Antiplasmodial Compounds from Clerodendrum rotundifolium Oliv. and Oral Toxicity Assessment of the Leaf Extracts.

ETHNOPHARMACOLOGICAL RELEVANCE: The growing resistance of Plasmodium strains to known conventional antimalarial drugs has increased challenges of malarial treatment in some parts of the world. Although plants are traditionally used to treat malaria, the antiplasmodial phytochemicals of many species remain uncharacterized. This study aimed to scientifically validate the traditional use of Clerodendrum rotundifolium Oliv. (Lamiaceae) in treating malaria by the herbalists of Prometra-Uganda. The antiplasmodial activity and toxicity of its leaf extracts were evaluated and the chemical structures o

Plasmodium falciparum Niemann-Pick Type C1-Related protein relies on its physicochemical properties for membrane contact site localization required for cholesterol homeostasis.

The Niemann-Pick Type C1-Related protein of the malaria parasite Plasmodium falciparum, PfNCR1, is a promising anti-malarial drug target facilitating cholesterol homeostasis at the interface of the malaria parasite with its host-red blood cell. PfNCR1 is localized to otherwise functionally uncharacterized regions covering ~half of the host-parasite interface. These regions are defined by exceptionally narrow membrane contact sites, leaving only ~3-4 nm vertical aqueous space in between the membranes. Determining the origin and functional consequence of localization to the closely apposed membr

The Role of Haemoglobin Variants and Red Blood Cell Polymorphisms in Asymptomatic Malaria: A Cross-Sectional Study.

BACKGROUND AND AIMS: Human phenotypes, particularly blood group polymorphisms (ABO) and haemoglobin variants (HbAA and HbAS), have been widely studied in relation to clinical malaria. However, their role in asymptomatic malaria remains underexplored. This study therefore, investigated the association of these phenotypic variants with the burden of asymptomatic malaria among Ghanaians. METHODOLOGY: Community members without clinical signs of malaria were recruited from four endemic communities. Whole blood samples were screened for Plasmodium falciparum histidine-rich protein 2 (PfHRP2). ABO b

Development and Synthesis of Bioactive (3-Arylediene-1-phenyl-1H-pyrazol-4-yl)acryloyl-4-hydroxy-6-methyl-2H-pyran-2-ones With Anti-Malarial and Anti-Inflammatory Activities.

Chalcones are versatile scaffolds with broad pharmacological relevance, particularly as anti-inflammatory and anti-malarial agents. In the current investigation, a series of (3-arylediene-1-phenyl-1H-pyrazol-4-yl)acryloyl-4-hydroxy-6-methyl-2H-pyran-2-one analogs/1,3-diarylprop-2-en-1-ones were successfully synthesized, leveraging dehydroacetic acid (DHA) as the precursor molecule. The choice of DHA is strategic as it is a bioactive and highly versatile scaffold bearing a 4-hydroxy-6-methyl-2H-pyran-2-one moiety, which enhances structural diversity and provides multiple reactive sites for func

Telomere-to-telomere genome assembly of Microsporidia sp. MB, a microsporidian symbiont of Anopheles coluzzii isolated from Burkina Faso.

BACKGROUND: Microsporidia sp. MB is an intracellular parasite of anopheline mosquitoes identified across the African continent. Microsporidia sp. MB infections appear to have no significant effect on mosquito fitness and can vertically transmit, whilst infected individuals have exhibited significantly reduced levels of Plasmodium falciparum transmission. These combined characteristics make Microsporidia sp. MB a promising candidate for use in malaria control. A comprehensive genome would greatly facilitate investigation into the evolution and biological pathways underlying important phenotypes

Acyl Carrier Protein is Essential for Apicoplast Biogenesis in Malaria Parasites Independent of Fatty Acid Synthesis.

Acyl carrier protein (ACP) and its 4-phosphopantetheine prosthetic group canonically function as the soluble scaffold for acyl chain assembly and elongation during type-II fatty acid synthesis (FASII). Plasmodium malaria parasites retain a FASII pathway in the apicoplast organelle that has been the subject of considerable scrutiny and confusion. Although apicoplast FASII is essential for P. falciparum growth within mosquitoes and the human liver, this pathway is dispensable and largely inactive in blood-stage parasites that can scavenge host fatty acids. In contrast to FASII enzymes that can b

Double Mutations in Plasmodium falciparum Kelch13 drive resistance to next-generation artemisinin derivatives in malaria parasites.

