Interferon gamma-induced MX1 and IFITM1 inhibit Varicella-Zoster Virus Replication.

UNLABELLED: Interferon gamma (IFN-γ) is a potent cytokine secreted following primary varicella-zoster virus (VZV) infection. Our previous studies demonstrated that pretreatment with IFN-γ completely inhibited VZV replication in lung fibroblast MRC-5 and retinal epithelial ARPE-19 but not in melanoma MeWo cells, suggesting that IFN-γ-stimulated protein(s) inhibit viral replication. Our microarray analysis revealed that 34 interferon-stimulated genes (ISGs) were upregulated by greater than 2.5-fold at 8 h post-treatment in both ARPE-19 and MRC-5 cells compared to those of MeWo cells. The depleti

Two Truncating MITF Variants Expand the Phenotypic Spectrum of COMMAD Syndrome.

Pathogenic variants in MITF (Microphthalmia Transcription Factor) gene are associated with a spectrum of disorders, including Waardenburg syndrome type II and Tietz syndrome. Rare reports suggest that biallelic loss-of-function variants may result in a severe multisystem phenotype. We report an infant presenting with a phenotype consistent with COMMAD-like syndrome (Coloboma, Οsteopetrosis, Μicrophthalmia, Μacrocephaly, Αlbinism, and Deafness) who died in the early neonatal period. In addition to the core COMMAD features, the proband exhibited ectopic ossification, cardiac fibrosis and fibroel

Gastrin and cholecystokinin 2 receptor, and gastrin-releasing peptide receptor in cancer: From pathogenesis to precision medicine.

Gastrin regulates gastric acid secretion and mucosal homeostasis via the cholecystokinin-2 receptor (CCK2R). Aberrant signaling via CCK2R (cholecystokinin family) and the distinct class A GPCR, gastrin-releasing peptide receptor (GRPR; bombesin family), drives oncogenesis across diverse GI and non-GI malignancies through calcium mobilization, MAPK/PI3K activation, EGFR transactivation, and constitutive splice variants like CCK2Ri4sv. Furthermore, precursor forms such as progastrin remodel the tumor microenvironment by interacting with non-CCK binding sites (p78, Annexin A2), converting fibrobl

Ectopic expression of MUC5B in the respiratory bronchiole initiates endoplasmic reticulum stress in the IPF lung.

RATIONALE: IPF is an irreversible and progressive type of lung fibrosis that is pathologically characterized as spatially heterogeneous. Despite the identified dominant risk factor for IPF as the gain-of-function MUC5B promoter variant, little is understood for how MUC5B drives lung fibrosis. OBJECTIVES: We used spatial transcriptomics from idiopathic pulmonary fibrosis (IPF) and unaffected control lung tissue to further understand the pathogenesis of MUC5B-driven lung fibrosis. METHODS: We captured 43 fields of view in 15 IPF and 13 controls with and without the MUC5B promoter variant using

Atypical collective oscillatory activity in cardiac tissue uncovered by optogenetics.

Many biological processes emerge as frequency-dependent responses to trains of external stimuli. Heart rhythm disturbances, that is cardiac arrhythmias, are important examples as they are often triggered by specific patterns of preceding stimuli. In this study, we investigated how ectopic arrhythmias can be induced by external stimuli in cardiac tissue containing a localised area of depolarisation. Using optogenetic in vitro experiments and in silico modelling, we systematically explored the dynamics of these arrhythmias, which are characterised by local oscillatory activity, by gradually alte

Extracardiac imaging biomarkers of cardiovascular health: a comprehensive review.

Cardiovascular disease (CVD) is the leading global cause of morbidity and mortality. Beyond clinical risk factors and cardiac imaging, extracardiac imaging biomarkers provide complementary insight into cardiometabolic health. Routine noncardiac imaging often depicts the liver, kidneys, pancreas, lung, brain, adipose tissue, muscle, vasculature, breast, and bone, enabling opportunistic risk assessment. Nonalcoholic fatty liver disease (NAFLD), detectable on MRI, CT, and ultrasound, independently predicts major adverse cardiovascular events (MACE). Chronic kidney disease-related markers includin

Spatial transcriptomics reveals altered communities and drivers of aberrant epithelia and pro-fibrotic fibroblasts in interstitial lung diseases.

