The identification of antiplasmodial lead compounds from the aerial parts of Vernonia glabra (Asteraceae) was carried out using a bioassay-directed protocol. The methanolic extract of the plant was partitioned into four main fractions using n-hexane, dichloromethane, ethyl acetate, and n-butanol. These fractions were then tested for their antiplasmodial activity against Plasmodium falciparum 3D7. The bioassay results indicated that the dichloromethane-soluble fraction had the highest potency, with an IC50 of 8.4 ± 0.5 μg/mL. Further purification of the active fraction through successive column

Artemisinin-based combination therapies (ACTs) remain the mainstay of treatment for Plasmodium falciparum malaria, despite reports of ACT treatment failure. ACTs consist of an artemisinin and a longer-lived partner drug, which is often a quinoline. Given that heme is central to the mechanism of action of artemisinins and some quinolines, we hypothesized that these antimalarials would exhibit strong drug-drug interactions. Previous studies using standard 48 hr or 72 hr assays identified additive to mildly antagonistic interactions between artemisinins and quinolines. Here, we sought to re-evalu

Plasmodium falciparum malaria relies on antigenic variation mediated by mutually exclusive expression of var genes. Repressed var genes cluster together and are bound by PfHP1 (Plasmodium falciparum heterochromatin protein 1), but the regulatory mechanism remains unclear. Here, we show that PfHP1 undergoes phase separation in vitro and compartmentalizes DNA. This process is tunable by RNA and PfH2A.Z. Our single-molecule assays show that PfHP1 preferentially compacts AT-rich DNA. We identify point mutations in the intrinsically disordered region of PfHP1 that disrupt phase separation and DNA c

BACKGROUND: Despite ongoing reports of malaria from health facilities in central Ethiopia, entomological evidence on local vectors remains scarce. This study investigated Anopheles species composition, host preferences, and entomological inoculation rates in this urban setting.
METHODS: A repeated cross-sectional and longitudinal entomological study was conducted in six districts (woredas), each of which had access to one of the three health centers (Akaki, Gelan, and Serti). Adult mosquitoes were collected from the houses of individuals with microscopically confirmed malaria, who served as t

The plant Chenopodium opulifolium is traditionally consumed as a food and for treating ailments. Despite its use in Uganda for treating malaria, there are no scientific studies on its phytochemicals and biological potential to ascertain its folkloric claims. In this study, bioassay (antiplasmodial activity) – guided fractionation, isolation and identification of the phytoconstituents of the leaves of C. opulifolium were carried out. The identified compounds were further investigated via in silico docking to propose the mechanism of action of the extracts. The leaves were extracted sequentially

CRISPR/Cas9-based gene editing of the malaria parasite Plasmodium falciparum has emerged as a transformative tool for advancing functional studies on parasite biology and identifying new therapeutic targets. Currently applied CRISPR/Cas9 methodologies depend on a limited set of heterologous drug resistance markers for the selection of transgenic parasites, which restricts the potential for iterative genetic modifications. Here, we developed a heterologous marker-free CRISPR/Cas9 gene editing strategy (CRISPR/Cas9pyrR) for P. falciparum based on the simultaneous editing of a gene of interest an

BACKGROUND: Ethiopia experienced a marked resurgence of malaria in 2024. Artemisinin-based combination therapies (ACTs) are first-line treatment for uncomplicated Plasmodium falciparum malaria and threatened by the emergence of artemisinin partial resistance (ART-R), associated with mutations in the P. falciparum kelch13 (k13) gene, that could undermine treatment efficacy and accelerate transmission.
METHODS AND MATERIAL: We used the national Public Health Emergency Management (PHEM) surveillance system to characterize malaria resurgence and further investigate antimalarial drug resistance ma

The emergence and spread of drug resistance threatens malaria control in Uganda. This study reports new data on key polymorphisms associated with antimalarial drug sensitivity at 32 malaria reference centers in Uganda in 2023 and 2024. Samples were collected from patients aged >6 months presenting with uncomplicated falciparum malaria, sequenced using the MAD4HatTeR panel and Illumina platforms. Multiple validated or candidate K13 mutations associated with artemisinin partial resistance were detected at ≥20% prevalence at one or more sites across multiple timepoints. The K13 mutations A675V an

In malaria-endemic regions, women remain vulnerable to Plasmodium falciparum infection at the time of delivery. However, the immunological mechanisms underlying infection-associated inflammation in primigravid women remain poorly characterized. This exploratory study investigated cytokine-based immune profiles reflecting malaria infection status at delivery. We assessed 33 primigravid women from Nanoro, Burkina Faso (mean age 19 years; range 18-20.5) at childbirth. Antibody responses to P. falciparum antigens (PfCSP, PfAMA-1, and EBA-175) and plasma levels of cytokines (IL-4, IL-10, IL-6, TNF-

