New mercaptoacrylate derivatives and other constituents from the stem bark of Senegalia polyacantha: in vitro antioxidant and antiplasmodial activities with in silico target validation.

Antimalarial resistance remains one of the main obstacles in the fight against malaria, which continues to be a leading cause of death in Africa. This study aimed to evaluate the antiplasmodial, cytotoxic, and antioxidant properties of the extracts and compounds from the stem bark of Senegalia polyacantha (Willd.) Seigler & Ebinger (syn. Acacia polyacantha Willd.). The DCM/MeOH crude extract (SPDM) of S. polyacantha was further extracted with EtOAc (SPEA) and chromatographed, leading twenty-four compounds (1-24) which were all characterized using NMR and MS data. The SPDM, SPEA, and compounds

Pyrophosphate homeostasis in multiple subcellular compartments is essential in Plasmodium falciparum.

UNLABELLED: Pyrophosphate is a byproduct of numerous cellular reactions that use ATP or other nucleoside triphosphates to synthesize DNA, RNA, protein, and various molecules. Its degradation into monophosphate is thus crucial for the survival and proliferation of all life forms. The human malaria parasite Plasmodium falciparum encodes two classes of pyrophosphatases to hydrolyze pyrophosphate. The first consists of P. falciparum proton pumping vacuolar pyrophosphatases (e.g., PfVP1), which localize to the parasite plasma membrane and work as proton pumps. The second includes P. falciparum solu

Malaria in Bangladesh, 2000-2025: Navigating the Headwinds From Control to Elimination.

BACKGROUND AND AIMS: Bangladesh has dramatically reduced its malaria burden since 2000, confining the disease to specific geographic zones. However, the national 2030 elimination goal is now threatened by a complex interplay of biological, ecological, and social factors. This narrative review synthesizes the country's malaria landscape from 2000 to 2025, analyzes the headwinds to elimination, and proposes a strategic framework that integrates genomic surveillance to address the nuanced challenges of residual transmission for the "last-mile" effort. METHODS: A narrative review of scientific li

Haplotype diversity of Plasmodium falciparum circumsporozoite protein prior to malaria vaccine implementation in Kaelé Health District, Cameroon, 2022-2023.

Malaria remains a major public health concern in Cameroon, with Plasmodium falciparum responsible for most morbidity and mortality, particularly among children under five. In response to rising cases, Cameroon began implementing the RTS,S/AS01 malaria vaccine in early 2024. Given the vaccine's strain-specific efficacy, understanding haplotypic diversity is critical for evaluating long-term impact. We analyzed 100 P. falciparum-positive dried blood spots collected in Mapoussere, Kaele Health District (2022-2023). Using the 4CAST amplicon sequencing assay, we targeted four genes: csp (circumspor

Antimalarial Potency of Tinospora crispa Stem Extract and Artesunate Against Plasmodium berghei: A Study on Combination Therapy.

Malaria continues to be a significant global health burden, with over 249 million cases and 619,000 deaths reported in 2022, primarily affecting tropical and subtropical regions. The emergence of drug-resistant strains of Plasmodium falciparum has reduced the efficacy of artemisinin-based combination therapies (ACTs), necessitating the exploration of alternative treatment strategies. This study investigates the antimalarial efficacy of Tinospora crispa extract (TCE) and artesunate (ART) against Plasmodium berghei ANKA infection in ICR mice. Both ART and TCE demonstrated dose-dependent antimala

Dynamic regulation of long non-coding RNAs across asexual and gametocyte development in Plasmodium falciparum.

Long non-coding RNAs (lncRNAs) are critical regulators of gene expression in eukaryotes. Short reads from Illumina sequencing, reverse transcriptase artefacts, and incomplete second-strand degradation in strand-specific cDNA libraries hamper genome-wide identification of lncRNAs, especially in gene-dense genomes such as Plasmodium. Here, we integrated long-read Oxford Nanopore Technology direct RNA sequencing, ribosome profiling, and single-cell transcriptomics to generate a robust and stage-specific characterization of P. falciparum lncRNAs. We generated comprehensive annotations of lncRNAs e

From Hit to Lead: Systematic Optimization of the Quinazolinedione Scaffold as Potent and Noncytotoxic Antimalarial Agents.

