BACKGROUND: Malaria is a leading cause of under-5 mortality in sub-Saharan Africa. In Somalia, Plasmodium falciparum predominates and transmission shows spatial and seasonal variation. This study analyzed subnational inequalities in malaria incidence and mortality among children aged 0-4 years to inform targeted control strategies.
METHODS: This retrospective subnational analysis used modeled annual estimates of Plasmodium falciparum incidence, Plasmodium vivax incidence, and malaria mortality among children aged 0-4 years across Somalia's 18 regions (2010-2019), sourced from the Institute fo

Sexual maturation and fertility of the malaria causing parasite, Plasmodium spp., is critical to transmission between hosts. This developmental process involves an intricate gene expression regulatory network that drives gametocyte commitment, development, and the production of male and female gametes. Here, we characterized the role of the CCCH-type zinc-finger protein Plasmodium falciparum Male Development 3 ( Pf MD3) in the formation of gametes. Genetic disruption of Pf MD3 revealed a male fertility defect that decreased infectivity to mosquitoes. Molecular characterization of Pf MD3 reveal

Gene expression analysis in malaria parasites has been used to define transcriptional regulatory networks but has been used less frequently to characterize parasite response to drug treatment or to show how parasites may evade killing. Here, we applied single-cell RNA sequencing (scRNA-seq) to hundreds of thousands of individually infected asynchronous red blood cells to evaluate the parasite's response to treatment with three chemotypes that can be used for treatment (artemisinin) or prophylaxis and treatment (atovaquone, ganaplacide). We found that each treatment gave rise to different cell

A 12-month longitudinal entomological study (September 2023-August 2024) in the Western Highlands of Cameroon (Penka-Michel) investigated malaria transmission dynamics and insecticide resistance markers, including knockdown resistance mutations (L1014F, N1575Y) and metabolic resistance genes (G119S-Ace1, E205D-CYP6P3, L119F-GSTe2, CYP6P9a/b). Malaria transmission has depended on the coexistence of three species: Anopheles funestus (43.7%), Anopheles gambiae (42.9%), and Anopheles ziemanni (13.5%). The site was hyperendemic with annual EIR of 839.5 infective-bites/human/night. Plasmodium falcip

In low-resource clinical settings, Plasmodium falciparum infection and hypoglycemia frequently co-present yet require discrete diagnostic platforms, creating a practical barrier to timely patient management. This study describes a theoretically designed multilayer surface plasmon resonance (SPR) biosensor for label-free, simultaneous quantification of blood glucose and Plasmodium falciparum histidine-rich protein II (HRP-II) from a single analyte sample. The sensor stack comprises Ag/LiB3O5/BiB3O6/Cu/FASnI3/black phosphorus layers deposited on a BK7 glass prism in the Kretschmann attenuated tot

Despite the urgent need for new therapies to treat multidrug-resistant virulent Plasmodium falciparum, many promising antimalarial candidates fail to progress to clinical use because their pharmacokinetic (PK) profile is poorly characterized. This study predicted the PK profile of a new compound, 3-chloro-4-(4-chlorophenoxy)aniline (ANI), in silico using the pkCSM online platform. In vitro cytotoxicity in Vero E6 cells and antiplasmodial activity against chloroquine-sensitive (P. falciparum 3D7) and chloroquine-resistant (P. falciparum W2) strains were screened using the MTT colorimetric and t

The pathogenesis of Plasmodium falciparum malaria involves coordinated molecular events, including host cell invasion, intraerythrocytic replication cycles, and antigenic variation, which are critically dependent on the stage-specific regulation of parasite proteins. Despite their biological significance, the molecular governance of these pathogenic mechanisms remains inadequately characterized. Our investigation reveals that the expression dynamics of an ApiAP2 family transcription factor, PfAP2-V (PF3D7_1239200), exhibit strong temporal coordination with the transcriptional activation of vir

Severe Plasmodium falciparum malaria typically presents with fever, chills, and features of multiple organ dysfunction; however, gastrointestinal manifestations may occur early and can be mistaken for gastrointestinal infection. We report the case of a 29-year-old Ghanaian man working in South Sudan who initially presented with nausea, vomiting, and diarrhea. He had not taken malaria chemoprophylaxis and reported inconsistent use of mosquito-prevention measures. He was first diagnosed with acute gastrointestinal infection and treated accordingly. After 24 hours, he developed high-grade fever,

BACKGROUND: IgG-mediated effector functions are modulated by the antibody Fc-region glycan composition, and the absence of core fucose is a strong modifier of NK cell-mediated antibody-dependent cellular cytotoxicity (ADCC). Most plasma IgG is fully Fc-fucosylated, but antigens displayed on host-derived membranes can selectively induce Fc-afucosylated IgG. These antigens include alloantigens, enveloped virus surface proteins, and PfEMP1, a malaria parasite protein expressed on the surface of Plasmodium falciparum-infected erythrocytes (IEs).
METHODS: We determined levels and Fc-fucosylation o

