Leishmaniasis.

Leishmaniases comprise clinically distinct diseases caused by the protozoan parasite leishmania, which is transmitted through the bite of infected sand flies. Cutaneous leishmaniasis is the most common form and manifests as a localized skin lesion. Mucosal leishmaniasis causes destructive nose, mouth, and throat lesions. Visceral leishmaniasis is a potentially life-threatening form that results from bloodborne dissemination of the parasites. The number of cases of cutaneous leishmaniasis is increasing, particularly in the Eastern Mediterranean region, and the prevalence of visceral leishmanias

Temporal transcriptional dynamics in cutaneous leishmaniasis reveal novel targets for therapeutic interventions in a dermal mouse model.

Cutaneous leishmaniasis (CL) caused by Leishmania braziliensis results in chronic skin ulceration and remains challenging to treat. While human transcriptomic studies have identified pathways driving immunopathology, the early events of infection and the molecular transitions from lesion formation to healing are still poorly understood. Here, we performed a longitudinal transcriptomic analysis of skin lesions and draining lymph nodes (dLNs) in the BALB/c ear dermal model infected with L. braziliensis, which recapitulates features of human CL. Using bulk RNA sequencing at 2, 6, and 48 hours and

Evaluation of a new rapid diagnostic test, based on the chimeric protein Q5, for the diagnosis of human and canine forms of visceral leishmaniasis.

BACKGROUND: Visceral leishmaniasis (VL), caused by Leishmania infantum, is the most severe form of leishmaniasis, prevalent in many countries, but still with limitations in diagnosis for both human and canine hosts. Serological assays based on recombinant proteins are the most efficient diagnostic alternatives, with the rapid diagnostic test (RDT) being the most cost-effective. The previously described chimeric Q5 is a recombinant protein derived from three native L. infantum antigens, which is potentially useful for both human and canine VL diagnosis, through preliminary enzyme-linked immunos

Synergistic anti-leishmanial effects of Rosmarinic acid and Meglumine Antimoniate against Leishmania major: Integrative in silico, in vitro, and immunomodulatory investigations.

Cutaneous leishmaniasis (CL), mainly caused by Leishmania major, remains a major public health concern, particularly in endemic regions. The clinical use of pentavalent antimonials such as meglumine antimoniate (MA) is limited by toxicity, resistance, and treatment failure. Rosmarinic acid (RMA), a natural polyphenolic compound, exhibits antimicrobial and immunomodulatory properties that may enhance antileishmanial efficacy. The antileishmanial activity of RMA alone and in combination with MA was evaluated in vitro against promastigote and intracellular amastigote forms of L. major using MTT a

Immunomodulation by platelet-derived DKK1: potential for controlling disease and pathology in leishmaniasis and implications for other infectious diseases.

Parasitic infection caused by Leishmania major in BALB/c mice is a well-established example of a chronic inflammatory disease. Although chronic inflammation in parasitic infections stems from persistent interactions between parasites and host immune cells, the mechanisms by which parasitic infections induce and regulate chronic immune responses remain to be fully understood. Emerging evidence suggests that platelets contribute not only to hemostasis but also modulate immune response during infection and inflammation. This review examines the emerging role of platelet-derived Dickkopf-1 (DKK1),

Acriflavine attenuates inflammatory pathology in cutaneous leishmaniasis independently of parasite control.

Cutaneous leishmaniasis is a parasitic skin disease for which current treatments often fail, highlighting the need for new therapeutic approaches. While high parasite burdens are associated with delayed healing, excessive protective immune responses, including elevated IFN-γ production, have also been linked to worse clinical outcomes, indicating that immunopathology contributes significantly to disease progression. Acriflavine is an antimicrobial compound with reported anti-leishmanial activity and the ability to inhibit hypoxia-driven responses. Because Leishmania -infected skin is hypoxic i

In vitro evaluation of activatable melittin encapsulated in liposome and albumin nanoparticles against Leishmania.

Leishmaniasis comprises a spectrum of clinical manifestations caused by protozoan parasites of the genus Leishmania, order Trypanosomatida. Cutaneous leishmaniasis remains a significant zoonotic disease prevalent in tropical and subtropical regions, particularly in developing countries. Despite ongoing research, a definitive cure for this parasitic infection is still needed. This study explored the potential of activatable melittin (AM) as a selective treatment for cutaneous leishmaniasis caused by Leishmania major. The AM was designed using PepFold and ExPASy servers, incorporating a matrix m

Rapid phagosome formation drives parasite control in subclinical Leishmania braziliensis infection.

