Population genetics of Plasmodium vivax with transmission decline and rebound in two endemic areas of Papua New Guinea.

BACKGROUND: Global efforts to control and eventually eliminate malaria have been less effective for Plasmodium vivax relative to Plasmodium falciparum due to its unique biology, including dormant liver stages that cause later relapse, and earlier commitment to transmission stages. After the nationwide distribution of long-lasting insecticide treated nets (LLIN) in Papua New Guinea (PNG), P. vivax initially reduced to low prevalence, but again resurged to levels similar to those before LLIN distributions. METHOD: To explore changes in P. vivax population structure and identify sources of resur

Adaptations in Plasmodium tubulin determine distinct microtubule architectures, mechanics and drug susceptibility.

Microtubules are ubiquitous yet diverse cytoskeleton filaments. However, tubulin conservation presents challenges in understanding the origins of diverse microtubule architectures. The mechanisms by which microtubule architecture varies through the life cycle of the malaria-causing parasite Plasmodium are not understood and provide a valuable framework for exploring how intrinsic properties of tubulin contribute to architectural variety. Using parasite-purified tubulin, we determine P. falciparum microtubule structures by cryo-electron microscopy. Parasite-specific sequences change the tubulin

Who uses the only mosquito net? Sex and age disparities.

BACKGROUND: The percentage of the population sleeping under insecticide-treated nets (ITN) increased in sub-Saharan Africa from 3 to 59% between 2000 and 2023. However, achieving universal coverage remains far from realized. Because ITNs are the most effective tools for preventing malaria, it is important to identify barriers to their use and maximize their protective effects. We investigated sex and age disparities in ITN use in a region of western Kenya where Plasmodium transmission remains high. METHODS: We examined sex- and age-related disparities in ITN use under two scenarios: household

Fabrication and evaluation of quinine-loaded chitosan-polycaprolactone core-shell polymeric nanoparticles for the treatment of malaria: an in vitro and in silico study.

Malaria remains a major global health challenge, worsened by the rise of drug-resistant Plasmodium falciparum. Quinine is a cornerstone therapy for severe malaria, yet its clinical use is limited by rapid clearance, dose-dependent toxicity, and a narrow therapeutic window. To address these challenges, we developed chitosan-functionalized poly(ε-caprolactone) core-shell nanoparticles (QN-PCL/CS NPs) for enhanced quinine delivery. Fabricated via double emulsion solvent evaporation, the nanoparticles exhibited favorable characteristics: 312.9 ± 11.7 nm diameter, +30.8 ± 1.5 mV zeta potential, and

Baseline Procalcitonin and C-Reactive Protein Levels in Asymptomatic Individuals From West Africa With and Without P. falciparum Parasitemia.

We assessed baseline C-reactive protein (CRP) and procalcitonin levels in asymptomatic individuals from malaria-endemic West Africa. C-reactive protein remained unaffected by Plasmodium falciparum parasitemia, while procalcitonin (PCT) was more frequently detectable among malaria-positive individuals. These findings support that CRP thresholds remain valid and highlight the need to explore parasite density-PCT associations.

Plasmodium falciparum GTP-cyclohydrolase 1 (Pfgch1) Gene Copy Number Variation and Its Implications in antimalarial resistance from Northeast India.

BACKGROUND OBJECTIVES: Drug-resistant strains of Plasmodium falciparum pose a major challenge to malaria control and elimination. The present study focuses on the GTP-cyclohydrolase 1 (Pfgch1) gene, a key enzyme in the folate biosynthesis pathway, known to play a role in antimalarial (Sulfadoxine-Pyrimethamine, commonly used in combination therapies) resistance. An increase in Pfgch1 copy number is hypothesized to compensate for the mutations in dihydrofolate reductase (Pfdhfr) and dihydropteroate synthetase (Pfdhps) genes associated with Sulfadoxine-Pyrimethamine resistance. METHODS: We anal

Estimating malaria attributable fraction using quantitative PCR in a longitudinal cohort in Eastern Uganda.

Persistent, asymptomatic Plasmodium infections are common in areas of high transmission due to acquired immunity. When asymptomatically infected individuals seek care for a fever caused by something other than malaria parasites, they may test positive for parasites and be incorrectly diagnosed as having clinical malaria. This study used distributions of qPCR parasite densities to estimate the fraction of fever attributable to malaria (malaria attributable fraction, MAF) in a cohort of 659 individuals followed for up to 3 years from three geographically distinct zones in eastern Uganda. In- vitro assessment of the anti- malarial potential of Alpha Onocerin; cytotoxicity and hemolytic effect, multi-stage activity and molecular docking.

The surfacing of resistant strains of Plasmodium falciparum (P. falciparum) against various antimalarials on the market has intensified the need for novel antimalarial agents, particularly those with multistage efficacy. This study investigated the safety and anti-plasmodial efficacy of alpha onocerin (AOC), a triterpenoid compound derived from Huperzia phlegmaria, against both 3D7 and Dd2 lab strains of P. falciparum. In vitro anti-plasmodial assays were carried out to investigate the activity of AOC across trophozoite, schizont, and gametocyte stages. The SYBR Green assay was used to quantif

Structure-Based Screening of Deep-Sea Microbial Metabolites Against Plasmodium falciparum Dihydroorotate Dehydrogenase.

