Pyrazolo[1,5-c]quinazoline-based heterocycles exhibit potent antileishmanial activity: Mechanistic insights and toxicity profile.

Leishmaniasis remains a major global health challenge, with current chemo-therapeutics limited by high cost, toxicity, and poor bio-availability. In this study, seventeen new pyrazolo[1,5-c]quinazoline derivatives were designed and synthesized and evaluated for their in vitro antileishmanial activity against Leishmania donovani promastigotes. Compounds 19e and 20k demonstrated superior potency, exhibiting IC50 values lower than miltefosine, while showing minimal cytotoxicity in HeLa and RAW cells (CC50), resulting in favorable selectivity indices. Mechanistic investigations revealed that both

Lupoid Cutaneous Leishmaniasis: A Case Report.

BACKGROUND: Lupoid Cutaneous Leishmaniasis(LCL) is rare type of cutaneous leishmaniasis that clinically and histologically mimics other granulomatous skin conditions like Lupus Vulgaris. This similarity often cause difficulty in diagnosis, especially in endemic areas. CASE PRESENTATION: We report a case of a 29-year-old male who presented with isolated plaque on right cheek persisting for 2 years. The lesion was slowly progressive, non-tender and had not responded completely to local therapy. Smear of initial lesion revealed +LD bodies but the recurrent lesion showed no amastigotes on smear.

LABCG1 Affects Leishmania (Leishmania) Amazonensis Infectivity but Not Virulence or Sand Fly Infection.

Leishmaniases are a complex spectrum of diseases caused by protozoan parasites of the genus Leishmania. Overall, they can be classified into two main clinical forms: tegumentary and visceral leishmaniasis (TL and VL, respectively). Our group has been studying LV79 and PH8 strains of Leishmania (Leishmania) amazonensis, one of the predominant TL-causing species in Brazil. PH8 is more infective in vitro and more virulent in vivo compared to LV79. Comparative proteomic analysis of promastigotes of both strains revealed that the ABCG1 transporter is 23.57-fold more abundant in PH8 than in LV79 str

Trypanosomatidae (Kinetoplastea: Trypanosomatida) diversity infecting the South American coati Nasua nasua (Carnivora: Procyonidae) in urban forest fragments in the Brazilian Midwest.

INTRODUCTION: South American coatis are reported as key-hosts of trypanosomatid species in Brazilian Pantanal, presenting high parasitemia and infection rates, long-lasting infections, ability to bioaccumulate Trypanosoma evansi, T. rangeli and different genotypes of T. cruzi, and biological features that favor these parasites' transmission. Moreover, this mammal species was also reported parasitized by the monoxenous trypanosomatid Crithidia mellificae and included in the epidemiological scenario of Visceral Leishmaniasis in Campo Grande (CG), Brazilian Midwest. Within this context, the aim o

Trypanosomatid parasites, translational research in a One Health, One World environment.

Adequate policies to achieve universal access to high-quality diagnostics and therapeutics in the challenge represented by Trypanosomatidae-borne diseases remain essential to command surveillance as a successful tool and a way to delay the unavoidable surge of resistance toward the currently used drugs. In the case of leishmaniasis, even for uncomplicated cutaneous leishmaniasis, most patients in endemic regions are treated with systemic therapies due to reasons such as local protocol misalignment, limited access to new technologies, and lesions being located in anatomical areas other than the

A Trypanothione Synthetase-Based Subunit Vaccine Induces Protective Th1 Immunity against Visceral Leishmaniasis in Mice.

Leishmaniasis, caused by Leishmania species, remains a major global health challenge due to limited therapeutic options, drug resistance, and relapse. To achieve long-term protection, an effective vaccine is urgently needed. In this work, we evaluated Leishmania donovani trypanothione synthetase as a vaccine candidate in BALB/c mice, formulated with CpG oligodeoxynucleotides and interleukin-2 to enhance immunogenicity. Mice were immunized with LdTryS alone or in combination with CpG and IL-2, followed by a challenge infection. Immunological parameters assessed included parasite burden, cytokin

Mechanistic evaluation of niosomal Rhoifolin in antileishmanial therapy and its additive potential with meglumine antimoniate.

Leishmaniasis is a neglected tropical disease that urgently requires safer and more effective therapeutic strategies. Rhoifolin (RF), a natural flavonoid with anti-inflammatory and antiparasitic potential, has shown promising biological activities against several infectious diseases. In the present study, the antileishmanial effects of free RF and its niosomal formulation (NRF), alone and in combination with meglumine antimoniate (MA), were investigated against Leishmania major. Niosomes were prepared using Span 40, Tween 40, and cholesterol, resulting in high encapsulation efficiency (90.9%)

Gender-Attributed Persona Prompts and the Diagnostic Accuracy of Proprietary and Open-Weight Large Language Models in Chagas Disease and Visceral Leishmaniasis: A Paired Experimental Study.

Background: Large language models (LLMs) are increasingly considered as adjuncts for differential diagnostic reasoning, yet their sensitivity to gender-attributed cues in the persona prompt-particularly for neglected tropical diseases (NTDs) and in non-English clinical settings-remains poorly characterised. Objective: The objective of this study was to compare the diagnostic accuracy of one proprietary and three open-weight LLMs for Chagas disease (CD) and visceral leishmaniasis (VL) under paired persona-prompt conditions in which the only manipulated variable was the linguistic gender of the

Investigation of viscerotropism-associated genes in Leishmania tropica strains causing visceral leishmaniasis.

Leishmania infantum is the predominant etiological agent of visceral leishmaniasis (VL), whereas Leishmania tropica is classically associated with cutaneous leishmaniasis (CL). However, sporadic reports from Turkiye and other endemic settings indicate that L. tropica may also cause VL, suggesting that specific parasite determinants enable adaptation to visceral tissues. The molecular basis of this phenotypic shift remains insufficiently understood. This study aimed to define genetic factors associated with visceralization in L. tropica by integrating clinical isolates from patients with VL and

INTRODUCTION: Mucosal leishmaniasis (ML) is a rare, destructive, and recurrent protozoal infection caused by Leishmania species, most commonly Leishmania braziliensis. ML can develop months to years after initial cutaneous leishmaniasis (CL) and may lead to severe mucosal destruction if untreated. CASE PRESENTATION: We present a case of a 48-year-old healthy male from Venezuela with prior CL treated with intravenous meglumine antimoniate who subsequently developed progressive nasal symptoms consistent with ML. Initial therapy with oral fluconazole was ineffective, but treatment with miltefosi

Cross-Species Hepatic Metabolism of the Antileishmanial Chalcone NAT22 Generates Metabolites with Predicted Enhanced Affinity for the Parasite Target cTXNPx.

Background/Objectives: Human and canine leishmaniases are neglected diseases with limited therapeutic options. The nitrochalcone NAT22, a high-affinity inhibitor of the essential parasite enzyme tryparedoxin peroxidase (cTXNPx), has emerged as a promising antileishmanial candidate. Interestingly, NAT22 demonstrated superior efficacy when administered orally rather than intralesionally, suggesting a metabolism-driven activity enhancement. Since in vivo studies with chalcones have been conducted exclusively in mice, this study aimed to determine whether mice are suitable models for oral chalcone
