Modeling patient variants of Cnot1 and Cdc42bpb results in distinct forms of congenital diaphragmatic hernia in mice.

Congenital diaphragmatic hernia (CDH) is a severe congenital anomaly characterized by impairment of both diaphragm and lung development in utero. CDH presents as a spectrum of forms and severities, with diaphragm defects arising in the dorsal/posterior region typically correlating with more severe pulmonary disease and higher risk of mortality than those appearing in ventral/anterior regions. The genetic etiology underlying CDH is complex, with many implicated genes showing variable expressivity and incomplete penetrance in both human patients and mouse models. Here we present in vivo validati

4D Flow Acquisition Parameters as Technical Correlates of Hemodynamic Measurements: An Analysis of the multicenter Tetralogy of Fallot Registry of 4D flow Energetics and Kinetics (TREK).

BACKGROUND: Advanced 4D flow cardiovascular magnetic resonance (CMR) biomarkers such as kinetic energy (KE), vorticity (ω), and viscous energy loss (VEL) are increasingly used to characterize abnormal flow in repaired tetralogy of Fallot (rTOF), but their sensitivity to acquisition parameters including magnet vendor, sequence and signal quality has not been defined. We sought to evaluate the real-world variability in 4D flow sequences and technical correlates to 4D flow measurements of the aorta (Ao) and right ventricle, leveraging the multicenter Tetralogy of Fallot 4D flow Registry of Energe

Single-Ventricle Physiology as a Dominant Determinant of Mortality, Resource Utilization, and Pulmonary Vein Reintervention in Total Anomalous Pulmonary Venous Connection: A Large Multicenter Study.

BackgroundChildren with single ventricle (SV) physiology and total anomalous pulmonary venous connection (TAPVC) face high surgical risk. Despite advances in single ventricle care, survival in SV/TAPVC patients has plateaued. We investigated multicenter outcomes, reintervention rates, and healthcare burden in this complex population.MethodsThe Pediatric Health Information System was queried for TAPVC repairs (2015-2024). Infants were stratified by single-ventricle versus biventricular (BiV) anatomy.ResultsAmong 1258 infants undergoing TAPVC repair, 10% (N = 120) had SV and 90% (N = 1138) had B

Changes in birth certificate-reported prevalence of congenital anomalies at live birth during the COVID-19 pandemic: a nationwide study of 26 million United States live births.

BACKGROUND: Although the majority of studies have indicated that maternal severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and coronavirus disease 2019 (COVID-19) vaccination are not associated with neonatal birth defects, it is relevant to examine whether the disease burden changed during the COVID-19 pandemic, as other influencing factors may have also changed. We aimed to compare the birth certificate-reported prevalence of congenital anomalies at live birth before and during the pandemic. METHODS: Using public birth data, all live births from 2016 to 2022 in the Unit

The Spectrum of Mosaic Double Aneuploidy of Monosomy X and Trisomy 18: Two New Cases and Review of the Literature.

INTRODUCTION: Mosaic double aneuploidy (MDA) defines the presence of two chromosomally distinct cells lines derived from the same zygote arising from early cytogenetic events. MDA for monosomy X and trisomy 18 is a rare occurrence described in only 12 individuals in the medical literature to date. CASE PRESENTATION: Here, we present 2 individuals with MDA of monosomy X and trisomy 18 with a Turner predominant phenotype. Congruent with previous reports, the percentage of aneuploid cells is highly variable in differing tissues, without clear genotype-phenotype associations. Molecular mechanism

Impact of Socioeconomic Status and Rurality on the Rate of Prenatal Detection of Major and Critical Congenital Heart Disease in Western Australia.

AIM: Prenatal diagnosis of congenital heart disease (CHD) is associated with reduced infant morbidity and mortality. Location and subsequent socioeconomic status influence rates and timing of prenatal diagnoses of CHD. We investigated the impact of socioeconomic status and rurality on the rate of prenatal diagnosis of critical and major CHD in Western Australia (WA). METHODS: This retrospective cross-sectional study reviewed all critical (surgical intervention ≤ 30 days of life) and major (surgical intervention ≤ 1 year of life) CHD in WA between 2017 and 2021. Patients were identified from t

Beyond conventional anthropometry: Muscle-related indicators and muscle strength in adults with congenital heart disease: A retrospective study with a comparison group.

Anthropometric characteristics and muscle strength remain insufficiently characterized in adults with congenital heart disease (CHD). The aim of the study was to characterize anthropometric indicators and muscle strength in hospitalized adults with CHD and examine differences according to cardiac lesion complexity. Exploratory comparisons with adults without CHD were performed as secondary analyses. This cross-sectional study included 109 hospitalized adults with CHD and an external convenience group of 115 adults without CHD. Body mass index (BMI), mid-upper arm circumference (MUAC), triceps

Effectiveness of three-dimensional printed cardiac models in teaching congenital heart anatomy: A systematic review and meta-analysis of randomized controlled trials.

Congenital heart disease challenges spatial understanding. 3D printed cardiac models are increasingly used, but randomized quantitative evidence remains limited. This review and meta-analysis evaluated their effectiveness for teaching congenital heart anatomy. PubMed, Embase, Scopus, and Web of Science were searched through November 12, 2025. RCTs comparing 3D-printed cardiac models with conventional instruction were included. The primary outcome was objective knowledge performance. SMDs were pooled using random-effects REML models. Risk of bias was assessed with RoB 2 and the certainty of evi

Elevated lithium levels in drinking water are associated with higher prevalence of Wolff-Parkinson-White diagnoses.

