WEHI-601 is a statine-containing peptidomimetic that inhibits the malarial aspartyl protease, plasmepsin V. In this work, stereoselective fluorination is investigated as a strategy for enhancing the potency of WEHI-601 through conformational bias. Consistent with prediction, a fluorostatine-containing analogue of WEHI-601 is found to deliver stronger antimalarial activity than the lead compound, attributable to conformational preorganisation of the FCCO segment within the peptidomimetic backbone. However, the magnitude of the improvement is small, and this is discussed with reference to the

The efficacy of antimalarial treatment, particularly artemisinin-based combination therapy (ACT), continues to face persistent threats due to the development of drug resistance in Plasmodium falciparum. Although artemisinin derivatives remain effective, ACT regimen failure often results from decreased efficacy of the companion drug. Therefore, robust molecular surveillance focusing on genetic markers associated with companion drug resistance is essential to maintain global efficacy. The combination of dihydroartemisinin-piperaquine (DHA-PPQ) has long been used as first-line therapy and has pro

Cerebral malaria (CM) remains one of the most severe and life-threatening manifestations of Plasmodium falciparum infection, predominantly affecting children and pregnant women in Sub-Saharan Africa, where resource constraints compound diagnostic and therapeutic challenges. Despite significant advances in antimalarial therapy, CM continues to be associated with high mortality rates (15%-30%) and significant long-term neurological sequelae among survivors (10%-30%). Challenges in the diagnosis and management of CM in resource-limited African settings, include widespread misdiagnosis (up to 23%

Malaria remains one of the major threats to human health. Breakthrough drugs with high potency and low resistance risk are needed to combat the ever-increasing resistance to currently deployed antimalarials. Here, we explore a series of 4-amino-quinazoline-based sulfonamides, with drug-like physicochemical parameters and a synthetically accessible scaffold. Exemplars exhibit nanomolar potency against blood stage Plasmodium cultures, with up to 300-fold selectivity compared with a mammalian cell line. The compounds are also active against transmissible stages of P. falciparum and are refractory

Effective control of falciparum malaria depends on the sustained efficacy of frontline antimalarial drugs, particularly artemether-lumefantrine (AL), the most widely used therapy in Africa. However, the emergence of artemisinin partial resistance and reduced lumefantrine susceptibility in eastern Africa threaten malaria control and elimination. Robust genetic markers of decreased susceptibility to lumefantrine remain elusive, and our understanding of artemisinin resistance is incomplete. We report results of a Plasmodium falciparum genetic cross between a drug-sensitive line and a Ugandan stra

A highly efficient metal and catalyst-free approach for the preparation of synthetically challenged tri-OMe-aryl-substituted heterocyclic, di-/tri-OMe-aryl-based cyclic and acyclic 1,2,4,5-tetraoxanes (5a-l and 6a-k) is presented herein. Under green-chemistry conditions, the reactions occur in two key steps. The first one involves azeotropic activation of hydrogen peroxide for N-sulfonylpiperidone (2a-f), substituted cyclic and acyclic ketones (2a1-a4) oxidation, whereas the second one was H+[BF4]- (50-55% solun; 25 mol%)-catalyzed cyclization of the highly reactive gem-dihydroperoxide (3a-f a

Malaria continues to devastate humanity with significant global morbidity and mortality. The emerging resistance to antimalarials suggests artemisinin and its derivatives will encounter the same challenges as other antimalarial drugs. Strategies to accelerate developing new antimalarials include utilizing known scaffolds and making structural modifications to enhance properties and elucidate their mode of action. We have focused on acridine derivatives and describe 18 acridine/acridone analogs based on pyronaridine or quinacrine cores. Several molecules demonstrated potent in vitro activity ag