INTRODUCTION: Cutaneous leishmaniasis (CL) caused by Leishmania braziliensis displays a wide spectrum of clinical manifestations characterized by an inflammatory Th1 response that leads to the development of skin lesions. In endemic areas of L. braziliensis infection, individuals who present a delayed type of hypersensitivity reaction and/or IFN-γ production to soluble Leishmania antigen (SLA) without presenting or having a history of leishmaniasis are considered to have subclinical (SC) infection. SC subjects produce less IFN-γ and TNF-α as compared to CL subjects but control infection. OBJE

Cutaneous leishmaniasis promotes skin microbial dysbiosis and exacerbation of local inflammatory responses.

Cutaneous leishmaniasis (CL) is a neglected tropical disease caused by protozoan parasites belongs to the genus Leishmania transmitted to humans by the bite of the infected female sand fly. Increasing evidence suggested that alterations in the skin microbiome may influence local inflammatory responses and disease progression in CL. This study aimed to investigate the microbial community shifts associated with CL lesions using paired lesional and contralateral healthy skin samples from infected individuals (n = 8). Leishmania tropica was identified in all clinical samples by ITS-1 real-time PCR

Bone Marrow-Derived Mesenchymal Stem Cells Alleviate Cutaneous Leishmaniasis by Promoting M2 Macrophage Polarization and Skin Tissue Repair in a Murine Model.

Cutaneous leishmaniasis (CL) is the most common clinical form of leishmaniasis, characterized by persistent skin ulcers and nodules. Standard chemotherapeutic agents have substantial toxicity and do nothing to repair the damaged tissue, an unmet need that motivates the search for adjunctive strategies. Mesenchymal stem cells (MSCs) can modulate macrophage activity and support tissue regeneration, yet their role in CL has received limited attention. In this study, we tested whether bone marrow-derived MSCs (BM-MSCs) could attenuate Leishmania mexicana-induced inflammation and facilitate skin re

Dose- and time-dependent dissemination of viable Leishmania amazonensis following experimental footpad infection in mice.

Cutaneous leishmaniasis is typically characterized by localized skin lesions, but the extent to which Leishmania amazonensis disseminates beyond the primary site of infection remains incompletely defined. In this study, BALB/c mouse footpads were infected subcutaneously with L. amazonensis promastigotes at doses ranging from 105 to 108, and infection was evaluated over time up to 120 days. All infected groups developed local footpad infection, and amastigotes were detectable at the infection site early after challenge. Footpad swelling and parasite burden increased progressively, with more pro

Imported Cutaneous Leishmaniasis in India: Diagnosis and Treatment with Implications for Kala Azar Elimination.

Cutaneous leishmaniasis (CL) is an emerging infection in Kerala state. Recent reports showed that Leishmania donovani is causing the dual manifestations, CL and visceral leishmaniasis (VL). In addition, the influx of migrants and returnees from other countries has introduced CL caused by non-endemic variants of Leishmania. A 46-year-old migrant worker from Nepal presented with a single erythematous nodule with crusting and erosions. It was diagnosed as CL through molecular diagnosis, and L. donovani was proven as the parasite by ITS-1 RFLP and sequencing. Additionally, the strain was identifie

Spatiotemporal dynamics of cutaneous leishmaniasis in Yucatán, México: retrospective analysis of cases diagnosed between 2015 and 2024.

Localized cutaneous leishmaniasis (LCL) is a vector-borne disease endemic to southeastern México. Although the number of LCL cases has increased in Yucatán since 2015, detailed analyses of its epidemiological characteristics and spatiotemporal dynamics remain limited. A retrospective analysis of confirmed leishmaniasis cases reported in Yucatán from 2015 to 2024 was conducted using the state's nominal surveillance database. Incidence rates were estimated using population data from CONAPO (Consejo Nacional de Población), and spatiotemporal clusters were identified using Kulldorff's scan statist

The development of novel ferrocenyl quinoline-benzimidazole molecular hybrids against cutaneous leishmaniasis.

Cutaneous leishmaniasis, the most prevalent form of the parasitic and vector-borne disease of leishmaniasis, is mainly caused by Leishmania major (L. major) in the developing world. With no vaccine available for primary prophylaxis, secondary prophylaxis comprises commercially available therapeutic drugs, such as amphotericin B and miltefosine. However, these agents are associated with parasite-specific drug resistance, severe drug-induced toxicity to the host and high treatment costs. In this study, the strategies of molecular hybridisation of pharmacologically relevant scaffolds and metal in

Spatio-temporal modeling of zoonotic cutaneous leishmaniasis (ZCL) in the Algerian steppe: Epidemiological insights and climatic associations.

Zoonotic cutaneous leishmaniasis (ZCL) caused by Leishmania major remains a significant public health concern in Algeria. The disease is maintained through a zoonotic cycle involving wild rodents as reservoirs and sand flies as vectors, whose population dynamics are influenced by climatic conditions. In the context of global climate change, this study investigated the relationship between climatic factors and ZCL incidence, and mapped high-risk areas in the Algerian central steppe. Epidemiological and climatic data from Djelfa, Laghouat, and Tiaret Wilayas collected between 2010 and 2022 were
