Exploratory Computational Assessment of Lactiplantibacillus plantarum K25-Derived Fatty Acid Metabolites Against Leishmania donovani Centrin.

Probiotic derived metabolites have emerged as promising bio-active compounds with diverse antimicrobial and therapeutic applications. In the present study, an integrative metabolomic and computational approach was employed to investigate fatty acid metabolites produced by Lactiplantibacillus plantarum K25 and evaluate their potential activity against virulence associated targets to Leishmania donovani, the causative agent of visceral leishmaniasis (kala-azar). L. plantarum K25 exhibited robust growth across linoleic acid concentrations ranging from 1 to 10% (w/v), with maximum metabolite produ

Application of image analysis in the evaluation of blood feeding by Lutzomyia longipalpis using an artificial feeding system with synthetic membranes.

BACKGROUND: Blood feeding is essential for the maintenance of laboratory colonies and for experimental studies involving sand fly-parasite interactions. However, artificial feeding systems for Lutzomyia longipalpis, the principal vector of Leishmania infantum, the etiological agent of visceral leishmaniasis in the Americas, remain limited by low engorgement rates and methodological constraints in blood meal quantification. OBJECTIVES: This study evaluated the feeding performance of Lu. longipalpis on different membranes in an artificial feeder and applied an image-based approach to quantify b

Arthropod-Related Health Threats (ARHTs) and Nuisance Arthropods in North Khorasan Province, Iran: A Comprehensive Review.

The arthropod vectors, such as mosquitoes, sand flies, and ticks, transmit various pathogens to humans and animals, leading to public health issues. Therefore, an effective disease surveillance system is essential to provide information on these diseases in order to monitor their trends worldwide. The current study focused on the prevalence of arthropod-borne diseases and their vectors in North Khorasan Province located in northeastern Iran, bordering Turkmenistan. A narrative literature review was conducted to analyze the status of arthropod-related health threats (ARHTs) and their vectors in

Clinical and Laboratory Predictors for Differentiating Visceral Leishmaniasis from Hematological Malignancies in Adults with Pancytopenia.

Visceral leishmaniasis (VL) is a systemic, life-threatening zoonotic disease. VL and hematological malignancies (HMs) may present with overlapping clinical and laboratory features. The primary endpoint of our study was to identify the clinical and laboratory parameters for differentiating VL from HMs in adult patients with pancytopenia. The secondary endpoint was to evaluate the clinical and laboratory findings in patients with VL. In this 15.5-year retrospective case-control study, adult patients with pancytopenia who were diagnosed with VL or HMs at a tertiary referral hospital in Antalya we

Transcriptomic evidence of an IFN-gamma-associated inflammatory state in active visceral leishmaniasis: an exploratory comparison with pediatric HLH.

BACKGROUND: Visceral leishmaniasis (VL) can progress to secondary hemophagocytic lymphohistiocytosis (HLH), but transcriptomic similarities between active VL and pediatric HLH remain uncertain. METHODS: We integrated VL whole-blood expression data (GSE125992), sorted CD68-positive monocyte bulk-expression data from pediatric HLH and severe sepsis (GSE150707), and supportive Leishmania-infected mouse spleen single-cell data (GSE240385). Differential expression, pathway enrichment, an exploratory 132-gene HLH-versus-sepsis-derived ssGSEA score, FARDEEP-LM22 deconvolution, signature-excluded WGC

Pentalinonsterol at the Host-Parasite Interface: Antileishmanial Activity, Host Immunomodulation, and Unresolved Mechanisms.

Leishmaniasis remains difficult to treat because available therapies are constrained by toxicity, administration burden, variable efficacy, cost, and drug resistance. This critical narrative review evaluates the preclinical evidence for pentalinonsterol (PEN), a plant-derived cholestane-type steroidal ketone originally isolated from the Mayan medicinal plant Pentalinon andrieuxii. Experimental studies demonstrate PEN activity against Leishmania promastigotes and intracellular amastigotes, together with marked parasite ultrastructural injury; however, no parasite molecular target has been valid

Digital PCR-based quantification of Leishmania infantum in Lutzomyia longipalpis: comparison between promastigote and macrophage-derived amastigote infection models.

