Prevalence and determinants of zoonotic malaria in the Greater Mekong Subregion (GMS) and Malaysia: systematic review and meta-analysis.

BACKGROUND: Zoonotic malaria is a growing public health concern in Southeast Asia, with Malaysia and Thailand accounting for 95.2% of the 3290 global cases reported in 2023. It is caused by Plasmodium species primarily adapted to long- and pig-tailed macaques, transmitted to humans via certain Anopheles mosquitoes. This meta-analysis quantifies its prevalence and risk factors in the Greater Mekong Subregion and Malaysia. METHODS: This meta-analysis was conducted following the PRISMA (Preferred Reporting Items for Systematic) guidelines. A comprehensive literature search was conducted in PubMe

Novel single-cell preservation and RNA sequencing technology unlocks field studies for Plasmodium natural infections.

Single-cell RNA sequencing (scRNA-seq) is a powerful technology used to investigate cellular heterogeneity. When applied to unicellular eukaryotes such as Plasmodium, scRNA-seq provides a single-cell resolution valuable to study complex infections comprised of mixed lifecycle stages and clones. Until now, the application of scRNA-seq has been mainly limited to in vitro and animal malaria models due to the challenges of working with Plasmodium natural infections in endemic settings. We validated sample preparation methods and a single-cell RNA preservation and sequencing technology in P. knowle

Comparative immunological study of Plasmodium knowlesi infections in humans and macaques: insights into cytokine dynamics.

BACKGROUND: Plasmodium knowlesi, a simian malaria parasite endemic to Southeast Asia, is transmitted from macaques to humans via mosquitoes and has seen a surge due to human encroachment into macaque habitats. While the primary host, Macaca fascicularis, can regulate P. knowlesi and alleviate disease symptoms, infected humans face a different scenario. A study was conducted in Sabah, Malaysia to compare the effects of parasite genomic DNA (gDNA) and host (both human and macaques) mitochondrial DNA (mtDNA) release on cytokine profiles in humans and macaques infected with P. knowlesi. METHODS:

Establishment of a visual mixed-dye loop mediated isothermal amplification assay for detecting Plasmodium knowlesi.

Colorimetric detection of loop-mediated isothermal amplification (LAMP) amplicons is one of the easiest and most direct methods among all detection techniques. However, not all indicators exhibit a noticeable colour change for both positive and negative reactions. In this study, we present a novel and visualized dual colour LAMP assay for the detection of Plasmodium knowlesi. The assay utilizes two types of dyes (phenol red and hydroxynaphthol blue) to facilitate a clear and distinguishable colour change upon successful amplification. The colour changes generated by the current single dye LAMP

Impact of environmental factors on the bionomics of Anopheles mosquito vectors of zoonotic malaria: A narrative review.

The increasing burden of non-human primates (NHP) malaria, driven primarily by Plasmodium knowlesi, poses a growing public health threat in many countries across Southeast Asia. Compounding this challenge, the emergence of other NHP Plasmodium species infecting humans, including P. cynomolgi, P. inui, and P. fieldi, introduces additional complexity to malaria elimination efforts, particularly in countries like Malaysia, Thailand, and Vietnam. A complex interplay among human populations, vector dynamics, and environmental factors influences the transmission and prevalence of this disease. This

Molecular and Microscopic Challenges in Detecting Plasmodium cynomolgi Co-Infections with Plasmodium vivax: A Case Report.

The risk of non-human primate (NHP) malaria transmission to humans is increasing, with Plasmodium knowlesi and Plasmodium cynomolgi emerging as significant zoonotic threats, particularly in Malaysia. While P. knowlesi is well-documented, P. cynomolgi infections in humans remain underreported, largely due to diagnostic challenges. Routine microscopy and standard molecular diagnostic tools often misdiagnose P. cynomolgi infections as P. vivax due to morphological similarities and genetic homology. We report a new case of a human P. cynomolgi infection misdiagnosed as Plasmodium vivax in a 32-yea

First molecular detection of zoonotic Plasmodium knowlesi, Plasmodium cynomolgi, and Plasmodium inui in Assamese macaques from northern Thailand.

The growing proximity between non-human primates (NHPs) and human communities in Thailand has raised concerns about zoonotic disease transmission. Among these NHPs, Assamese macaques (Macaca assamensis) are understudied in Thailand, particularly regarding their role as reservoirs for zoonotic malaria. The limited data on Plasmodium infections in this species highlights the need for further investigation. Therefore, this study determined the prevalence and molecular characterization of zoonotic Plasmodium spp. in Assamese macaques from Chiang Rai Province, northern Thailand. In total, 133 blood

Sialyl-T Antigen: A Novel Red Blood Cell Determinant for Plasmodium falciparum Invasion.

Malaria continues to pose significant health challenges globally despite advances in control measures. Plasmodium falciparum, the parasite responsible for most severe malaria cases, uses multiple redundant invasion pathways to enter the red blood cell (RBC) during the blood stage of infection. Through a combination of RNA interference screening in erythroid cells and validation by CRISPR/Cas9-mediated knockout in primary human hematopoietic stem cells, we identified the glycosyltransferase Core 1 Synthase Glycoprotein-N-Acetylgalactosamine 3-Beta-Galactosyltransferase 1 (C1GALT1) as a novel ho

High-sensitivity detection of asymptomatic malaria in high-risk Thai populations using droplet digital PCR.