New antimalarial compounds are urgently required to overcome artemisinin partial resistance that has emerged in Asia and now Africa. Ozonides are promising next-generation artemisinins that offer the improved pharmacokinetic property of a prolonged in vivo half-life. To assess the potential for parasite resistance to ozonides in an artemisinin-resistant background, we subjected Cambodian Kelch13 (K13) mutant parasites to increasing artefenomel (OZ439) pressure up to in vivo physiological concentrations. Whole-genome sequencing identified a novel non-propeller K13 A212T mutation in OZ439-resist

Molecular surveillance of Falciparum malaria in Rwanda: Shifts in parasite prevalence and risk factors between the 2014-15 and 2019-20 Rwanda Demographics and Health Surveys.

Rwanda is a malaria endemic country and a focal point for emerging Plasmodium falciparum artemisinin partial resistance (ART-R). While Demographic and Health Surveys (DHS) provide both national and province-level representative data, malaria testing in Rwandan DHS (RDHS) studies has been limited to a subset of adult women and children under 5 years using RDT and/or microscopy. Recent work using ultra-sensitive quantitative real time PCR on residual dried blood spots (DBS) from the 2014-15 RDHS revealed a significantly higher P. falciparum prevalence than detected by standard DHS diagnostics. Malaria: 11-Year Experience With Imported Cases at a German University Hospital and Epidemiological Trends Amid the COVID-19 Pandemic.

BACKGROUND: By the mid-20th century, Europe eradicated endemic malaria. However, the significance of imported infections in nonendemic regions is growing. These cases often constitute the majority, can sustain transmission, trigger relapses, and increase mortality in these areas. METHODS: We conducted a retrospective, single-center study of patients with malaria treated at the University Hospital Regensburg located in the State of Bavaria, Germany, between 2012 and 2023. Our single-center epidemiological data were analyzed in the context of the COVID-19 pandemic and compared with overall data

Deciphering Membrane Protein Complexes in Plasmodium falciparum Gametocytes via Integrative Structural Systems Biology.

Malaria is caused by protozoan parasites of the genus Plasmodium that proliferate asexually in human erythrocytes. During each replication cycle, a small fraction of the parasites differentiates into gametocytes. These sexual gametocytes are the only stages of the parasite that can infect the mosquito vector and transmit malaria. Their progression and maturation depend on a profound remodeling of the erythrocyte. This is achieved by the export of parasite proteins into the erythrocyte, leading to structural and mechanistic changes in the cytoskeleton and membrane of the host cell, crucial for

Plasmodium falciparum ring-infected erythrocyte surface antigen 3 (PfRESA3) is a cytoskeleton-interacting protein and potential co-chaperone.

Plasmodium falciparum exports proteins into host erythrocytes for survival, but the roles of many of these proteins remain unexplored. Here, we used recombinant protein constructs and antibodies corresponding to Plasmodium falciparum ring-infected erythrocyte surface antigen-3 (PfRESA3) to identify it as a dense granule merozoite protein exported early to the inner erythrocyte membrane, where it associates with the cytoskeleton. Constructs lacking the J domain bound inside-out vesicles, implicating PRESAN and DnaJ-X in cytoskeletal interactions. Recombinant PfRESA3 constructs stimulated the AT

Phase I/IIa study to assess the safety, immunogenicity and efficacy of ChAdOx1-MVA vectored vaccines expressing a novel liver-stage malaria dual antigen LS2 by sporozoite challenge in malaria-naïve adults.

BACKGROUND: Induction of CD8 + T-cells using viral vectors is a promising strategy in developing effective vaccines against pre-erythrocytic malaria. A recent comparative assessment of candidate antigens using this approach in a mouse model had identified Liver Stage Antigen 1 (LSA1) and Liver Stage Associated Protein 2 (LSAP2) as more protective than TRAP and CSP antigens, which have been the dominant focus of clinical testing. We proposed that combining these within a novel dual antigenic insert (LS2), encoded alongside an orthologous immunogenic domain from invariant chain in ChAdOx1, and t

Indole Peptidomimetics Show Potent and Selective Activity against Drug-Resistant Plasmodium falciparum.