Visceral leishmaniasis (VL) remains a major public health concern in Latin America, particularly in Brazil, where Leishmania infantum is primarily transmitted by the sand fly Lutzomyia longipalpis. Understanding parasite development within the vector is essential for elucidating transmission dynamics, yet classical microscopic assessments often lack sensitivity for detecting early or low intensity infections. In this study, Lu. longipalpis females were experimentally infected with either promastigote or amastigote forms of Le. infantum and the infection dynamics were evaluated by both optical

Phosphatidylserine Blockade Limits Systemic Dissemination of Leishmania Amazonensis Amastigotes.

Leishmania amazonensis is associated with cases of cutaneous, disseminated, diffuse, mucosal, and visceral leishmaniasis (VL). The mechanisms underlying its ability to persist and spread after the initial clinical cure are unclear, although intrinsic characteristics of the parasite and the host may be involved. Phosphatidylserine (PS) is a pathogenic factor in both experimental models and in patients. It is related to macrophage infection, regulation of inflammatory and immune responses, and the extent and severity of the clinical outcomes of the disease. In a murine model of systemic infectio

Immune-mediated remodelling of tissue architecture in Post-Kala-Azar Dermal Leishmaniasis (PKDL).

BACKGROUND: Post-Kala-Azar Dermal Leishmaniasis (PKDL), a dermal sequel of apparently cured visceral leishmaniasis (VL), presents with macular or polymorphic lesions. A relationship, if any, between immune cell infiltration and cytokines potentially causing tissue-remodelling remains poorly defined, and was the focus of this study. METHODOLOGY/PRINCIPAL FINDINGS: Skin biopsies along with plasma from PKDL patients and healthy controls were analyzed. Tissue sections were assessed for immune cell subsets (CD8 ⁺ T cells, CD68 ⁺ macrophages, CD20 ⁺ B cells, CD66b⁺ neutrophils), associated cytokine

Case Report: Multiple brain abscess-like lesions with concurrent detection of Nocardia amamiensis and Leishmania infantum in cerebrospinal fluid.

Central nervous system nocardiosis is rare and potentially life-threatening, and human infection due to Nocardia amamiensis has seldom been reported; central nervous system involvement in visceral leishmaniasis is likewise uncommon. We report a 69-year-old man who developed progressive impairment of consciousness and coma after recurrent fever. Brain magnetic resonance imaging revealed multiple abscess-like lesions in both cerebral hemispheres, the cerebellum, and the brainstem, accompanied by acute or subacute cerebral infarctions. The cerebrospinal fluid opening pressure was 300 mmH2O. The w

Targeting the proteasome in Leishmania donovani: developments from preclinical to Phase I and ii trials.

INTRODUCTION: Visceral leishmaniasis (VL), a neglected tropical disease caused by Leishmania donovani and L. infantum, which affects poor areas of South Asia, East Africa, and Brazil, and is fatal if left untreated. Current treatments, including liposomal amphotericin B, miltefosine, paromomycin, and pentavalent antimonials, are limited by emerging resistance, high cost, cold storage, need for parenteral administration. These drawbacks of the currently available therapy entail the development of a safe, short, and oral therapy for VL treatment. AREA COVERED: This review summarizes current kno

Effects of centrifugation-based therapeutic plasma exchange on coagulation parameters in dogs with canine visceral leishmaniasis.

As in humans, therapeutic plasma exchange (TPE) has been investigated in veterinary medicine for managing immune-mediated diseases such as canine visceral leishmaniasis (CVL). TPE may improve clinical outcomes by removing circulating immune complexes and excess antibodies; however, its physiological effects must be understood to ensure safety. Given the scarcity of studies in animals, this study evaluated the effects of centrifugation-based TPE in dogs with CVL receiving fresh frozen plasma (FFP) as replacement fluid. Thirty-one dogs with CVL that failed to respond to conventional therapy were

Haemophagocytic Lymphohistiocytosis Triggered by Visceral Leishmaniasis Masquerading as Epstein-Barr Virus (EBV)-Associated Haemophagocytic Lymphohistiocytosis in Rheumatoid Arthritis: A Case Report and Literature Review.

Haemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory syndrome caused by uncontrolled immune activation. In adults, it is usually secondary to an underlying condition, such as infection, malignancy, or autoimmune disease. Identifying this trigger is essential, as treatment depends on the cause. Visceral leishmaniasis (VL) is a systemic parasitic infection that may mimic or trigger HLH, particularly in immunosuppressed patients. We report a 59-year-old man with seropositive rheumatoid arthritis, for which he received immunosuppressive treatment with methotrexate and

The gene expression signature of multinucleated giant cells in the spleens of Leishmania donovani-infected mice.

Multinucleated giant cells (MGCs) are associated with chronic inflammation and infectious diseases, but their biological characteristics in vivo remain poorly understood because intact cells are difficult to isolate from tissues. In visceral leishmaniasis, heavily parasitized, hemophagocytic MGCs are observed in the spleens of Leishmania donovani-infected mice, suggesting that these cells may contribute to parasite persistence and disease pathology. In this study, we established a method for isolating intact MGCs from the spleens of chronically infected BALB/c mice and performed transcriptomic

Widespread transmission in diverse ecotypes challenges visceral leishmaniasis control in East Africa.

East Africa is emerging as the global hot spot of visceral leishmaniasis (VL), yet efforts to eliminate it are hindered by substantial knowledge gaps in its ecoepidemiology. Here, we report on the high prevalence of Leishmania infection in Phlebotomus orientalis in Marsabit county, Kenya (3.9%), and Gedaref state, Sudan (3.6%), where this species comprised 99.8% (n = 1185) and 100% (n = 1350) of captured Phlebotomus females, respectively. In Aba Roba, Ethiopia, Phlebotomus martini accounted for 94.4% of 184 collected Phlebotomus females and had a lower infection rate of 1.5%. Phlebotomus orien

Leishmania respiration profiling as a One Health functional marker associated with antimony resistance in human and canine leishmaniases.

Rapid functional tools for assessing antimony susceptibility in Leishmania are needed to strengthen resistance surveillance across human and veterinary settings. We developed a real-time oxygen-consumption assay using the Resipher analyzer and evaluated normalized oxygen-consumption rate (OCR) as a functional correlate of antimony (Sb) resistance in Leishmania infantum and Leishmania donovani. A density of 1 × 107 parasites/mL and a 48-h readout provided the most robust analytical window, and rotenone markedly reduced OCR, demonstrating assay responsiveness to respiratory perturbation. Antimon

Synthesis, Antiparasitic Activity, and ADME Profile of Morpholinephenylbenzamides: A Pharmacological Study Against Chagas Disease and Visceral Leishmaniasis.

Chagas disease and leishmaniasis are neglected tropical diseases with limited treatment options. This study reports a hit-to-lead optimization of morpholinephenylbenzamides, starting from hit 1, which showed potent activity against Trypanosoma cruzi (IC50 0.29-0.76 μM) and Leishmania infantum (IC50 10.7 μM), despite a suboptimal ADME profile. A structure-activity relationship study of twenty-four novel derivatives provided key pharmacophoric insights. While Trypanosoma cruzi was less susceptible to chemical modifications, compound 20 exhibited potency comparable to benznidazole (IC50 3.58 μM)

Canine exposure to vector-borne pathogens in a rural Brazilian landscape.

Vector-borne pathogens (VBPs) of medical and veterinary concern have mostly been studied in urban dog populations, while animals in rural settings, with limited access to ectoparasite prevention, have received less attention. In this study, we assessed canine exposure to VBPs in a rural Brazilian landscape. From August to October 2024, we collected 155 blood samples from dogs domiciled in a rural area of the municipality of Garanhuns, Pernambuco, Northeastern Brazil. Samples were assessed using an immunofluorescence antibody test (IFAT) to detect antibodies against Trypanosoma cruzi and using