Interstitial lung diseases (ILD) are characterized by fibrotic scarring of the lung parenchyma with remarkably unfavorable prognosis. Using single-nucleus RNA sequencing and spatial transcriptomics, we generated a comprehensive cellular network of the distal lung and its alterations in fibrosis. Integration with histopathology revealed that the transformation of normal parenchyma into fibrotic tissue is accompanied by ectopic bronchiolization and decellularization. Areas of active fibrosis were characterized by co-localization of pro-fibrotic CTHRC1-hi fibroblasts and aberrant transitional epi

[A rare differential diagnosis for pulmonary calcifications].

A 77-year-old patient developed community-acquired pneumonia four times within two years. During the work-up, the CT chest showed typical signs of dendriform pulmonary ossification (DPO), a rare disease characterized by bone formation in the lung tissue. DPO usually occurs in chronic lung damage, its clinical significance is still unclear, but it could become more relevant with increasing high-resolution diagnostics.

Metabolic Checkpoints and Lymphoid Neogenesis in Lung Dendritic Cells: Mechanisms Guiding Tolerance and Chronic Lung Inflammation.

Dendritic cells (DCs) are key sentinels in the lung mucosa that interpret environmental signals to either promote tolerance or trigger inflammation, influencing the development of chronic lung diseases. This review highlights recent mechanistic insights showing that metabolic checkpoints serve as upstream regulators of DC fate and activity: inflammatory stimuli activate HIF-1α/mTOR-linked glycolytic pathways that drive maturation, cytokine secretion, antigen presentation, and migration. In contrast, AMPK-related oxidative and lipid metabolism pathways support tolerogenic states that encourage

Metabolic Dysfunction-Associated Steatotic Liver Disease and Respiratory Disorders: A Systematic Review of Clinical and Pathophysiological Associations.

PURPOSE OF REVIEW: Metabolic dysfunction–associated steatotic liver disease (MASLD) represents the hepatic manifestation of ectopic fat accumulation and adipose tissue dysfunction. Excess visceral adiposity promotes adipose tissue hypoxia, macrophage infiltration, and chronic low-grade systemic inflammation, generating systemic metabolic stress that may extend to the lung through a proposed liver–lung axis (hepatopulmonary crosstalk). This systematic review synthesizes current evidence on the associations between MASLD and major respiratory diseases, with emphasis on shared obesity-driven mech

Semaglutide Reverses Ectopic Lipid Accumulation, Impaired Myocardial Perfusion Reserve, and Diastolic Dysfunction in a Mouse Model of Cardiometabolic Heart Disease.

Semaglutide (SEMA) improves cardiometabolic outcomes in obesity, but its mechanisms remain incompletely understood. We examined the effects of SEMA in mice fed a high-fat, high-sucrose diet. Obese mice were treated with SEMA and compared with pair-fed controls to account for reduced dietary intake with SEMA. Multiparametric cardiovascular magnetic resonance was used to assess epicardial adipose tissue volume and composition, myocardial fat fraction, adenosine myocardial perfusion reserve, systolic strain, and diastolic function, with histological evaluation of myocardial fibrosis. Safety of intravascular administration of umbilical-cord-derived mesenchymal stromal cells: an updated systematic review and meta-analysis.

BACKGROUND: Mesenchymal stromal cells (MSCs) have the capacity to self-renew and exert immunomodulatory and paracrine effects. Our previously published systematic review of 55 randomized controlled trials (RCTs) of all MSC sources found an overall favorable safety profile for MSCs aside from more frequent fever in the MSC group. OBJECTIVE: Umbilical cord MSCs (UC-MSCs) possess potential biological and economic advantages over other MSC sources. Since several UC-MSC RCTs have been recently published in a heterogenous group of disorders, we performed a systematic review to specifically examine

FSTL4 interaction with Gαi proteins activates the Akt-mTOR pathway to drive malignant phenotypes in non-small cell lung cancer.

Non-small cell lung cancer (NSCLC) presents a critical challenge clinically; thus, identifying novel drivers and reliable prognostic biomarkers is imperative. Here, we investigated the clinical relevance, cell-type-specific localization, and mechanistic role of Follistatin-like 4 (FSTL4) in NSCLC. Analysis of The Cancer Genome Atlas (TCGA) and local resected tissue samples show that FSTL4 expression is significantly elevated in NSCLC tissues compared to normal lung tissues, and serves as an independent prognostic indicator for reduced overall survival. Single-cell RNA sequencing confirmed that

SATB2 dysregulation generates a novel circular RNA and drives KRAS-like transcriptional reprogramming and transformation-associated phenotypes.