BACKGROUND/OBJECTIVES: The GMZ2.6c malaria vaccine candidate is a multi-stage P. falciparum chimeric protein that contains a fragment of the sexual stage Pfs48/45-6c protein genetically fused to GMZ2, which is an asexual stage vaccine construct consisting of conserved domains of Glutamate-Rich Protein (GLURP) and Merozoite Surface Protein-3 (MSP-3). Previous studies showed that GMZ2.6c is widely recognized by antibodies from individuals living in endemic areas of Brazil and that levels of anti-GMZ2.6c increase with malaria exposure and may contribute to immunity against the parasite. Insecticide-treated nets (ITNs), particularly long-lasting insecticide-treated nets (LLINs), are important for malaria control; however, the rise of insecticide resistance, behavioral adaptations in mosquito vectors, and diminishing net durability may affect their efficacy. The objective of this systematic literature review and meta-analysis to synthesize recent epidemiological evidence (2021-2025) on the correlation between ITN/LLIN use and Plasmodium falciparum infection prevalence and to explore sources of heterogeneity across populations, settings, and type of nets. Background/Objectives: Severe Plasmodium falciparum malaria remains a leading cause of childhood morbidity and mortality in sub-Saharan Africa. The clinical outcome is largely driven by the balance between pro-inflammatory and anti-inflammatory cytokines. However, immunological data from Sudanese children remain limited and the diagnostic utility of cytokine biomarkers has not been formally evaluated in this population. Therefore, this study aims to compare plasma levels of IFN-γ and IL-10 among Sudanese children with severe malaria (SM), uncomplicated malaria (UM) and healthy controls (HC) an

The malaria parasite Plasmodium falciparum depends entirely on de novo pyrimidine synthesis, as it is unable to salvage these essential nucleotides. This reliance makes the pyrimidine biosynthesis pathway a compelling target for antimalarial drugs, with several inhibitors targeting its rate-limiting enzyme, dihydroorotate dehydrogenase (PfDHODH), already in clinical development. In this study, we investigated the roles of three other pathway enzymes: aspartate transcarbamoylase (PfATC), carbamoyl phosphate synthetase II (PfCPSII), and dihydroorotase (PfDHO). PfATC features a unique N-terminal

BACKGROUND: The Ethiopian highlands >2000 m above sea level are classified as experiencing very low malaria transmission, but clinical case numbers have increased in recent years. Transmission intensity, and how many infections are imported from the lowlands, are not known.
METHODS: Surveys on subclinical prevalence were conducted in 8 sites across an altitude range of <1900 to 2900 m. Samples were screened by quantitative polymerase chain reaction (qPCR) and microscopy or rapid diagnostic testing (RDT). Data on recent travel were collected from subclinical and clinical individuals.

Malaria in pregnancy is associated with substantial maternal and fetal morbidity, yet pregnancy-specific guidance remains limited, particularly in non-endemic settings where low clinical suspicion, non-specific symptoms, and testing barriers may delay diagnosis. This report presents a case of severe third-trimester Plasmodium falciparum malaria and reviews management considerations. A nulliparous patient at 36 weeks and 4 days of gestation presented to a U.S. community hospital with fever, headache, and abdominal discomfort, after recent travel to Ghana. Initial evaluation showed maternal feve

BACKGROUND: The description of genetic diversity of malaria parasite populations in malaria-endemic areas may help determine the effectiveness of malaria interventions in the region. This study aimed to evaluate the genetic diversity and allele frequencies of the merozoite surface protein 1 gene (Msp1) and merozoite surface protein 2 gene (Msp2) in Plasmodium falciparum populations collected from Paletwa Township in western Myanmar and two regions along the China-Myanmar border (the Myitsone and Bhamo regions of Myanmar).
METHODS: Between 2013 and 2022, malaria cases in China were predominant

In response to the challenge of drug-resistant malaria, this study investigated the antimalarial potential of B. sapida ethanolic leaf extract using an in silico approach. Gas chromatography-mass spectrometry (GC-MS) analysis was performed to identify compounds present in the extract. Molecular docking and dynamics simulations were conducted to assess interactions and stability between selected lead compounds and the target protein PfDHODH using AutoDock Vina and GROMACS, respectively. A total of twenty-one compounds were identified and the three most abundant were D-Fructose 3-O-methyl- (21.0

Antimalarial chemotherapeutics, including artemisinin derivatives and their combination regimens, sustain clinical effectiveness against Plasmodium species, including resistant strains. However, these chemotherapeutics face challenges, including poor aqueous solubility and membrane permeability, a short elimination half-life, erratic oral bioavailability, and delayed parasite clearance. These formidable challenges have been widely addressed by developing new chemical entities, combining artemisinin-based combination therapies, exploring alternative routes of administration, or prolonging the d

Plasmodium falciparum malaria parasites harbor an essential plastid organelle, called the apicoplast, which produces key metabolites required for organelle function and parasite viability. Apicoplast functions depend on iron and other metals, but the membrane transporters that mediate metal import into this organelle have been challenging to identify. Tetracycline antibiotics, including doxycycline, specifically target the apicoplast and can exhibit metal-dependent activity. Using tetracycline-affinity proteomics, we identified a doxycycline-interacting, uncharacterized transmembrane protein (
