Arthropod-Related Health Threats (ARHTs) and Nuisance Arthropods in North Khorasan Province, Iran: A Comprehensive Review.

The arthropod vectors, such as mosquitoes, sand flies, and ticks, transmit various pathogens to humans and animals, leading to public health issues. Therefore, an effective disease surveillance system is essential to provide information on these diseases in order to monitor their trends worldwide. The current study focused on the prevalence of arthropod-borne diseases and their vectors in North Khorasan Province located in northeastern Iran, bordering Turkmenistan. A narrative literature review was conducted to analyze the status of arthropod-related health threats (ARHTs) and their vectors in

In Silico Design and Immunogenicity Evaluation of a Multiepitope Vaccine Candidate Against Leishmania tropica in BALB/c Mice.

BACKGROUND: Cutaneous leishmaniasis caused by Leishmania tropica remains a major neglected parasitic disease with limited therapeutic options and frequent treatment failure, highlighting the urgent need for effective vaccine-based interventions. Multiepitope vaccine (MEV) strategies provide a rational and targeted approach to induce defined cellular and humoral immune responses against Leishmania infection. METHODS: In this study, a MEV candidate was designed using conserved cytotoxic T lymphocyte (CTL), helper T lymphocyte (HTL), and linear B-cell epitopes derived from immunodominant Leishma

Performance of recombinase polymerase amplification assay for cutaneous leishmaniasis detection: a systematic review and meta-analysis.

BACKGROUND: Sensitive and robust point-of-care (PoC) assays are urgently needed for the diagnosis of cutaneous leishmaniasis (CL). Recombinase polymerase amplification (RPA), an isothermal molecular assay, has shown promise for CL diagnosis using either a lateral-flow assay (LFA) or a mobile-suitcase-laboratory (MSL) platform. However, its diagnostic performance has been inconsistent across studies. This systematic review and meta-analysis aimed to determine the pooled sensitivity and specificity of RPA for CL diagnosis. METHODS: A comprehensive search was conducted in Google Scholar, EMBASE,

Neglected tropical diseases in the Arabian Peninsula: a systematic review of leishmaniasis and rickettsioses.

BACKGROUND: Vector-borne neglected tropical diseases are still an underrecognized public health problem in the Arabian Peninsula. Of these, leishmaniasis is endemic in multiple regions, while the epidemiology of rickettsial infections remains incompletely understood. This systematic review synthesizes existing evidence on the epidemiology, distribution, and surveillance of these two diseases in the region. METHODS: Following PRISMA 2020 guidelines, a systematic review was performed. The protocol was registered with PROSPERO. We searched PubMed/MEDLINE, Scopus, Web of Science, EMBASE, and rele

Clinical and treatment-related predictors of failure of pentavalent antimonial therapy for cutaneous leishmaniasis: a systematic review.

BACKGROUND: Pentavalent antimonials remain widely used for cutaneous leishmaniasis (CL), but treatment failure prolongs disease and increases patient burden. We synthesized routinely available clinical and treatment-related predictors of antimonial failure. METHODS: PubMed/MEDLINE and Europe PMC were searched through 14 August 2026 and Scopus through 30 August 2026, supplemented by citation and targeted website searches. Cohort, case-control, and prediction-model studies of antimony-treated CL were eligible. QUIPS assessed risk of bias. Heterogeneous outcomes, regimens, and overlapping cohort

Molecular screening for Wolbachia endosymbionts in phlebotomine sand flies within the North Central Province, Sri Lanka.

Cutaneous leishmaniasis is a growing public health threat in Sri Lanka, transmitted primarily by phlebotomine sand flies. While biological control using Wolbachia has proven effective against other vector-borne diseases, its potential application in phlebotomine sand flies remains largely unexplored. Therefore, investigating the natural prevalence of these endosymbionts in Sri Lankan sand fly populations is well justified. To address this knowledge gap and evaluate the feasibility of symbiont-mediated control, we conducted a six-month entomological survey across four endemic sampling areas in

One Health determinants of clinically assessed cutaneous leishmaniasis in Sodo District, Ethiopia: a community-based cross-sectional study.

Cutaneous leishmaniasis (CL) is a neglected tropical disease shaped by interacting environmental, wildlife, household, and behavioral factors, yet few community-based studies have evaluated these determinants simultaneously within a One Health framework. We assessed multidomain determinants of clinically assessed CL among 1,356 individuals from six kebeles in Sodo District, Ethiopia. CL was defined as an active lesion or typical scar consistent with current or previous infection. Exposures were grouped into four analytic domains: household/structural, environmental, behavioral, and reservoir/a

microRNA and Regulatory Networks of Human Macrophage During Leishmania major In Vitro Infection.

