With resistance to current frontline antimalarial treatments rapidly emerging in malaria endemic regions, there is an urgent need to identify new antimalarial compounds with novel mechanisms of action. Currently, no clinically used antimalarials target erythrocyte invasion, the process by which the short-lived, extracellular merozoite form of Plasmodium falciparum parasites enters a host erythrocyte. Following invasion, the parasite grows and then divides to form a schizont, containing merozoites that exit the host cell and invade new erythrocytes. To identify novel egress and invasion inhibit

Artemisinin resistance in Plasmodium falciparum is now well established in three continents and challenges the efficacy of antimalarial drug regimens. This study examined whether methylene blue, an ancient antimalarial drug with a broad spectrum of activity against Plasmodium blood stages, retains its activity against the young rings of artemisinin-resistant parasites. Clinical isolates carrying kelch13 wild type (n = 3), R561H (n = 9), or P441L (n = 10) genotypes were tested with a modified ring survival assay whereby 0- to 3-h post-invasion rings were exposed to a range of methylene blue con

BACKGROUND: Due to the increasing resistance of malaria parasites to antimalarial drugs, medicinal plants are increasingly being employed to cure malaria. The purpose of this study is to establish the cytotoxicity, antioxidant, and antiplasmodial activity of a mixture of Terminalia macroptera and Erythrina sigmoidea extracts, two plants that have been traditionally used together in the management of malaria in Cameroon's Western Region.
METHODS: The stem bark extracts of both plants were formulated according to established protocols. Previous studies reported the use of blends of E. sigmoidea

INTRODUCTION: In low-transmission malaria settings, routine surveillance often fails to detect asymptomatic infections in partially immune populations. P(Detect), defined as the proportion of infections captured by surveillance systems, serves as a proxy for both population immunity and surveillance completeness. We evaluated whether antibody responses to Plasmodium falciparum antigens could classify low vs. high P(Detect) populations and identify surveillance blind spots.
METHODS: We analyzed data from 5,300 seropositive individuals across 32 villages in The Gambia following mass drug admini

The life cycle of Plasmodium falciparum is complex, involving asexual and sexual reproduction in humans and mosquitoes, respectively. The parasite transmission from humans to mosquito vectors requires the formation of female and male gametes through gametogenesis. The MKT1 domain proteins are key molecules involved in posttranscriptional gene regulation and cellular differentiation in protozoans, plants, and animals, and the presence of an MKT1 ortholog suggested a possible role in stage differentiation in Plasmodium. Here, we report that PfMKT1 was expressed in both asexual and sexual stages.

Our group explores the antimalarial compounds from the Streptomyces culture library. Pilot screening showed significant antimalarial activity in Streptomyces antibioticus HUT6035 (synonym of strain NBRC3117). We here isolated antimalarial compounds through large-scale fermentation and subsequent purification using chromatographies. Strain HUT6035 accumulated actinomycins X2 and X0ß, which were 4-oxo and 4-hydroxy proline derivatives of actinomycin D, respectively. The former showed an antimalaria activity (IC50 = 0.241 nM) and a cytotoxicity (CC50 = 6.71 nM), while the latter showed IC50 = 7.6

Malaria constitutes a major public health burden in sub-Saharan Africa. It remains a key health concern and the leading cause of death in children under five in Benin. Since October 2021, the World Health Organization has recommended the use of malaria vaccines for the prevention of Plasmodium falciparum malaria in children living in malaria endemic areas, prioritizing areas of moderate and high transmission in sub-Saharan Africa. However, with the exception of Ghana, there is a scarcity of studies modelling the potential impact of the RTS,S/AS01 vaccine in the context of West Africa. A compar

Plasmodium falciparum parasites with deletions of the histidine-rich protein 2 and 3 (hrp2 and hrp3) genes evade detection by common rapid diagnostic tests (RDTs) and pose a growing threat to malaria control. While these deletions have emerged in multiple regions globally, the evolutionary forces driving their spread remain unclear. Here, we analyze 1,215 P. falciparum samples collected between 2003 and 2018 in Loreto, Peru. This region experienced a major decline in malaria transmission following the Project for Malaria Control in Andean Border Areas (PAMAFRO) and now harbors a high proportio

Cerebral malaria (CM) due to Plasmodium falciparum ( Pf ) infection is a major cause of death in African children. Bradykinin (BK) is a mediator of vasogenic edema. It could contribute to the pathogenesis of central nervous system malaria in Kenyan children and P. berghei ANKA ( PbA ) infected C57BL/6J mice with experimental cerebral malaria. Cleaved plasma high molecular weight kininogen (cHK) is a marker for prior BK release. 40% of children with central nervous system malaria had plasma cHK versus 18% of children with uncomplicated malaria. Wild-type PbA -infected mice had circulating plasm

