Protozoan parasites remain a major threat to global health, affecting both humans and livestock. Theileria annulata, an apicomplexan parasite of cattle, transforms host leukocytes into a cancer-like state, while Leishmania donovani causes visceral leishmaniasis in humans. Given their reliance on host signaling and metabolic pathways, we hypothesized that FDA-approved anticancer drugs could be repurposed as antiparasitic agents. A library of 436 compounds was screened against T. annulata, yielding 130 hits at 10 µM, of which 63 exhibited minimal cytotoxicity toward healthy PBMCs. Secondary scre

BACKGROUND: Leishmaniasis presents in three main clinical forms: Cutaneous (CL), Visceral (VL), and Mucocutaneous (MCL). In Hatay, CL is generally caused by L. infantum and L. tropica, with L. major appearing less frequently. Following the 2011 migration wave from Syria, we aimed to identify the causative Leishmania species, particularly L. major, among Syrian patients living in households and tent cities in Hatay, Turkey.
METHODS: Smear preparations from 250 Syrian patients diagnosed with CL at Hatay Mustafa Kemal University, Turkey between 2011 and 2019 were retrospectively analyzed. DNA wa

Obesity significantly increases the risk of developing other chronic diseases and shortens the life expectancy by 5 to 8 years. Childhood obesity is particularly prominent as 80% of teenagers with obesity have shown it to persist into adulthood. With reference to the development of adult medication, the present review summarises the recent research progress in the development of pediatric anti-obesity drugs. The current management guidelines support the use of medication for adolescents aged 12 and above when lifestyle measures are ineffective. Early weight-loss drugs like the serotonergic ago

This narrative review synthesizes the current evidence on the geographic distribution, diagnostic approaches, and clinical and epidemiological characteristics of Viscerotropic leishmaniasis (VTL) in both human and animal hosts in Iran. A comprehensive search of electronic databases (PubMed, Web of Science, etc.) was conducted for articles (1997 to 2025) reporting VTL in Iran with PCR-based species confirmation. Twenty-three studies met the inclusion criteria and were analyzed. The findings indicate that VTL in Iran is predominantly caused by Leishmania tropica, with notable cases attributed to

BACKGROUND: Cutaneous leishmaniasis (CL) is a major public health challenge in Ethiopia, with 20-30,000 new cases annually. Limited awareness and misconceptions contribute to delayed care seeking, stigma, prolonged healing, and reduced uptake of preventive measures in CL-endemic communities. To address these challenges, a participatory community dialogue (CD) intervention was implemented to evaluate a structured CD intervention in improving knowledge, attitudes, and preventive practices related to CL in Kalu District, Ethiopia.
METHODS: A convergent parallel mixed-methods study was conducted

INTRODUCTION: Cutaneous leishmaniasis (CL), caused by protozoa of the genus Leishmania , is a neglected tropical disease. Limited evidence exists to inform the management of CL in children and adolescents, and experience in non-endemic settings varies significantly. We report case data and treatment outcomes for 17 children and young people diagnosed with CL and treated at a specialist tropical medicine center in the United Kingdom.
METHODS: The electronic patient records of 17 patients less than 18 years old with CL who were treated at University College London Hospitals between October 2015

BACKGROUND/AIM: Visceral leishmaniasis, primarily caused by Leishmania (L.) infantum, remains a major global health challenge due to limitations in current chemotherapeutic options, including toxicity and emerging drug resistance. Host-directed therapeutic approaches are increasingly recognized as promising alternatives. All-trans retinoic acid (ATRA) is an immunomodulatory molecule with host-dependent effects on macrophage function; however, its therapeutic use is hindered by instability and poor solubility. Solid lipid nanoparticles (SLNs) offer a controlled and biocompatible delivery platfo

American cutaneous leishmaniasis (ACL) comprises a complex spectrum of immunopathological conditions in which Leishmania (V.) braziliensis and L. (L.) amazonensis elicit markedly divergent host responses. Mucosal leishmaniasis (ML) is characterized by a hyperergic, pro-inflammatory state, whereas anergic diffuse cutaneous leishmaniasis (ADCL) involves a profound impairment of cell-mediated immunity, respectively. The molecular sensors and metabolic circuits that dictate this immunological divergence remain poorly understood. We hypothesized that parasite species-specific programs orchestrate d

BACKGROUND: Hemophagocytic lymphohistiocytosis (HLH) is a rare yet life-threatening hyperinflammatory syndrome, increasingly reported in patients with inflammatory bowel disease (IBD) receiving biologic therapy. While Epstein-Barr virus and cytomegalovirus represent the most frequent infectious triggers, visceral leishmaniasis (VL) is extremely uncommon and may be easily ignored outside endemic areas.
CASE PRESENTATION: We report the case of a 31-year-old man with Crohn's disease treated with escalated anti-TNFα therapy who presented with persistent fever, pancytopenia, hyperferritinemia, hyp