Malaria is a major global health concern caused by Plasmodium parasites, among which Plasmodium falciparum is responsible for the most severe and fatal cases. The emergence of drug resistance to existing antimalarial therapies necessitates the discovery of novel molecular targets and chemically distinct inhibitors. Current study employed an integrated in silico drug discovery pipeline combining high-throughput structure-based virtual screening of 1549 deep-sea marine microbial metabolites with MM-GBSA binding free-energy estimation, QikProp-based ADME/Tox profiling, and 100 ns molecular dynami

The Plasmodium falciparum PPCS is a unique heteromeric complex with prokaryote-like activity and is a target of pantothenate analogs.

Coenzyme A (CoA) is essential for the intraerythrocytic stage of Plasmodium falciparum. Phosphopantothenoylcysteine synthetase (PPCS) catalyzes the second of five steps of the CoA biosynthesis pathway. Most apicomplexan parasites express one PPCS, whereas two PPCSs are expressed in P. falciparum. Here, we demonstrate that the two P. falciparum PPCSs (PfPPCSs) associate into a single, functional PPCS heteromeric complex that, unlike any other eukaryotic PPCS reported to date, is unable to use adenosine 5'-triphosphate. We identify a prokaryote-like helical component of PfPPCS as important for t

Profiling 26S Proteasome Activity of Plasmodium falciparum Monitored by a Live-Cell Assay.

Malaria remains a major global health challenge, driven in part by widespread antimalarial drug resistance in Plasmodium parasites. Artemisinin-based combination therapies (ACTs) are currently the first-line treatment; however, resistance has also emerged. Artemisinin damages parasite proteins, promoting their ubiquitination and subsequent proteasomal degradation. Because inhibitors of the Plasmodium 26S proteasome synergize with artemisinin, the proteasome has emerged as a promising drug target, yet tools to monitor its function in live parasites remain limited. Here, we generated a P. falcip

Antimalarial compounds from Solanum nigrum: In vitro antiplasmodial evaluation and network pharmacology-based investigation of host immunomodulatory mechanisms.

ETHNOPHARMACOLOGICAL RELEVANCE: Malaria is a significant global health issue, made worse by the growing resistance of Plasmodium falciparum to current antimalarial drugs. This highlights the need for new, safe, and multi-target therapeutic agents. Solanum nigrum L., a wild vegetable used in traditional herbal medicine in Cameroon, helps treat several ailments, including some cancers, urinary issues, malaria, fever, and joint pain. However, we have not studied this plant in depth to identify its potential antimalarial extracts and compounds or understand their action mechanisms. AIM OF THE STU

Emerging Artemisinin Partial Resistance in Southern Africa.

A new focus of probable artemisinin-partial resistance (ART-R) has been identified in southern Africa, driven by the P441L mutation in the Plasmodium falciparum Kelch13 gene. This emerging cross-border cluster across Angola, Namibia, and Zambia represents the fourth major ART-R focus identified in Africa, adding to the previously reported foci in East Africa and the Horn of Africa. Findings from three new studies reveal 7.3%, 33%, and 47% prevalence of P441L in specific sites in Angola, Namibia, and Zambia, respectively. The Genomics of Malaria in the Elimination 8 project enabled harmonized,

Targeted Genomic Surveillance Unveils Genetic Variations Linked to Regional Malaria Drug Resistance Dynamics in India.

BACKGROUND: India has made substantial progress in reducing Plasmodium falciparum malaria cases and has set a target to eliminate malaria by 2030. Although artemisinin-based combination therapy (ACT) treatment remains effective, tracking regional differences in genetic variants associated with antimalarial resistance is required for effective drug policy implementation. METHODS: We analyzed 238 P. falciparum clinical samples from 6 Indian states by sequencing 15 parasite genes associated with reduced drug effectiveness. The method involved nanopore sequencing of target gene amplicons derived

Microfluidic platform for automatic quantification of malaria parasite invasion under physiological flow conditions.

Understanding the impact of forces generated by blood flow on biological processes in the circulatory system, such as the invasion of human red blood cells by malaria parasites, is currently limited by the lack of experimental systems that integrate them. Recent systematic quantification of the growth of Plasmodium falciparum, the species that causes the majority of malaria mortality, under a range of shaking conditions has shown that parasite invasion of erythrocytes is affected by the shear stress to which the interacting P. falciparum merozoites and their target red blood cells are exposed.

Hydroxamate-based inhibitors reveal structural determinants of selectivity for Plasmodium falciparum aminopeptidase P.

The malarial parasite, Plasmodium falciparum (Pf), utilizes aminopeptidases in the breakdown of hemoglobin-derived oligopeptides to release amino acids for protein synthesis during growth and asexual reproduction of erythrocytic stages of the parasite. However, a N-terminal peptide bond that involves proline is difficult to hydrolyze. Aminopeptidase P (APP) is capable of cleaving peptide bonds with proline in the second position. Inhibition of PfAPP is therefore an attractive strategy for developing therapeutics for the treatment of malaria by limiting the supply of amino acids at the erythroc
