A Human Monoclonal Antibody Targeting α-Gal Ameliorates Experimental Cutaneous Leishmaniasis.

Leishmaniasis is a vector-borne disease transmitted by female sand flies infected with Leishmania parasites. Cutaneous leishmaniasis is primarily caused by Leishmania major in the Old World and typically manifests as skin ulcers. The carbohydrate-rich molecules on the surface of Leishmania are crucial for the parasite's infectivity in the vertebrate host and vector. Among these, galactose moieties have been identified as promising targets for vaccines against leishmaniasis. This study investigates the potential of human monoclonal antibodies (mAbs) targeting α-Gal to prevent L major infection.

Improving Leishmania isolation in field conditions: Efficacy of caspofungin against yeasts contamination.

Cutaneous leishmaniasis (CL), a neglected disease prevalent in 88 countries, is commonly caused by Leishmania (L.) major in the Old World, posing significant public health concerns. Isolation from sand flies or infected mammals and in vitro cultivation of Leishmania parasites are critical for epidemiological studies, but these cultures are often compromised by bacterial and fungal contamination, especially when outsourced from vector digestive tracts. While the Leishmania parasite's natural resistance to antibiotics simplifies bacterial control, most antifungals also inhibit the parasites grow

Beyond the Skin: Topical Amphotericin B Nanocarriers Targeting Cutaneous Leishmaniasis with Suppression of Lymphatic Parasite Burden.

BACKGROUND/OBJECTIVES: Cutaneous leishmaniasis (CL) remains a global health challenge, with treatment options often limited by drug resistance and systemic toxicity. Amphotericin B (AmB) represents a promising alternative. but intravenous administration causes severe systemic adverse effects. Despite growing interest in topical therapies, knowledge gaps remain regarding the comparative efficacy of delivery systems, including the influence of treatment timing and potential intrinsic effects. This study aimed to develop and characterize different topical AmB formulations (polymeric nanoparticles

Tri-Layer Composite Nanofiber Wound Dressing Incorporating Glucantime and Silver Nanoparticles for Cutaneous Leishmaniasis Management.

Cutaneous leishmaniasis is a zoonotic disease caused by Leishmania parasites and leads to chronic, non-healing skin lesions. Although current drugs can control the disease, their use is limited by systemic side effects, low efficacy, and inadequate lesion penetration. Therefore, innovative local delivery systems are required to enhance drug penetration and reduce systemic toxicity. To address these challenges, silver nanoparticles (AgNPs) were synthesized using propolis extract through a green synthesis approach, and a tri-layer wound dressing composed of polyvinyl alcohol and gelatin containi

Pentamidine-Functionalized Polycaprolactone Nanofibers Produced by Solution Blow Spinning for Controlled Release in Cutaneous Leishmaniasis Treatment.

Leishmaniasis, a widespread, neglected infectious disease with limited effective treatments and increasing drug resistance, demands innovative therapeutic approaches. In this study, we report the fabrication of pentamidine (PTM)-loaded polycaprolactone (PCL) nanofibers using solution blow spinning (SBS) as a potential topical delivery system for cutaneous leishmaniasis (CL). Homogeneous PCL fiber mats were produced using a simple SBS set-up with a commercial airbrush after optimizing several working parameters. Drug release studies demonstrated sustained PTM release profile over time, which wa

Econazole Exhibits In Vitro and In Vivo Efficacy Against Leishmania amazonensis.

Background/Objectives: Cutaneous leishmaniasis (CL) remains a major neglected tropical disease, and current chemotherapeutic options are limited by toxicity and emerging resistance. Repurposing azole antifungals is a promising approach, as they target ergosterol biosynthesis, a pathway also essential in Leishmania spp. This study investigated the antileishmanial potential of econazole through in vitro and in vivo assays. Methods: Econazole activity was evaluated against Leishmania amazonensis promastigotes and intracellular amastigotes using MTT and luminescence-based methods. Cytotoxicity in

Treatment-Refractory New World Cutaneous Leishmaniasis Misdiagnosed as Recurrent Cellulitis in a UK Traveler Returning From the Amazon.

Cutaneous leishmaniasis is an uncommon consideration in non-endemic settings, where it may closely resemble bacterial skin infections and result in diagnostic delay. We report the case of a previously healthy young man in the United Kingdom who developed a progressively enlarging ulcer on the dorsum of the hand with associated nodular lymphocutaneous spread following recent travel to the Amazon rainforest. The lesion evolved over several weeks and was repeatedly treated as recurrent cellulitis with multiple courses of antibiotics, without clinical improvement. Histopathological examination and

The Unusual Invader in a Patient with Long-Standing Rheumatoid Arthritis: A Case of Leishmania major Colonization of Rheumatoid Nodules.

Rheumatoid nodules are the most common extra-articular manifestation of rheumatoid arthritis. Long-term immunomodulatory therapies, including corticosteroids, used in the management of rheumatoid arthritis are associated with a higher risk of infections. Leishmaniasis is a neglected protozoal infection that may arise in these patients. Cutaneous presentation is the most common and is characterized by a wide spectrum of clinical manifestations and courses, depending on the interplay between species involved and the host's immune response. Here, we report the rare and intriguing case of a patien

Intervention framework towards a community-driven approach to improve health-seeking in cutaneous leishmaniasis in rural Sri Lanka.

BACKGROUND: Cutaneous leishmaniasis (CL) is a re-emerging neglected tropical disease (NTD) in Sri Lanka. The prevention and control of CL mainly rely on early detection and treatment. The WHO roadmap for NTDs highlights the importance of community-based approaches to reduce disease burden. Accordingly, our study aimed to design and evaluate the feasibility of a community-based intervention framework to improve early and appropriate health-seeking in CL in rural Sri Lanka. METHODS: We designed the intervention by integrating key principles of community engagement and involvement (CEI) with the

Long pentraxin 3 (PTX3) regulates IL-17A-mediated immunity to Leishmania major infection in mice.

Cutaneous leishmaniasis, caused by protozoan parasites of the Leishmania genus, remains a significant health concern in endemic regions such as the Middle-East, Asia, Latin America, and North Africa. The disease affects millions of people worldwide, with over one million new infections reported annually. Despite its health impact, there is currently no approved vaccine largely due to limited understanding of immunological mechanisms underlying protective immunity and disease pathogenesis. We previously reported that long pentraxin 3 (PTX3), a pattern recognition molecule involved in inflammati
