A single archaeon couples long-chain fatty acid oxidation to methanogenesis.

Anaerobic degradation of complex organic compounds to methane and carbon dioxide is common in the deep subsurface biosphere and has long been considered to require syntrophic cooperation, with methanogens confined to the terminal use of simple C1 and C2 substrates. Thermodynamic considerations, multi-omics analyses and microcosm experiments suggest that archaeal lineages are capable of methanogenic degradation of complex substrates1-6. However, experimental validation in pure culture was lacking. Here we report the isolation of a thermophilic archaeon, Methanoglobus dolicholipophagus AA-75, af

Five-country analysis of geographic and dietary drivers of gut microbiome composition and genomic variation.

Human diet and lifestyle vary widely across cultures, shaping global diversity in gut microbiome composition and function. However, comparable cross-population studies have been limited by methodological heterogeneity. The Human Diets and Microbiome Initiative (THDMI) integrates standardized recruitment, dietary assessment, and shotgun metagenomics across 1,976 participants from the United States, United Kingdom, Spain, Mexico, and Japan to disentangle biological from technical variation. We observed pronounced geographic differences in nutrient intake, food consumption, and microbial composit

Scientific Remix: Epistemic Idioms and the Spread of Data Science.

Scholarship on science has long contained a tension between accounts emphasizing convergent forces and those highlighting divergent practices. This article addresses that tension through an ethnographic study of data science. It advances the concept of epistemic idiom-a recognizable and portable pattern of practices that evolve alongside knowledge-making tools, analogous to musical idioms arising from the interaction of material affordances and learned techniques. Through analysis of a project attempting to sequence chromosomal centromeres, I show participants enacting the nomad idiom of data

PhyDiff: Physics-guided latent diffusion model for sparse-view CT projection interpolation.

Sparse-view Computed Tomography (SVCT) is an important strategy for minimizing radiation exposure and accelerating industrial inspection, yet it suffers from severe streak artifacts under angular undersampling. Existing sinogram completion methods often process detector rows independently, failing to exploit cross-row correlations to maintain the structural consistency required for high-fidelity reconstruction. To address this, we propose PhyDiff, a physics-guided latent diffusion framework that reformulates sparse-view projection completion as an angle-conditioned sequence interpolation. PhyD

Migrasome-driven mitocytosis relieves mitochondrial damage induced by chikungunya virus TF and 6K.

Chikungunya virus (CHIKV) infection induces the formation of migrasomes, yet their specific role in CHIKV pathogenesis remains unclear. This study explores the mechanisms underlying mitochondrial damage induced by CHIKV 181 clone 25 (CHIKV 181/25) and the role of migrasomes in mitigating this damage. Using cultured cell lines, we assessed the impact of CHIKV infection on mitochondrial integrity and function, with particular emphasis on the viroporin proteins transframe (TF) and 6K. We utilized fluorescence microscopy and transmission electron microscopy to visualize the interplay between migra

Public trust and moral perceptions as predictors of anticipated preventive behavior toward future epidemics among adults in Riyadh, Saudi Arabia: a cross-sectional study.

BACKGROUND: Future epidemic preparedness depends partly on public willingness to follow preventive measures. This study examined whether public trust and moral perceptions were associated with anticipated preventive behavior toward future epidemics among adults in Riyadh, Saudi Arabia. METHODS: A cross-sectional survey included 256 adults. Three 10-item constructs measured public trust, moral perceptions, and anticipated behavior using five-point Likert scales. Internal consistency, correlations, subgroup comparisons, multiple regression, and a fully adjusted general linear model were evaluat

Biomaterial-Based Activation Enables Scalable Expansion of Stable Human Regulatory T-cells.