BACKGROUND: Asymptomatic infections present a major obstacle to malaria elimination efforts, particularly within high-risk populations where low-density parasitemia often escapes detection by conventional diagnostic methods. In such cases, onward transmission remains possible, sustaining the parasite reservoir. Therefore, highly sensitive screening techniques are essential for effective malaria prevention and control. Molecular methods provide greater sensitivity and specificity in detecting low-level infections compared to traditional approaches. In this study, we evaluated droplet digital PCR

Insights into AI-Driven malaria diagnosis: A systematic review with implications for Plasmodium knowlesi.

Plasmodium knowlesi has emerged as a significant zoonotic malaria threat, particularly in Southeast Asia, where its incidence continues to rise. Timely and accurate diagnosis of its blood stages is critical for effective diagnosis and treatment, as disease severity and transmission dynamics vary across different stages. Microscopic examination is the gold standard for malaria diagnosis; however, it is labour-intensive and requires professional interpretation. This makes it prone to variability and possible misclassification, especially among morphologically identical Plasmodium species. Recent

Vernonia amygdalina (Delile) exhibits in vitro anti-plasmodial activities against Plasmodium knowlesi.

Malaria is a global public health concern, where Plasmodium knowlesi contributes to most human malaria cases in Malaysia. The leaves of Vernonia amygdalina (Delile) were found to exhibit significant anti-plasmodial properties and are commonly used in sub-Saharan Africa countries for malaria treatment. Nonetheless, studies are lacking in the effect of this plant against P. knowlesi specifically. This study aimed to investigate the in vitro anti-plasmodial activity of crude extracts from V. amygdalina (Delile) against P. knowlesi. The fresh leaves of V. amygdalina were sequentially extracted in

Babesiosis and Malaria in the United States: Epidemiology, Research Funding, Medical Progress, & Recommendations for Improvement.

BACKGROUND: Babesiosis and malaria are infectious diseases caused by the intraerythrocytic parasites Babesia and Plasmodium, respectively. While no human red blood cell (RBC) receptors have been shown to be essential for B. microti (Bm) invasion, Duffy (ACKR1) was reported to be essential for P. knowlesi and P. vivax invasion in 1975 and 1976, respectively. This suggests additional medical progress is needed for babesiosis, warranting a detailed analysis. METHODS: Given similarities in the target cell of infection, data about babesiosis and malaria cases in the US were obtained from the Cente

Evaluating the potential of cross-species neutralization of anti-PfCyRPA and anti-PfRIPR Monoclonal Antibodies.

PfCyRPA and PfRIPR are promising next-generation malaria blood-stage vaccine candidate antigens that play an essential role in erythrocyte invasion of Plasmodium falciparum. CyRPA and RIPR orthologs are present in all human-infecting Plasmodium species, suggesting the potential for a cross-species vaccine. Using Growth Inhibition Assays (GIA), this study investigates seven anti-PfCyRPA and three anti-PfRIPR monoclonal antibodies targeting P. falciparum for their inhibitory activity against P. knowlesi, a non-falciparum species that contributes to a significant burden of zoonotic disease in Sou

Effect of periostin (OSF-2) on phagocytosis of Plasmodium-infected erythrocytes.

INTRODUCTION: Phagocytosis is a pivotal component of the human innate immune defense against malaria. This essential defense mechanism is often modulated by various host-derived soluble factors. We investigated the phagocytosis of Plasmodium falciparum- and P. knowlesi-infected erythrocytes (IRBC) by human monocytic THP-1 cells in the presence of periostin (OSF-2), a human secretory protein involved in inflammation and tissue repair. This focus was prompted by the fact that OSF-2 is a potent stimulator of Plasmodium IRBC rosette formation, a parasite-derived cytoadherence phenomenon known to i

Acute Kidney Injury in Severe Malaria: A Serious Complication Driven by Hemolysis.

Acute kidney injury (AKI) is a common clinical complication in malaria, with AKI reported across all species that cause severe disease, including Plasmodium falciparum, Plasmodium knowlesi, and Plasmodium vivax. AKI during malaria varies based on host and parasite factors, including the growth potential of the parasite within host red blood cells, the extent of red blood cell lysis, and the capacity of the parasite to sequester within the microvasculature. In this review, we focus primarily on P. falciparum pathogenesis and the role of intravascular hemolysis in AKI through the depletion of en

Current update on malaria in pregnancy: a systematic review.

BACKGROUND: Malaria during pregnancy poses significant risks to both the mother and the developing fetus. For pregnant women, the infection can result in severe illness and even death. Parasite sequestration in the placenta can cause maternal anemia and increase the risk of mortality both during and after childbirth. Malaria is also a major contributor to stillbirths and preterm births. Infected placental tissue can impede fetal growth, resulting in low birth weight, which is linked to delayed growth and cognitive development in the child. Furthermore, malaria during pregnancy remains a major