The emergence of Plasmodium falciparum resistance to artemisinin-based therapies necessitates the urgent discovery of new antimalarials with novel scaffolds and mechanisms. The quinazolinedione scaffold, exemplified by the TCAMS hit TCMDC-125133, was identified as a promising, noncytotoxic starting point. Herein, we report a systematic lead optimization campaign commencing from a simplified and synthetically tractable analogue, compound 9 (IC50 = 586 nM), which was previously reported by our group. A focused library of 57 novel derivatives was designed and synthesized via a concise 5-step rout

Protective immunity against malaria by a nanoparticle CIS43-based junctional vaccine alone or in combination with R21.

Repetitive display of the major repeats of the Plasmodium falciparum circumsporozoite protein (PfCSP) is the basis for two WHO-recommended vaccines: RTS,S/AS01 and R21/Matrix-M. Recently, however, the CIS43 monoclonal antibody that preferentially targets the junctional region of PfCSP has been shown to be highly protective in humans, highlighting its junctional epitope as a key vaccine target. Here, we develop a vaccine based on tandem repeats of the junctional epitope displayed on a self-assembling nanoparticle and compare this CIS43-based junctional vaccine alone or in combination with the b

Defining the signature of deformable and infectious Plasmodium falciparum gametocytes.

BACKGROUND: One of the major challenges for malaria elimination is combating the highly efficient spread of the disease. Despite progress in understanding the development of malaria transmission stages, there remain many unanswered questions about how gametocytes transition from being immature to infectious. In Plasmodium falciparum, immature gametocytes are rigid and sequester outside of circulation in the extravascular space of the bone marrow, while deformable, mature stages are found in circulation and transmitted to mosquitoes. It is currently unclear whether deformable gametocytes are im

Stabilizing Plasmodium falciparum proteins for small molecule drug discovery.

Early-stage drug discovery relies on the availability of stable protein for reliable biophysical characterization of ligand binding. However, many Plasmodium falciparum proteins are challenging to produce in heterologous systems, which limits their experimental utility. To address this, we tested whether ProteinMPNN-guided sequence design could generate stabilized surrogate constructs that retain wild-type-like structure and binding thermodynamics. Designs were generated with constraints to maintain conserved and binding-site residues for three therapeutically relevant targets: PfBDP1-BRD, PfB

Repositioning Antibiotics Against Plasmodium falciparum RAD5 and WD11 as Novel Antimalarial Targets.

BACKGROUND: The emergence of artemisinin resistance in Plasmodium falciparum threatens the sustainability of malaria control programs and underscores the need for new therapeutic strategies. Recently, two proteins, RAD5 (PfRAD5) and WD40-repeat protein 11 (PfWD11), have been implicated in parasite survival and drug resistance. Their druggability, however, remains underexplored. METHODS: We integrated computational and experimental approaches to assess PfRAD5 and PfWD11 as antimalarial targets. Structural models were retrieved from AlphaFold and aligned with orthologues across Plasmodium speci

Repurposing Drugs for Malaria through a Human Dose Prediction: A Case Study with Berzosertib.

Repurposing drugs whose clinical safety has been established offers a valuable approach to reduce the cost and time associated with the development of new drugs for malaria. Here, we investigate the potential to repurpose the anticancer kinase inhibitor berzosertib for the treatment of malaria, by assessing whether a predicted efficacious human dose for malaria is within the clinically established safety and tolerability profile. First, an oral dose fractionation study was performed in a Plasmodium falciparum-infected humanized mouse model to evaluate the antimalarial pharmacokinetic-pharmacod