BACKGROUND: Supraventricular tachycardia (SVT) and Wolff-Parkinson-White (WPW) are potentially highly symptomatic and, rarely, lethal arrhythmias associated with atrioventricular accessory pathways. While the etiology of accessory pathways remains largely unknown, they are frequently observed with Ebstein's Anomaly, a congenital heart disease associated with maternal medicinal lithium use. Therefore, we evaluated whether in utero exposure to lithium from environmental sources is associated with the prevalence of accessory pathways. METHODS: We conducted a county-level ecological comparison of

Family-Centered Care and Sociodemographic Characteristics as Predictors of Caregiving Burden and Parental Self-Efficacy in Mothers of Infants With Congenital Heart Disease.

BACKGROUND: Congenital heart disease requires complex care, placing mothers at risk of high burden and low self-efficacy. Family-centered care (FCC), emphasizing collaboration with healthcare providers, may alleviate these challenges. However, its predictive role in the Turkish context remains underexplored. METHODS: This cross-sectional, descriptive correlational study included mothers of 131 children aged 0 to 2 years who were followed at a pediatric cardiac surgery unit. Data were collected using the FCC Scale, Zarit Caregiving Burden Scale, and Parental Self-Efficacy Scale. Analyses invol

Multicentric Longitudinal Performance Monitoring of Different Prenatal Cell-Free DNA Screening Technologies in Belgium.

OBJECTIVE: Prenatal cell-free DNA screening is widely used to assess the risk of fetal trisomy 21, 18, and 13. Although commercial and laboratory-developed tests (LDTs) show high accuracy in validation studies, real-world longitudinal performance data are lacking. METHOD: In Belgium, prenatal cell-free DNA screening is a population screening offered to all pregnant people, regardless of their a priori risk. Over 3 years, all confirmatory diagnostic tests after a positive prenatal cell-free DNA screening result were recorded along with the technologies used and their outcomes. Positive predict

Coarctation of the aorta as the presenting cardiovascular manifestation of Williams syndrome in an infant.

Williams syndrome (WS) is a multisystem genetic disorder caused by a 7q11.23 microdeletion and is classically associated with supravalvular aortic stenosis (SVAS) and peripheral pulmonary artery stenosis. We report a male infant in whom discrete juxtaductal coarctation of the aorta (CoA) was incidentally detected during pre-operative evaluation for left inguinal hernia. Echocardiography demonstrated a peak gradient of 25 mm Hg without SVAS or branch pulmonary artery stenosis. Recognition of characteristic facial features prompted genomic evaluation, which confirmed a pathogenic 1.6 Mb 7q11.23

Advancements in echocardiography for congenital heart defects (CHDs): a literature review.

Congenital heart defects (CHDs) are the most prevalent congenital condition globally, affecting approximately 1% of live births and contributing to significant morbidity and mortality. While advancements in diagnostic imaging have improved early detection and management, CHDs remain a leading cause of infant mortality, particularly in low- and middle-income regions. Echocardiography has evolved as a cornerstone in CHD diagnosis, with traditional modalities such as 2D echocardiography, M-mode, and Doppler imaging providing essential structural and hemodynamic assessments. However, these convent

Epilepsy-associated potassium channel KCNT1 is required for multiciliated cell specification in Xenopus.

Pathogenic, presumed gain-of-function variants in the gene KCNT1, which encodes a sodium-activated potassium channel, cause a severe neurodevelopmental disorder with intractable epilepsy. In addition to seizures, affected individuals commonly present with severe respiratory issues and structural heart defects not commonly observed in other genetic pediatric epilepsies, suggesting additional developmental functions for KCNT1 in organs beyond the brain. Here we characterized the spectrum of clinical diagnoses present in a cohort of 46 individuals with genetic variants in KCNT1, ranging from 0 to

Alcohol Exposure Alters Proximodistal Nephron Segmentation During Zebrafish Embryogenesis.

Fetal alcohol spectrum disorders (FASD) encompass a broad range of effects caused by prenatal alcohol exposure. In the United States, conservative estimates place the incidence of FASD at a rate of 5 in 100 births. Children with FASD can have morphological afflictions including congenital defects of the brain, spinal cord, heart, eyes, and skeleton, as well as poor formation of facial structures such as the nose and mouth. Cognitive and behavioral abnormalities such as learning disabilities, socio-developmental delays and emotional disorders are also highly prevalent. There is no known cure fo

Sudden death following a single coronary ostium located below an aortic valve leaflet: A rare autopsy case and review of the literature.

Congenital coronary anomalies are rare but can cause serious cardiovascular events, including sudden death, in both young and old. A forensic autopsy of a 67-year-old woman found dead at her home revealed a single low coronary ostium located under an aortic valve leaflet, with an unobstructed coronary pathway. Myocardial scarring from a previous infarction was identified in the interventricular septum. This case illustrates the importance of recognising congenital coronary anomalies, even in elderly people, and evaluating their possible role in a sudden and unexplained deaths. A review of the
