Altitudinal cross-sectional study of the sand fly fauna of the highlands of Tigray Region, northern Ethiopia.

BACKGROUND: Cutaneous leishmaniasis (CL), caused by Leishmania aethiopica and transmitted by Phlebotomus longipes, is endemic in northern Ethiopia. However, sand fly fauna is poorly documented in the northern Tigray region. High-altitude CL cases have also been observed, but their local acquisition is uncertain. To identify the faunal composition, their geographical and altitudinal distribution, cross-sectional surveys were conducted in ten villages (average altitude range: 2411 - 3186 meters) with confirmed CL cases across four endemic districts of the region. A total of 170 light trap-nights

Macroecological synchronization of vector-borne diseases under climate variability in Colombia.

BACKGROUND: Climate variability may synchronize vector-borne diseases across distinct transmission systems. This study evaluates climatic coherence among malaria, cutaneous leishmaniasis, and Chagas disease in Colombia and projects changes in environmental suitability under future climate scenarios. METHODS: National surveillance, climatic, environmental, and socioeconomic data from 1980 to 2025 were analyzed using generalized additive models, distributed lag nonlinear models, and wavelet coherence. Annual series were used for interannual analyses, whereas monthly epidemiological and climatic

Haemophagocytic Lymphohistiocytosis Triggered by Visceral Leishmaniasis Masquerading as Epstein-Barr Virus (EBV)-Associated Haemophagocytic Lymphohistiocytosis in Rheumatoid Arthritis: A Case Report and Literature Review.

Haemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory syndrome caused by uncontrolled immune activation. In adults, it is usually secondary to an underlying condition, such as infection, malignancy, or autoimmune disease. Identifying this trigger is essential, as treatment depends on the cause. Visceral leishmaniasis (VL) is a systemic parasitic infection that may mimic or trigger HLH, particularly in immunosuppressed patients. We report a 59-year-old man with seropositive rheumatoid arthritis, for which he received immunosuppressive treatment with methotrexate and

Antileishmanial potential of polar fractions from Amaranthus blitoides S. Watson, Caucalis platycarpos L., and Cistanche flava (C.A.Mey.) Korsh.: evidence from in vitro and in vivo studies.

BACKGROUND: Cutaneous leishmaniasis (CL) is recognized by the World Health Organization (WHO) as a research priority, yet its treatment remains challenging. CL lesions arise from complex interactions between the parasite and host immunity, where excessive inflammation can delay healing. Given the anti-inflammatory and immunomodulatory properties of medicinal plants, this study evaluated the effects of polar fractions from methanolic extracts of Amaranthus blitoides S. Watson, Caucalis platycarpos L., Cistanche flava (C.A.Mey.) Korsh., and their mixtures on CL caused by Leishmania major. METHO

The gene expression signature of multinucleated giant cells in the spleens of Leishmania donovani-infected mice.

Multinucleated giant cells (MGCs) are associated with chronic inflammation and infectious diseases, but their biological characteristics in vivo remain poorly understood because intact cells are difficult to isolate from tissues. In visceral leishmaniasis, heavily parasitized, hemophagocytic MGCs are observed in the spleens of Leishmania donovani-infected mice, suggesting that these cells may contribute to parasite persistence and disease pathology. In this study, we established a method for isolating intact MGCs from the spleens of chronically infected BALB/c mice and performed transcriptomic

Widespread transmission in diverse ecotypes challenges visceral leishmaniasis control in East Africa.

East Africa is emerging as the global hot spot of visceral leishmaniasis (VL), yet efforts to eliminate it are hindered by substantial knowledge gaps in its ecoepidemiology. Here, we report on the high prevalence of Leishmania infection in Phlebotomus orientalis in Marsabit county, Kenya (3.9%), and Gedaref state, Sudan (3.6%), where this species comprised 99.8% (n = 1185) and 100% (n = 1350) of captured Phlebotomus females, respectively. In Aba Roba, Ethiopia, Phlebotomus martini accounted for 94.4% of 184 collected Phlebotomus females and had a lower infection rate of 1.5%. Phlebotomus orien

Leishmania respiration profiling as a One Health functional marker associated with antimony resistance in human and canine leishmaniases.

Rapid functional tools for assessing antimony susceptibility in Leishmania are needed to strengthen resistance surveillance across human and veterinary settings. We developed a real-time oxygen-consumption assay using the Resipher analyzer and evaluated normalized oxygen-consumption rate (OCR) as a functional correlate of antimony (Sb) resistance in Leishmania infantum and Leishmania donovani. A density of 1 × 107 parasites/mL and a 48-h readout provided the most robust analytical window, and rotenone markedly reduced OCR, demonstrating assay responsiveness to respiratory perturbation. Synthesis, Antiparasitic Activity, and ADME Profile of Morpholinephenylbenzamides: A Pharmacological Study Against Chagas Disease and Visceral Leishmaniasis.

Chagas disease and leishmaniasis are neglected tropical diseases with limited treatment options. This study reports a hit-to-lead optimization of morpholinephenylbenzamides, starting from hit 1, which showed potent activity against Trypanosoma cruzi (IC50 0.29-0.76 μM) and Leishmania infantum (IC50 10.7 μM), despite a suboptimal ADME profile. A structure-activity relationship study of twenty-four novel derivatives provided key pharmacophoric insights. While Trypanosoma cruzi was less susceptible to chemical modifications, compound 20 exhibited potency comparable to benznidazole (IC50 3.58 μM)

Canine exposure to vector-borne pathogens in a rural Brazilian landscape.