Regulatory T-cells (Tregs) hold significant promise for the treatment of autoimmune diseases and transplant rejection, yet their clinical translation is limited by inefficient and inconsistent manufacturing strategies that fail to generate stable and functional cell populations at scale. Current ex vivo expansion approaches rely on bead-based activation systems that often induce transient FOXP3 expression and variable product quality. Here, we present a bioinspired hydrogel-based activation platform that enables robust expansion and stabilization of regulatory T-cell phenotypes from primary hu

ICE-CAP: ice crystal engineering for cryogel architecture programming. Modulation of material properties with ice-binding polymers.

Macroporous cryogels are formed by polymerising monomers around ice crystal templates, with water as the only waste product, making them appealing as tuneable and sustainable scaffolds. Cryogels have found diverse applications spanning pollutant removal, vaccine delivery, tissue culture and purification. However, an unmet challenge remains: ice crystal templates are intrinsically dynamic and evolve during polymerisation, leading to heterogeneity and complicating reproducibility. Here we overcome this challenge using ice-binding protein-inspired polymers that pre-program the ice crystal templat

Nuclease-Free Scalable Purification of Physalis Mottle Virus-Like Particles Expressed in Escherichia coli.

Plant virus-like particles (VLPs) are promising nanotechnology platforms, but their translation requires purification strategies suitable for large-scale production. Here, we developed a nuclease-free downstream process for Physalis mottle virus (PhMV) VLPs expressed in Escherichia coli, combining ammonium sulfate precipitation, low-pH extraction, ultrafiltration/diafiltration (UF/DF), and flow-through anion-exchange (AEX) chromatography. Ammonium sulfate precipitation separated the VLPs from bulk host nucleic acids, whereas extraction at low pH removed most host cell proteins (HCPs) while ret

A Hybrid Mechanistic Model Capturing Lactate Run-Away and Back-Metabolism in Mammalian Cell Culture.

Mechanistic modeling is important in biopharmaceutical manufacturing by enabling in-silico experimentation. However, developing a robust mechanistic model for mammalian cell culture remains challenging due to the complexity of the underlying bioprocesses. Existing data-driven models often suffer from limited extrapolation capability and require extensive datasets. Not all modeling approaches can accurately represent differing metabolic scenarios such as lactate consumption and production. To address these limitations, this study proposes a hybrid mechanistic model for mammalian cell culture to

On-Surface Synthesis of Hexabenzocoronenes via a Retro-Büchner Reaction of Heptaarylcycloheptatrienes.

On-surface synthesis has become a powerful technique for obtaining various nanostructures, mainly by linking small precursor molecules. In contrast, the cleavage of specific moieties from precursor molecules is also an important and promising strategy to increase the variety of products. Here, we report the on-surface synthesis of hexabenzocoronene (HBC) and its hexa-fluorinated analogue (6F-HBC) from the parent heptaphenylcycloheptatrienes, via the systematic extrusion of a phenyl-CH: carbenic fragment and a subsequent cyclodehydrogenation process on a Au(111) substrate. Their structures and

In vitro Transdifferentiation Approaches for Dissecting Vascular Cell Identity in Arabidopsis.

Plant vascular tissues consist of multiple cell types that together coordinate long-distance transport, signaling, and perform diverse metabolic functions. While extensive research uncovered key regulators of vascular patterning and function, the dissection of individual vascular cell types has been hindered by their inaccessibility in planta and the interdependence among the cell types. To overcome these limitations, in vitro transdifferentiation-based approaches have emerged as powerful experimental tools. Here, we provide a comprehensive review of current transdifferentiation strategies use

Queer Difference in Ghana's Cultural Publics: Gender, Sexuality, and Historical Memory.

This paper examines how sex and gender are distinguished or conflated, how gender nonconformity is recognized and negotiated, and what oral histories and spiritual narratives reveal about locally situated understandings of gender and sexual difference in Ghana. We draw on African feminist, queer, and intersectional approaches and use ethnographic interviews, informal group discussions, oral histories, archival materials, and cultural artifacts collected between August and December 2023. Findings show that local languages and social practices distinguish bodily sex, gendered expectations, gende
