Evaluating the effect of immunization with DNA encoding Phlebotomus sergenti apyrase protein (PsSP42) against Leishmania tropica infection in BALB/c mouse model.

BACKGROUND: Leishmania parasites are transmitted through the bite of infected female sand flies. The sand fly inoculum contains both the parasite and the salivary proteins, which can modulate the immune system's function. Some of these salivary proteins have the potential to be used as a vaccine candidate. Since there have been fewer studies investigating the salivary proteins of Phlebotomus (Ph.) sergenti, this prompted us to select among the three protein members of Ph. sergenti apyrase family (PsSP40, PsSP41, and PsSP42) and measure its effectiveness as vaccine candidate against Leishmania

Antibody response induced by structural proteins from Triatoma virus as potential adjuvants in experimental immunisation models.

INTRODUCTION: Virus-Like Particles (VLPs) are viral protein structures widely used as adjuvants in vaccine formulations due to their ability to stimulate the innate immune response, thereby contributing to the activation of adaptive immunity through the production of different IgG subclasses. The present study evaluated the adjuvant potential of recombinant structural proteins of the Triatoma virus VLPs (TrV-VLPs: VP1, VP2, and VP3) in experimental immunisation protocols for American Cutaneous Leishmaniasis and Chagas disease. METHODS: BALB/c mice were immunised with native antigens of Leishm

Activity-based chemoproteomic profiling reveals the active kinome of Leishmania.

BACKGROUND: Leishmania parasites cause neglected tropical diseases such as cutaneous and visceral leishmaniasis, which have limited treatment options and rising drug resistance. Protein kinases are pivotal in Leishmania biology and attractive drug targets, but their functional status in the parasite remains largely unexplored. METHODS: We applied activity-based protein profiling (ABPP) with custom in-house cell-permeable ATP-site directed probes to map the ligandable, "active kinome" of Leishmania mexicana. Three related covalent probes featuring an ATP-mimetic scaffold, electrophilic warhead

Platelet DKK1 promotes tolerogenic dendritic cells and non-healing responses in cutaneous leishmaniasis.

Activation of platelet TLR1/2 at the initiation of Leishmania major infection results in the release of the Wnt antagonist Dickkopf-1 (DKK1). To examine the role of platelet MyD88 and/or DKK1 in regulating the immune response, genetic approaches using BALB/c mice deficient in platelet MyD88 (MyD88PKO) or DKK1 (DKK1PKO) were used. Genetically deficient mice exhibit significant changes in the immune response throughout infection. Initially, the levels of activated neutrophils and neutrophil-platelet aggregates were reduced at the site of infection. Subsequently, an anti-leishmanial Th1-response

In vitro, In silico, and Scanning Electron Microscopy Investigations of Anti-leishmanial Efficacy of New Synthetic 3-Imidazolyl flavan oxime on Leishmania tropica Parasites.

INTRODUCTION: Antimonials have been traditional therapeutic agents for leishmaniasis for decades, but selecting the optimal treatment regimen remains challenging. Alternative systemic treatments, including liposomal amphotericin B and sodium stibogluconate, have shown unclear efficacy in Leishmania tropica cutaneous leishmaniasis. The objective of this study was to evaluate the efficacy of a new synthetic 3-Imidazolyl flavan oxime (IFO) against Leishmania tropica, using in vitro, in silico, and Scanning Electron Microscopy (SEM) analyses. METHODS: A standard strain of Leishmania tropica was c

Natural infection by Trypanosoma spp. (Trypanosomatida: Trypanosomatidae) in Micropygomyia cayennensis cayennensis (Diptera: Psychodidae) on Northern Colombia.

In Colombia, sand flies (Diptera: Psychodidae) are recognized as primary vectors of Leishmania spp. However, there are reports of infections caused by unidentified trypanosomatid parasites in sand flies circulating within leishmaniasis-endemic areas (2017 to 2018). This study aimed to identify trypanosomatid parasites other than Leishmania spp. infecting sand flies in a visceral leishmaniasis focus in northern Colombia. Between March 2017 and March 2018, sand flies were actively collected from tree bases and cavities and subsequently dissected for taxonomic identification and the detection of

Functional Effects of Cyclization on Li1 Peptide Activity against Metacyclic Leishmania amazonensis Internalization.

Leishmaniases are caused by protozoa of the genus Leishmania, whose metacyclic promastigote forms initiate infection in the mammalian host. Building upon previous work with Leishmania infantum, this study evaluated the cyclic peptide Li1 and its linear analogue Li1nc regarding their capacity to interfere with Leishmania amazonensis infection. Both peptides were synthesized by solid-phase peptide synthesis (SPPS-Fmoc) and tested after pre-exposure of metacyclic promastigotes prior to infection of murine peritoneal and THP-1-derived macrophages. In contrast to the previous study conducted by our

Impact of Integrated Control Interventions on Sandfly Populations in Human and Canine Visceral Leishmaniasis Control in Araçatuba, State of São Paulo, Brazil.