Malaria caused by Plasmodium falciparum remains a major global health challenge, exacerbated by the emergence of resistance to artemisinin-based combination therapies. Dihydroorotate dehydrogenase (PfDHODH), an essential enzyme in parasite pyrimidine biosynthesis, represents a validated and selective antimalarial drug target. In this study, an integrated multilevel computational strategy was employed to identify novel PfDHODH inhibitors from a structurally diverse chemical library. Structure-based virtual screening yielded 1,500 initial hits, from which three top-ranking compounds (17507474, 2

BACKGROUND: The increasing insecticide resistance of malaria vectors is an urgent concern for disease control and novel vector control strategies are needed. Wolbachia are endosymbiotic bacteria that can invade mosquito populations and reduce transmission of human pathogens. Wolbachia strains in wild Anopheles (An.) malaria vectors are rare, with only two known genuine symbioses; An. moucheti with wAnM and An. demeilloni with wAnD. In this study, we set out to determine if there was a correlation between wAnD in different An. demeilloni mosquito body parts, infective stage Plasmodium (Pl.) fal

BACKGROUND: Malaria is one of the most devastating infectious diseases in humans, and antimalarial drugs have been used to combat it with minimal success. Worldwide, malaria treatment is threatened by the emergence and spread of artemisinin resistance, which is associated with mutations in the PfK13 propeller domain. In Sub-saharan Africa, data relating to the prevalence of Plasmodium falciparum malaria infection in association with the Kelch 13 mutations are mainly from research settings outside disease-endemic areas. This study is aimed at establishing the prevalence of P. falciparum malaria

Malaria, caused by the parasite Plasmodium falciparum, remains a significant global health threat, with multidrug resistance posing a major challenge to treatment. The P-glycoprotein homolog P. falciparum Multidrug Resistance Protein 1 (PfMDR1) is a key determinant of resistance to first-line antimalarials like mefloquine (MFQ) and chloroquine. ACT-451840, a clinical phase I drug, has been developed as an antimalarial candidate, but its mechanism of action and interaction with drug resistance markers remain to be fully understood. Here, we present the cryo-electron microscopy structure of PfMD

BACKGROUND: Malaria, caused by Plasmodium spp., remains a major global health threat. Among them, Plasmodium falciparum is the most pathogenic, and its asexual intraerythrocytic proliferation is the pathological basis. This process has enormous biosynthetic demands and highly relies on the coordinated function of the endomembrane and vesicular transport systems. The transition from asexual proliferation to sexual differentiation similarly involves remodeling of internal membrane complexes, membrane reshaping, and precise protein sorting. In eukaryotic cells, the Endosomal Sorting Complexes Req

Mosquito-borne diseases are responsible for over 600,000 deaths annually, mainly in sub-Saharan countries. Mosquito surveillance is a crucial element of vector control programmes to assure they remain effective. This study focused on optimizing the use of the MinION sequencer for interrogating mosquito pools from surveillance programmes, with simplified bioinformatic workflows for use in African laboratories nearer to the field. Mosquito pools were created using different human disease vector species, mimicking possible field trap contents. Some pools were spiked with DNA from the malaria para

BACKGROUND: The growing number of international travellers aged ≥60 years reflects global demographic changes and improved life expectancy. However, this group faces specific health risks due to age-related physiological changes and comorbidities, which may influence the presentation and management of travel-related illnesses. This study aimed to describe the epidemiological and clinical characteristics of imported diseases amongst older travellers compared with younger adults in Spain.
METHODS: A retrospective observational study was conducted using data from the Spanish National Network (+R

Quinolines are well-established scaffolds in drug discovery and form the core of many FDA-approved medicines. Among their reduced analogues, hexahydroquinolines (HHQs) have gained increasing attention due to their structural adaptability and broad pharmacological relevance. Research has largely focused on the 1,4,5,6,7,8-hexahydroquinoline framework, although interest in other variants, including 2-oxo derivatives, is steadily growing. This review summarizes recent progress in HHQ chemistry, with emphasis on synthetic strategies and emerging therapeutic applications. Traditional multicomponent

Plasmodium falciparum is responsible for what appears to be a never-ending public health issue in the developing world. With repeated infections, a gradual semi-immunity to severe malaria can be acquired, but this is disrupted when women become pregnant as the parasite cytoadheres in the placenta to prevent splenic clearance. This change in tissue tropism is due to specific transcription of the antigenically variable adhesin VAR2CSA. To better understand the molecular mechanisms activating var2csa and antigenic variation overall, we used a combination of phenotypic and systems biology assays.

Splenic abscess is a rare but life-threatening condition with poorly understood links to malaria and hemoglobinopathies. We present the case of a young male from West Africa who developed a pyogenic splenic abscess with concurrent Plasmodium falciparum malaria and sickle cell trait. Clinical data were collected through retrospective chart review, and a literature review was conducted focusing on splenic abscess in the context of malaria and hemoglobinopathies. The patient presented with abdominal pain and fever after initiating antimalarial treatment. Imaging revealed a large splenic abscess.