Leishmania (L.) major modulates host microRNA (miR) expression to promote intracellular survival, yet the roles of specific miRs during early infection of human macrophages remain poorly defined. We investigated the dynamic regulation of miR-21-5p and miR-146a-5p in THP-1-derived macrophages infected with L. major over an early post-infection time course (3-48 h), alongside miR-30a-3p and miR-101-3p. Both miR-21-5p and miR-146a-5p were significantly upregulated during the early stages of infection (3-24 h) before declining by 48 h. Using antisense LNA inhibitors, we achieved independent single

[Cutaneous coinfection with tuberculosis and leishmaniasis].

We describe the case of a 16-year-old female patient from Bolivia, with a family history of pulmonary tuberculosis, who presented with a two-month history of ulcerated lesions on the dorsum of her left hand. A skin biopsy revealed amastigotes consistent with Leishmania spp., and antiparasitic treatment was initiated. Subsequently, Mycobacterium tuberculosis was also isolated, and antituberculosis treatment was started, resulting in a favorable outcome.

Temperature seasonality and spatially structured variation in cutaneous leishmaniasis in Algeria: A source-verified national panel, 2000-2024.

Algeria carries one of the highest cutaneous leishmaniasis (CL) burdens worldwide, yet the climatic structure of its provincial distribution has not been quantified nationally with year-specific covariates. We assembled a balanced panel of all 48 wilayas over 2000-2024 (1200 province-years), linking notified CL counts to annual ERA5-Land bioclimatic indicators, and verified the case series province-by-province against the primary national epidemiological bulletins. Counts were strongly overdispersed and modelled by negative binomial regression with a log-population offset and province-clustere

Travel history and occupation among diagnosed Cutaneous Leishmaniasis cases in Guyana: A multi-site retrospective case series, 2017-2021.

BACKGROUND: Cutaneous Leishmaniasis (CL) in Guyana has not been systematically characterised in the peer-reviewed literature for several decades. This study aimed to describe the demographic, occupational, geographic, and clinical profile of diagnosed CL cases at Guyana's principal diagnostic sites, and to test, within this case series, whether field-based occupation is associated with reported interior travel and whether travel history is associated with time to presentation. METHODOLOGY/PRINCIPAL FINDINGS: Retrospective case series (chart review) of CL diagnoses recorded at three facilities

Systemic IgG responses to Leishmania braziliensis-derived extracellular vesicles vary across clinical forms of American tegumentary leishmaniasis and associate with early therapeutic outcome.

BACKGROUND: American tegumentary leishmaniasis (ATL) caused by Leishmania braziliensis is characterized by marked clinical and immunopathological heterogeneity. While cellular immune responses have been extensively investigated, the contribution of humoral immunity, particularly against parasite-derived extracellular vesicles (EVs) components, remains poorly understood. Here, we evaluated systemic IgG responses against EVs derived from L. braziliensis isolates associated with distinct clinical forms of ATL and assessed their association with disease presentation and therapeutic outcome. Altitudinal cross-sectional study of the sand fly fauna of the highlands of Tigray Region, northern Ethiopia.

BACKGROUND: Cutaneous leishmaniasis (CL), caused by Leishmania aethiopica and transmitted by Phlebotomus longipes, is endemic in northern Ethiopia. However, sand fly fauna is poorly documented in the northern Tigray region. High-altitude CL cases have also been observed, but their local acquisition is uncertain. To identify the faunal composition, their geographical and altitudinal distribution, cross-sectional surveys were conducted in ten villages (average altitude range: 2411 - 3186 meters) with confirmed CL cases across four endemic districts of the region. A total of 170 light trap-nights

Antileishmanial potential of polar fractions from Amaranthus blitoides S. Watson, Caucalis platycarpos L., and Cistanche flava (C.A.Mey.) Korsh.: evidence from in vitro and in vivo studies.

BACKGROUND: Cutaneous leishmaniasis (CL) is recognized by the World Health Organization (WHO) as a research priority, yet its treatment remains challenging. CL lesions arise from complex interactions between the parasite and host immunity, where excessive inflammation can delay healing. Given the anti-inflammatory and immunomodulatory properties of medicinal plants, this study evaluated the effects of polar fractions from methanolic extracts of Amaranthus blitoides S. Watson, Caucalis platycarpos L., Cistanche flava (C.A.Mey.) Korsh., and their mixtures on CL caused by Leishmania major. METHO

Skin Disease After Refugee Resettlement: A Practical 90-Day Approach for Dermatologists and Primary Care Physicians.

The first months after refugee resettlement create an opportunity to identify transmissible, disabling, and undertreated skin disease. In the United States, the domestic medical examination generally occurs 30-90 days after arrival, but the clinical principles are relevant across resettlement systems. We conducted a structured narrative review of peer-reviewed publications and organizational or public-health sources. Evidence varied by migration stage, region, age, and referral pathway; camp and specialty-clinic data should not be treated as post-resettlement prevalence estimates. Physicians s