BACKGROUND: Even though epigenetic factors contribute to oncogenesis, most human cancer models still assume that disease originates from driver DNA mutations. Thus, it is still unclear if non-genetic mechanisms are sufficient to trigger malignant transformation. Special AT-rich binding protein 2 (SATB2) is a chromatin organizer that brings distal DNA elements into close proximity, thus remodeling chromatin structures to reprogram cell-specific and/or developmentally sensitive gene networks. METHODS: Here, we chronically exposed human bronchial lung epithelial cells to ≤ 2 µM inorganic arsenic

DDIT3, OTUB2, and ASS1 regulate arginine biosynthesis in colorectal cancer cells under arginine deficiency.

Argininosuccinate synthetase 1 (ASS1) is a rate-limiting enzyme in arginine biosynthesis, and its stability is regulated by TRAF2-mediated ubiquitination. Here, we report OTUB2 as a major deubiquitinase to stabilize ASS1, resulting increased arginine biosynthesis in colorectal cancer (CRC) cells; OTUB2 expression is elevated in CRC tissue, and patients with high OTUB2 expression exhibit shorter overall survival. As such, ectopic expression of OTUB2 promotes growth of CRC cells and xenograted tumors and accelerates cell migration and lung metastasis. Moreover, arginine deprivation induces marke

SRGAP2-AS1 Regulates RSV Replication and EMT-Associated Changes Through PTBP1 and ZEB1.

Respiratory syncytial virus (RSV) is a leading cause of lower respiratory tract disease, but the contribution of long non-coding RNAs (lncRNAs) to RSV-induced epithelial-mesenchymal transition (EMT)-associated changes remains unclear. We used transcriptomic screening in RSV-exposed BEAS-2B cells to identify candidate lncRNAs and then examined SRGAP2-AS1 in cell and mouse models. Expression was measured by RT-qPCR, and loss- and gain-of-function experiments were combined with RNA immunoprecipitation, co-immunoprecipitation, Western blot analysis, and TCID50 assays. SRGAP2-AS1 was increased in R

Fatty Pancreas and Risk of Type 2 Diabetes, Chronic Kidney Disease and Cardiovascular Events: Evidence From a Population-Based Cohort.

BACKGROUND: Fatty pancreas is a metabolically active ectopic fat depot, but its cardiometabolic implications have been assessed using heterogeneous thresholds. We investigated the association of fatty pancreas, quantified using MRI-derived proton density fat fraction (PDFF) and categorised according to 2026 international consensus thresholds, with prevalent and incident type 2 diabetes (T2D), chronic kidney disease (CKD) and major adverse cardiovascular events (MACE). METHODS: We analysed 19,255 European-ancestry participants from the UK Biobank imaging sub-study. Pancreatic PDFF was categori

Microstructural disease and hypoperfusion in dilated cardiomyopathy underpin midwall septal fibrosis.

BACKGROUND: The presence of midwall septal fibrosis (MSF) in dilated cardiomyopathy (DCM) has been shown to be associated with adverse clinical outcomes, but the underlying pathophysiological mechanisms are incompletely understood. We investigated whether MSF associates with a distinct pattern of myocardial microstructural and microvascular abnormalities using advanced cardiovascular magnetic resonance (CMR). METHODS: This was a prospective, multi-referral, single-center study comparing the hearts of patients with a current or prior diagnosis of DCM with and without MSF ("MSF+"/"MSF-"), to a

Ectopic breast cancer in vulva and other non-axillary sites: a systematic review.

BACKGROUND: Ectopic breast cancer (EBC) is a rare malignancy (0.3–0.6% of all breast cancers) that arises along the embryological milk line. While axillary EBC is better characterised and typically managed similarly to orthotopic breast cancer, the clinical features and management of non-axillary EBC remain poorly defined. AIM: To synthesise available data on the clinical presentation, histopathology, management, and outcomes of non-axillary EBC to refine treatment strategies. METHODS: A systematic review of case reports and case series was conducted following PRISMA 2020 guidelines and regist
