Mapping historical diaries into knowledge graphs: a multi-granular approach.

PLAIN ENGLISH SUMMARY: There are new ways to engage people with science and research but many patient support groups and charitable organisations still hold traditional meetings to provide updates on their activities and to report new developments in their field of interest. These meetings often feature presentations given by medical doctors or, in the case of research-focussed organisations, by research scientists.Receiving feedback from people who are confused and sometimes upset by some types of information, and the way it is presented at meetings, made us think about better ways for resear

Harnessing Artificial Intelligence to Restore Standards in Medical and Dental Education in Pakistan.

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

Unveiling new perspectives about the onset of neurological and cognitive deficits in cerebral malaria: exploring cellular and neurochemical mechanisms.

Cerebral malaria is the most severe and lethal complication caused by Plasmodium falciparum infection, leading to critical neurological impairments and long-term cognitive, behavioral, and neurological sequelae in survivors, particularly affecting children under the age of five. Various hypotheses have been proposed to explain the neurological syndrome associated to cerebral malaria condition, including vascular occlusion and sequestration, cytokine storm or inflammatory response, or a combination of these mechanisms and despite extensive research and a growing range of scientific information,

Unraveling the Role of Proteinopathies in Parasitic Infections.

Proteinopathies, characterized by the misfolding, aggregation, and deposition of proteins, are hallmarks of various neurodegenerative and systemic diseases. Increasingly, research has highlighted the role of protein misfolding in parasitic infections, unveiling intricate interactions between host and parasite that exacerbate disease pathology and contribute to chronic outcomes. The life cycles of parasitic protozoa, including Plasmodium, Toxoplasmosis, and Leishmania species, are complicated and involve frequent changes between host and vector environments. Their proteomes are severely stresse

Severe imported Plasmodium falciparum malaria with hyperparasitaemia: evaluation of determinants of critical disease in adult returning travellers.

BACKGROUND: Severe Plasmodium falciparum (P.f.) malaria remains a major health threat for travellers. World Health Organization (WHO) defines criteria for severe malaria, including hyperparasitaemia ≥10% infected red blood cells (iRBCs), as major risk factors for adverse outcome. Additionally, WHO recognizes 'uncomplicated hyperparasitaemia' with 4-10% iRBC, a parasite density usually defining severe malaria outside endemic areas. Overall, the role of hyperparasitaemia as an independent risk factor in imported severe malaria is unclear, with most data predating the artemisinin era. METHODS: W

Phenotypic screens reveal Plasmodium falciparum genetic factors associated with infection of sickle-trait cells.

BACKGROUND: Malaria causes over 200 million cases and more than half a million deaths annually. In many African regions, hemoglobinopathies, such as sickle cell trait (HbAS), confer partial protection against severe P. falciparum malaria. HbAS significantly reduces the risk of severe, life-threatening malaria by over 90 %. This study aims to describe a new analysis for the piggyBac transposon-based mutagenesis phenotypic screen to identify genes that influence the mechanisms behind this protection and tolerance of P. falciparum to the HbAS intracellular microenvironment, providing insights int

Guillain-Barré syndrome following falciparum malaria infection: a case report.

BACKGROUND: Malaria is an infectious disease caused by Plasmodium parasites, transmitted to humans by infected female Anopheles mosquitoes. Five Plasmodium species infect humans: P. vivax, P. falciparum, P. ovale, P. malariae, and P. knowlesi. Guillain-Barré Syndrome (GBS) is an inflammatory condition that can lead to paralysis, autonomic dysfunction, respiratory failure, and sensory symptoms. GBS typically follows an infection with Campylobacter bacteria, commonly found in undercooked poultry, but is rarely associated with malaria. CLINICAL PRESENTATION: A 16-year-old female patient presente

Management and monitoring strategies for severe cerebral malaria: a guide for the intensivist.

Severe malaria, caused by Plasmodium falciparum, poses a critical public health challenge, with cerebral malaria (CM) representing its most severe and life-threatening neurological manifestation. Defined by impaired consciousness (Glasgow Coma Score < 11) after the exclusion of other causes of encephalopathy, CM remains a critical condition with a mortality rate of 15-25% and long-term neurological sequelae in survivors. CM pathogenesis involves parasitized erythrocyte sequestration in cerebral microvasculature, immune hyperactivation, blood-brain barrier disruption, and cerebral edema, potent

Critical Understanding of the Influence of Cellular Aging Biomarkers on Host-Parasite Relationships Serving as a Key Platform for Malaria Eradication.

Plasmodium parasites are the causative agents of malaria and can infect humans and other vertebrates, impacting socioeconomic development and causing significant health issues globally. Plasmodium falciparum causes the most severe type of infection, which can lead to chronic morbidity and other severe complications like anemia and cerebral malaria. The onset of infection is marked by the injection of sporozoites into the skin through the bite of a female Anopheles mosquito. This triggers a cascade of reactions elicited both by the host immune system in response to infection and by the parasite

Current update on malaria in pregnancy: a systematic review.

BACKGROUND: Malaria during pregnancy poses significant risks to both the mother and the developing fetus. For pregnant women, the infection can result in severe illness and even death. Parasite sequestration in the placenta can cause maternal anemia and increase the risk of mortality both during and after childbirth. Malaria is also a major contributor to stillbirths and preterm births. Infected placental tissue can impede fetal growth, resulting in low birth weight, which is linked to delayed growth and cognitive development in the child. Furthermore, malaria during pregnancy remains a major

Quantifying aminoglycoside resistance in extended-spectrum beta-lactamase (ESBL)-producing Enterobacterales clinical isolates: a retrospective cohort study.

AIMS: Aminoglycoside resistance is frequently detected in extended-spectrum-beta-lactamase (ESBL)-producing Enterobacterales (ESBL-PE), questioning the appropriateness of aminoglycosides as empiric therapy in patients with suspected ESBL-PE infections. Therefore, we aimed to evaluate the frequency of aminoglycoside resistance in patients harbouring ESBL-PE and identify patient-related risk factors associated with aminoglycoside resistance to facilitate early detection of at-risk patients. Cerebral malaria pathogenesis: Dissecting the role of CD4+ and CD8+ T-cells as major effectors in disease pathology.

Cerebral malaria (CM) is a severe complication of Plasmodium falciparum (P. falciparum) infection, with complex pathogenesis involving multiple factors, including the host's immunological response. T lymphocytes, specifically CD4+ T helper cells and CD8+ cytotoxic T cells, are crucial in controlling parasite growth and activating cells for parasite clearance via cytokine secretion. Contrary to this, reports also suggest the pathogenic nature of T lymphocytes as they are often involved in disease progression and severity. CD8+ cytotoxic T cells migrate to the host's brain vasculature, disruptin