Visceral leishmaniasis (VL) is a serious vector-borne disease affecting humans and dogs, posing major public health challenges in endemic regions. Control efforts often target sandfly vectors, whose larvae and pupae develop in soil. Environmental management, such as removing organic matter, reducing moisture, and pruning vegetation, aims to limit breeding sites and reduce sandfly populations. This study evaluated the impact of integrated interventions on sandfly behavior in priority areas for VL control in Araçatuba, São Paulo, Brazil. The control strategy combined environmental management, ca

Molecular Detection of Leishmania Species on Microscopically Negative Smears in Gonbad-e-Kavus, Northeastern Iran.

BACKGROUND: The diagnosis of cutaneous leishmaniasis (CL) is typically based on microscopic examination of Giemsa-stained smear. However, this method can lead to false negative and false positive results. Therefore, the study aimed to identify L. major and L. tropica species by investigating microscopically negative smears suspected of CL using the multiplex nested polymerase chain reaction (PCR) method. MATERIALS AND METHODS: This study was conducted on 227 microscopically negative specimens from individuals suspected of having CL referred to the CL Treatment and Diagnosis Center in Gonbad-e

Differential microRNA expression profiles and predicted miRNA-mRNA regulatory networks in human macrophage-like cells infected with Leishmania infantum.

MicroRNAs (miRNAs) regulate gene expression and play a crucial role in numerous diseases, including infections. Leishmaniasis is a neglected infectious disease occurring in different forms (i.e., cutaneous, mucocutaneous, and visceral) caused by a protozoan belonging to the Leishmania genus. The parasite infects mainly the macrophages, establishing a niche permissive for its proliferation. Leishmania parasites are known to modulate host gene expression, including miRNAs; however, the specific role of these miRNAs in driving host transcriptomic changes remains to be fully characterized. The aim

An Overview of the Factors Related to Leishmania Vaccine Development.

Leishmaniasis is an infectious disease caused by several species of Leishmania parasites that preferentially infect macrophages as host cells. These intracellular parasites can evade the main microbicidal effector mechanisms of phagocytic cells and, in turn, are able to stimulate marked Th2 or regulatory T cell immune responses, which are not protective for the host. The presence of a non-protective immune response, together with the multiplication and spread of Leishmania parasites throughout the tissues, leads to the main clinical forms of leishmaniasis, such as cutaneous and visceral leishm

Retrospective entomological and eco-epidemiological survey of a leishmaniasis outbreak in southern Spain.

Leishmaniasis is a well-known vector borne disease with a high incidence in southern European countries. In Spain, Leishmania infantum infections typically occur as sporadic and infrequent outbreaks. However, in recent years, several outbreaks have been reported in different regions, accompanied by increasing hospitalization rates. In 2023, 31 human cases of Leishmania infection were reported in hospitals across 15 municipalities in Córdoba (southern Spain). Visceral leishmaniasis was the predominant form (77.4%), followed by cutaneous (19.4%), and mucocutaneous (3.2%) forms. A retrospective a

Protein undernutrition alters the colonic bacteriome, disrupts intestinal immune homeostasis, and impairs control of Leishmania infantum infection in a murine model of visceral leishmaniasis.

INTRODUCTION: Undernutrition is a significant global health issue that exacerbates susceptibility to infectious diseases, including visceral leishmaniasis (VL), caused by Leishmania infantum. Here, we investigated the interplay between undernutrition, immune responses, and the colonic microbiota composition in a murine model of VL. METHODS: We used BALB/c mice subjected to a low-protein diet and infected with L. infantum to analyze the effects on systemic and local immune responses, microbiota composition, and parasite load. RESULTS: Undernutrition significantly downregulated the mRNA expres

Potentiality of LPS in Ameliorating the Histopathological Responses in Visceral Leishmaniasis-Infected Mice.

OBJECTIVE: Visceral leishmaniasis is the second most fatal parasite illness worldwide, and it is the most severe type of leishmaniasis. LPS is a crucial chemical compound on the bacterial cell wall that the host recognizes and uses to launch an immune response to eliminate invasive infections. MATERIALS AND METHODS: Four concentrations of LPS were used to treat infected mice with visceral leishmaniasis (20, 40, 60, and 80 ng/mL). Differential cell count and phagocytic index tests were done, then the liver and spleen were separated, and a histopathological assay was performed. RESULTS: The fi