BACKGROUND: Cameroon bears a high malaria burden, and sulfadoxine-pyrimethamine (SP) remains widely used for intermittent preventive treatment in pregnancy despite the adoption of artemisinin-based combination therapies. Continued programmatic use of SP under sustained malaria transmission exerts selective pressure for resistant Plasmodium falciparum strains. This study investigated the molecular epidemiology of resistance-associated mutations in the P. falciparum dihydropteroate synthase (Pfdhps) gene in two health facilities in the Mbouda Health District, Cameroon.
METHODOLOGY: Blood sample

The aspartic protease plasmepsin X (PMX) of the parasite Plasmodium is a promising drug target for novel malaria therapies. Two potent inhibitors of PMX are WM382 and WM4, which both include a guanidinium group that is in contact with the two catalytic aspartates of PMX in the bound complexes. In structural representations of the inhibitors, the guanidinium group is typically depicted as uncharged. However, pKa predictions with standard tools presented in this article indicate that the guanidinium groups of WM382 and WM4 are protonated and, thus, positively charged in the bound complexes. This

Malaria research is facing a concerning challenge due to the continuously increasing annual reported number of cases and deaths, along with the Plasmodium parasite's resistance against current first-line treatments, which emphasise the need for novel effective antimalarials to combat this resistance. While curcumin, a polyphenolic compound from Curcuma longa, has previously demonstrated in vivo efficacy in mediating malaria infections through glycogen synthase kinase-3 beta (GSK-3β) inhibition, the potential was suboptimal due to its bioavailability constraints. Building on this, novel sugar-c

INTRODUCTION: Recent evidence indicates that an increasing number of endemic countries have deployed and are using genomic surveillance to determine and monitor the trends and patterns of malaria transmission. This study aimed to evaluate and identify the most informative genetic metrics for establishing and monitoring the genetic diversity of Plasmodium falciparum and its correlation with malaria transmission intensities in Mainland Tanzania.
METHODS: A cross-sectional survey of symptomatic patients was conducted in 100 health facilities from February to July 2021 and covered 10 regions cate

Members of the myosin superfamily are found in all eukaryotes, including Plasmodium falciparum, the parasite that causes the majority of severe malaria. The P. falciparum genome encodes six myosins, but apart from PfMyoA and B, these remain largely uncharacterised. Here, we characterise PfMyoF, a class XXII myosin, using structural prediction, biochemical assays, and imaging. We show that PfMyoF is a plus-end-directed, processive motor with a long neck region with capacity to bind up to six copies of the calmodulin homologue PfCaM. The PfMyoF tail contains predicted WD40 and Rab-like domains t

Malaria, caused by Plasmodium spp. parasites, presents treatment challenges due to the emergence of resistant strains to frontline antimalarials. In this study, we explored activities of 24 furoxan derivatives, originally designed for leishmaniasis treatment, against intraerythrocytic Plasmodium falciparum parasites. The studies were carried out with genetically-modified P. falciparum expressing GCaMP3, a cytosolic calcium indicator, and maintained in human red blood cell culture. The most potent compound Furoxan (1,2,5-oxidiazole-2N-oxide) 4m exhibited significant antimalarial activity (EC50

Malaria control remains constrained by an incomplete understanding of the molecular interactions that enable Plasmodium falciparum transmission within mosquito vectors. Although blood-stage parasite biology has been extensively studied, the mosquito-stage processes required for transmission are less well defined. Here, we characterize a previously unannotated Anopheles gambiae midgut factor, Midgut Ookinete Binding Protein 1 (AgMOBP1; AGAP008138), and define its role in parasite infection. AgMOBP1 is a membrane-associated protein localized predominantly to the apical surface of midgut epitheli

Analogues of tadalafil, an FDA-approved inhibitor of human phosphodiesterase 5, have been reported to block the growth of human Plasmodium falciparum in vitro. Herein, we synthesized and evaluated 56 tadalafil analogues prepared as pure diastereomers. Some of the analogues showed potent antiplasmodial activity at nanomolar concentrations with selectivity indices >20-250 in vitro. Compound 33 was the most potent analogue, with an IC50 of 80 nM against cultured parasites and an IC50 > 20 μM on HeLa cells, resulting in a selectivity index >250. Several compounds were tested for potential inhibiti

BACKGROUND: Currently, the widely used antigen-based malaria rapid diagnostic tests (RDTs) in sub-Saharan Africa primarily detect Plasmodium falciparum histidine-rich protein 2 (PfHRP2). However, their diagnostic accuracy is being compromised by the increasing report of evolving parasite strains with deletion of pfhrp2 and pfhrp3 genes in malaria endemic regions. Therefore, elucidating local heterogeneity and addressing methodological variability are critical for guiding diagnostic strategies.