Vector-borne pathogens (VBPs) of medical and veterinary concern have mostly been studied in urban dog populations, while animals in rural settings, with limited access to ectoparasite prevention, have received less attention. In this study, we assessed canine exposure to VBPs in a rural Brazilian landscape. From August to October 2024, we collected 155 blood samples from dogs domiciled in a rural area of the municipality of Garanhuns, Pernambuco, Northeastern Brazil. Samples were assessed using an immunofluorescence antibody test (IFAT) to detect antibodies against Trypanosoma cruzi and using

Skin Disease After Refugee Resettlement: A Practical 90-Day Approach for Dermatologists and Primary Care Physicians.

The first months after refugee resettlement create an opportunity to identify transmissible, disabling, and undertreated skin disease. In the United States, the domestic medical examination generally occurs 30-90 days after arrival, but the clinical principles are relevant across resettlement systems. We conducted a structured narrative review of peer-reviewed publications and organizational or public-health sources. Evidence varied by migration stage, region, age, and referral pathway; camp and specialty-clinic data should not be treated as post-resettlement prevalence estimates. Physicians s

First record of Phlebotomus sergenti (Diptera: Psychodidae) in China and its potential epidemiological implications.

BACKGROUND: Leishmaniasis is a severe parasitic disease transmitted by sand fly bites. Phlebotomus sergenti is a principal vector of Leishmania tropica, causing anthroponotic cutaneous leishmaniasis (ACL) across the Mediterranean, North Africa, South Asia, and West Asia. The Xinjiang Uygur Autonomous Region, located in northwestern China, is a well-recognized endemic focus of visceral leishmaniasis (VL), with sporadic cutaneous leishmaniasis cases also documented in the region. Nevertheless, Ph. sergenti has never been reported in China prior to this study. METHODOLOGY/PRINCIPAL FINDINGS: Fie

Hepatic lipogranulomas in canine visceral leishmaniasis: Morphological and immunohistochemical characterization using IBA-1 and MAC387.

Hepatic involvement is common in canine visceral leishmaniasis (CVL), yet uncommon inflammatory patterns remain poorly documented. This retrospective study aimed to describe the occurrence, histopathological features and immunohistochemical characterization of hepatic lipogranulomas in dogs naturally infected with Leishmania spp. Liver samples from 30 necropsied dogs with confirmed leishmaniasis (between 2010 and 2025) and 60 control dogs without leishmaniasis were evaluated histologically. Cases containing lipogranulomas and/or epithelioid granulomas were further assessed using periodic acid-

Targeting environmental determinants and settlements in public health planning for the containment of visceral leishmaniasis: a One Health perspective.

Visceral leishmaniasis (also known as kala-azar) continues to be a major public health challenge in endemic regions. While vector control and clinical management have gained considerable attention, environmental determinants of transmission are often not prioritized in elimination strategies. In this article, we highlight gaps in surveillance that overlook environmental risks and propose their integration into routine elimination activities. At the same time, we also propose strengthening community-based monitoring, systematic environmental assessments, and harmonized data collection as tools

The utility of point-of-care haemoglobin A1c tests in a very remote Aboriginal and Torres Strait Islander community: an agreement analysis with routine laboratory testing.

BACKGROUND: Leishmaniasis is a prevalent tropical disease caused by more than 20 Leishmania species (Protozoa, Kinetoplastida and Trypanosomatidae). Among different clinical forms of the disease, cutaneous leishmaniasis is the most common form, with an annual 0.6-1 million new cases reported worldwide. This disease's standard treatment is pentavalent antimonial (SbV) that have been used successfully since the first half of the 20th century as a first-line drug. However, treatment failure is an increasing problem that is persistently reported from endemic areas. It is important to define and st

Engineered invasive Lactococcus lactis as an oral vaccine delivery platform is associated with compartmentalized immune modulation and enhances control of Leishmania infantum infection.

Visceral leishmaniasis caused by Leishmania infantum remains a major challenge for vaccine development due to the intracellular localization of the parasite and the need for effective antigen delivery and cellular immune responses. Genetically engineered lactic acid bacteria have emerged as promising platforms for mucosal vaccination because of their genetic tractability and capacity to interact with the host immune system. Here, we evaluated an engineered invasive Lactococcus lactis strain expressing fibronectin-binding protein A (FnBPA) and carrying the chimeric TryPan antigen-encoding plasm

Assessing Designed Model Membrane-Targeting Peptides Found as Potential Molecular Scaffolds against Cutaneous Leishmaniasis.

Recent research has demonstrated therapeutic potential against visceral leishmaniasis, a neglected disease with high prevalence and severity in very populated low- and middle-income regions; however, effective treatments for the cutaneous or mucocutaneous forms remain limited, despite their higher incidence. The negatively charged composition of Leishmania membranes differentiates it from the zwitterionic nature of mammalian cells, providing a basis for selective targeting by cationic peptides. Here, designed peptides (DecP-9, DecP-17, and DecP-18), spanning a broad range of charge and hydroph

Tomatine Augments T Cell-Mediated Protective Immunity Induced by First-Generation Leishmania Antigen Against Visceral Leishmaniasis.

First-generation vaccine candidates against leishmaniasis, based on chemically or physically inactivated whole parasite, have attracted considerable attention owing to their feasibility, scalability of production, and comparatively low manufacturing costs. Vaccine formulations typically require immunostimulatory compounds to elicit robust cell-mediated immunity. Traditionally, adjuvants have been incorporated into vaccine formulation to augment their immunogenic potential. Recent advances over the past decade have yielded novel adjuvant system capable of stimulating both antibody-mediated and