Pleural involvement in visceral leishmaniasis (VL) is rare and may pose a diagnostic challenge, particularly when it mimics pleural tuberculosis. We report the case of a 39-year-old immunocompetent man who presented with a dry cough and night sweats. Initial workup revealed a small lymphocytic exudative pleural effusion, and pleural biopsy showed granulomatous inflammation with a tuberculoid appearance, leading to empirical antituberculosis treatment. Because of clinical deterioration, persistent pleural effusion, and negative molecular testing for Mycobacterium tuberculosis, further investiga

BACKGROUND: Cutaneous leishmaniasis (CL) is a neglected tropical disease with limited therapeutic options due to drug resistance, systemic toxicity, and prolonged treatment duration associated with pentavalent antimonials such as meglumine antimoniate (MAT). Lucilia sericata larvae produce hemolymph containing bioactive compounds with antimicrobial and immunomodulatory properties, suggesting potential as an alternative or adjunct therapy for CL.
METHODS: Hemolymph was extracted from sterile third-instar L. sericata larvae and characterized using SDS-PAGE and Fast Protein Liquid Chromatography

Cutaneous leishmaniasis (CL) is highly prevalent in northern Ethiopia but data on community knowledge, attitudes, and health-seeking behaviours remain limited. A cross-sectional survey was conducted between November and December 2022 in CL-endemic areas of Tigray using mixed sampling and a structured questionnaire administered to 512 households. Knowledge of CL transmission was poor: only 1% correctly identified sand flies as the vector, while 25% believed the disease was genetically acquired. Approximately 67% of participants perceived CL as stigmatizing, and 63.3% reported a preference for t

Dogs play a major role in sustaining the transmission of Leishmania infantum to people, making prevention and treatment of canine leishmaniosis (CanL) public health priorities. However, immune mechanisms underlying progression from subclinical stages to terminal disease in dogs remain ill-defined. To address this gap, we generated a single-cell RNA sequencing map of peripheral immune cells from uninfected and naturally infected dogs across well-defined stages of L. infantum infection. Disease progression was marked by a shift from a lymphoid-to a myeloid-dominated immune landscape. CD4+ T cell

Cutaneous leishmaniasis (CL) is a vector-borne disease caused by Leishmania spp. Molecular methods such as conventional PCR (cPCR) and real-time PCR (qPCR) show high accuracy but remain restricted to reference centers. Loop-mediated isothermal amplification (LAMP) enables rapid DNA amplification under isothermal conditions. LAMP targeting 18S rDNA using WarmStart® Colorimetric LAMP 2× Master Mix and Bst 2.0 DNA Polymerase with SYBR® Green I (BstSg) was evaluated in swab samples (extracted by kit-SW-kit and direct boil-SW-DB) and specimens collected through biopsy from ulcerated lesions of pati

INTRODUCTION: Individual participant data meta-analysis (IPD-MA) is the gold-standard approach for evidence synthesis. Poverty-related infectious diseases, a context in which standalone trials often have small sample sizes and consequently observe relatively few treatment failures, can benefit from this approach.
METHODS: In collaboration with the global research community, the Infectious Diseases Data Observatory (IDDO) has developed a data platform for visceral leishmaniasis (VL)-a neglected tropical disease (NTD), primarily to facilitate IPD-MA aimed at addressing questions of public healt

Leishmaniases are infectious diseases whose vectors are female sand flies that depend on the blood of vertebrates for egg maturation. In this study, we investigated the sand fly species and DNA from Leishmania spp. This entomological/epidemiological study was conducted in a rural area in Nova Andradina, Mato Grosso do Sul, Brazil. From March 2019 to February 2020, five modified Falcão light traps were installed at night once a month. The collected insects were separated by sex into pools of 10 specimens. A sample was used for species identification via microscopic analysis, and another sample

BACKGROUND: Skin NTDs cause mental ill health, primarily through social stigma and discrimination, overburdening services and undermining elimination efforts. A holistic One Health (OH) approach is essential for NTD elimination, but evidence and operational guidance lack. This scoping review documents the current state of integration of mental health and OH-based skin-NTD programmes. It forms part of a series of reviews on the role of OH in skin NTDs, led by the WHO skin NTD working groups.
METHODOLOGY: We searched four peer-reviewed databases for terms related to skin NTDs, OH, mental health

BACKGROUND: Invasive fungal diseases (IFDs) cause significant morbidity and mortality, and liposomal amphotericin B (L-AmB) is a key treatment option for both IFDs and visceral leishmaniasis. Despite extensive clinical experience, real-world data on L-AmB use across diverse clinical settings remain limited.
METHODS: The AMPHO-SITA study was a multicentre, retrospective observational study conducted across 14 Italian centres (September 2020-October 2024). Adult patients receiving at least one dose of L-AmB were included. Baseline demographics, comorbidities, infection and L-AmB treatment chara