Malaria remains a global health challenge exacerbated by emerging drug-resistant Plasmodium falciparum strains. Here, we report the design, synthesis, and biological evaluation of indole-based peptidomimetics against P. falciparum sensitive and multidrug-resistant strains. Structure-activity relationship analysis indicated that aromatic and halogen substituents, as well as modifications at the indole nitrogen and amide linkage, strongly influence potency and selectivity. LSPN954 (4i) and LSPN959 (4k) emerged as front-runner compounds, displaying low micromolar potency against the sensitive str

Heat shock protein Gp96 (Grp94) in malaria: functional insights at the host-parasite interface and therapeutic perspectives.

Heat shock protein Gp96 (also known as Grp94 or endoplasmin) is the endoplasmic reticulum (ER)-resident paralog of the Hsp90 family and a central regulator of ER proteostasis and immune receptor biogenesis in mammalian cells. By controlling the folding, quality control, and trafficking of a restricted yet functionally critical set of client proteins, including Toll-like receptors, integrins, and immunoglobulins, Gp96 plays an essential role in innate immunity and inflammatory signaling. In the context of malaria, accumulating evidence suggests that host-derived Gp96 is involved in immune activ

Malaria in the 21st Century: Global Disease Burden, Epidemiological Insights, and Strategic Control Approaches.

Malaria remains a major public health issue worldwide and a repeated cause of illness and death in tropical and subtropical areas. It is caused by protozoan parasites of the genus Plasmodium and transmitted through bites of infected female Anopheles mosquitoes, but it can also be transmitted via blood transfusions, organ transplants, and congenitally from mother to child. Despite decades of intervention efforts, millions of new cases and hundreds of thousands of deaths still occur each year, primarily in low- and middle-income countries. This review summarizes current epidemiological data on t

upsML: A high-accuracy machine learning classifier for predicting Plasmodium falciparum var gene upstream groups.

Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1), encoded by the hypervariable var gene family, is central to malaria pathogenesis, influencing both disease severity and immune evasion. Classifying var genes into upstream groups (upsA, upsB, upsC, upsE) is important for understanding parasite biology and clinical outcomes, but remains challenging, especially with partial sequences, such as the DBLα tag or RNA-Seq assemblies. We developed upsML, a machine-learning-based classifier trained on 2,530 curated var genes, to accurately assign upstream groups based on sequence features fr

Congenital and Neonatal Malaria in Ethiopia: A Narrative Review.

BACKGROUND: Malaria remains a major public health problem in Ethiopia. Congenital malaria results from transplacental transmission of malaria parasites from mother to fetus before or during delivery, whereas neonatal malaria is acquired after birth, typically through mosquito bites within the first 28 days of life. Both conditions are associated with significant neonatal morbidity and mortality in endemic settings. Despite Ethiopia's high malaria burden, evidence on congenital and neonatal malaria remains fragmented and limited. OBJECTIVE: This narrative review synthesizes the existing litera

The hidden malaria: Misidentification of Plasmodium malariae and Plasmodium ovale in low-transmission setting targeted for elimination in Ethiopia.

BACKGROUND: A quartan malaria caused by Plasmodium malariae (Pm) and a tertian Plasmodium ovale (Po) malaria has been largely overlooked in Ethiopia due to milder clinical symptoms, lower prevalence, and the frequent misdiagnosis of these species during routine microscopic. This study estimated the prevalence of Pm and Po infections misdiagnosed as P. vivax (Pv) in selected health facilities in southwestern Ethiopia. METHODS: A cross-sectional study was conducted among study participants who were positive for Pv by microscopy or rapid diagnostic test (RDTs) and who were attending health facil

Genome-wide detection of conserved stage-specific RNA editing events for nuclear genes in the Plasmodium falciparum 3D7 malaria parasite.

RNA editing is an important post-transcriptional modification of RNA. While transcriptional variation is a well-studied phenomenon in Plasmodium, post-transcriptional modifications, such as RNA editing, have not received as much scrutiny. We detected genome-wide RNA editing in a developmental stage-specific manner at 16 h, 24 h, 32 h, and 40 h of tightly synchronized P. falciparum 3D7 by using the RNA editing computational approaches REDItools1-De novo and REDItools2. REDItools1-De novo approach revealed extensive A-to-G, G-to-A, and T-to-C type variations in almost all stages. With the REDIto