Malaria remains a major global health burden, with an estimated 282 million cases and 610,000 deaths reported in 2024, disproportionately affecting children under five years of age and pregnant women in sub-Saharan Africa. Although antimalarial drugs are highly effective at clearing infections, their reliance on timely diagnosis and treatment limits their scalability as a population-wide control strategy. Vaccines therefore represent a critical tool for reducing malaria-associated morbidity and mortality, as well as interrupting parasite transmission, by inducing durable protective immunity. H

BACKGROUND: Malaria prevention in Senegal includes the use of Seasonal Malaria Chemoprevention with sulfadoxine-pyrimethamine plus amodiaquine. Prolonged use of sulfadoxine-pyrimethamine, however, can select for mutations in the Plasmodium falciparum mutations in pfdhfr and pfdhps genes.
METHODS: We analyse dried-blood-spot samples collected during therapeutic-efficacy studies in two SMC districts (Kolda, Kédougou) between 2020 and 2023. Using targeted amplicon deep sequencing we determine the prevalence of pfdhfr and pfdhps mutations by year and site.
RESULTS: The pfdhfr triple mutant genot

Malaria blood stages build and maintain an intricate system of membranes during their cycle of rapid growth and schizogony (daughter-cell formation), requiring precise mechanisms of lipid synthesis and trafficking. Lipid transfer proteins (LTPs) at ER membrane contact sites (MCSs) have emerged as key for lipid distribution processes but remain largely unexplored in protozoans. Here we use the ER adapter VAP to identify essential mechanisms of lipid transfer at ER-MCSs in P. falciparum. One PfVAP-interacting LTP is the bridge-like PfVPS13L1, which allows bulk flow of lipids between two apposed

UNLABELLED: Large Language Models (LLMs) are being rapidly deployed in higher education, yet institutions lack integrated, theoretically grounded frameworks to guide responsible adoption. While meta-analytic evidence reports large positive effects on academic performance (g = 0.867; Wang and Fan 2025), emerging research reveals a "cognitive paradox": performance gains may coincide with diminished metacognitive accuracy and self-regulation-a gap no existing framework comprehensively addresses across adaptation design, data governance, pedagogical targeting, and learner agency simultaneously.

Sequestration of Plasmodium falciparum-infected erythrocytes (IE) in the microvasculature is a major virulence determinant. While the sequestration of mature stage parasites (trophozoite and schizonts) to vascular endothelium is well established, the conditions that promote ring-stage IE sequestration are less understood. Here, we observed in ring-stage parasites that febrile exposure increased transcript levels of several exported parasite genes involved in the trafficking of the P. falciparum erythrocyte membrane protein 1 (PfEMP1) ligand responsible for adherence to the endothelium of blood

Malaria remains a major global health problem, particularly in Africa. Although transmission is primarily vector-borne, Plasmodium can also be transmitted through blood transfusion, resulting in post-transfusion malaria (PTM). PTM is typically linked to asymptomatic parasitemia in donors and, while uncommon, may be rapidly progressive and potentially fatal. We conducted a systematic review to summarize published PTM cases in African countries considered non-endemic and to identify risk factors and prevention gaps relevant to transfusion services. We searched PubMed/MEDLINE, Google Scholar, and

OBJECTIVE: Malaria remains a significant global health challenge, with Plasmodium falciparum causing the most severe cases and deaths, particularly in sub-Saharan Africa. An effective universal or pan vaccine is needed to augment current concerted control efforts. This in silico study investigated the global population coverage of human leukocyte antigen (HLA) that recognize experimentally validated CD8+ T cell epitopes derived from three P. falciparum antigens (CSP, AMA1 and TRAP), focusing on their potential for inclusion in a globally effective multi-epitope malaria vaccine.

While vaccines are central to eradicate malaria, they remain elusive, with numerous malaria vaccine candidates showing limited efficacy in Phase II and III studies. Controlled human malaria infection studies have showed that human volunteers, immunized with Plasmodium falciparum sporozoites under drug cover, were protected experimentally from a subsequent challenge. Here, to identify new targets associated with protection, we utilized a previously developed screening approach, where we screened sera from protected and non-protected individuals against newly included hypothetical antigens in a

The ongoing rise in antimalarial drug resistance underscores the urgent need for new drug candidates that specifically target novel mechanisms. Malaria parasites employ various epigenetic strategies to regulate gene expression throughout their complex life cycle, with histone lysine acetylation and methylation being well-studied and targeted by new antiplasmodials. By contrast, arginine methylation remains poorly explored. Plasmodium falciparum possesses three protein arginine methyltransferases (PRMTs) that maintain a unique and combinatorial histone arginine methylation landscape. Here, we p
