Zoonotic transmission and genetic diversity of Leishmania major in Shiraz, Iran: An integrated entomological, clinical, and molecular study.

Cutaneous leishmaniasis (CL), endemic in Shiraz, Iran, is caused by Leishmania major (zoonotic cutaneous leishmaniasis, ZCL) and L. tropica (anthroponotic cutaneous leishmaniasis, ACL). This study investigated transmission drivers, including sand fly vectors, zoonotic and environmental factors, and the genetic diversity of L. major. A total of 1029 sand flies were collected indoors and outdoors in Shiraz from August to October using sticky traps. Samples from patient lesions (n = 30) and pooled female sand flies (n = 40 pools) were examined by microscopy and screened by PCR targeting kDNA and

In vitro and in vivo studies of dehydroxylated-isoquines and -isotebuquines against trypanosomatids: a preclinical drug candidate for treatment of cutaneous leishmaniasis.

The 4-aminoquinoline scaffold represents a promising platform for designing anti-trypanosomatid agents targeting essential survival pathways in Leishmania and T. cruzi. Herein we demonstrated the potential of the dehydroxylated isoquines (deOH-IQ) and isotebuquines (deOH-ITBQ) against in vitro and in vivo models of cutaneous leishmaniasis (CL) and acute Chagas disease (CD). The most active compounds displayed excellent in vitro efficacy against non-infectious and intracellular infective forms of L. braziliensis and T. cruzi, with low EC50 magnitudes. A detailed cytotoxicity analysis across div

Limonene exerts antileishmanial effects via mevalonate pathway in Leishmania amazonensis.

Limonene, a monoterpene abundant in essential oils of citrus plants, exhibits well-documented activity against both promastigote and amastigote forms of Leishmania in vitro and in vivo. Leishmania (L.) amazonensis is one of the etiological agents of localized and diffuse cutaneous leishmaniasis in the Americas, clinical forms frequently associated with limited therapeutic efficacy and recurrent treatment failure. Here, we show that limonene inhibits the biosynthesis of key isoprenoids in L. amazonensis promastigotes, including ubiquinone, dolichol, and ergosterol. Mechanistically, limonene dis

Evaluating the effect of immunization with DNA encoding Phlebotomus sergenti apyrase protein (PsSP42) against Leishmania tropica infection in BALB/c mouse model.

BACKGROUND: Leishmania parasites are transmitted through the bite of infected female sand flies. The sand fly inoculum contains both the parasite and the salivary proteins, which can modulate the immune system's function. Some of these salivary proteins have the potential to be used as a vaccine candidate. Since there have been fewer studies investigating the salivary proteins of Phlebotomus (Ph.) sergenti, this prompted us to select among the three protein members of Ph. sergenti apyrase family (PsSP40, PsSP41, and PsSP42) and measure its effectiveness as vaccine candidate against Leishmania

Antibody response induced by structural proteins from Triatoma virus as potential adjuvants in experimental immunisation models.

INTRODUCTION: Virus-Like Particles (VLPs) are viral protein structures widely used as adjuvants in vaccine formulations due to their ability to stimulate the innate immune response, thereby contributing to the activation of adaptive immunity through the production of different IgG subclasses. The present study evaluated the adjuvant potential of recombinant structural proteins of the Triatoma virus VLPs (TrV-VLPs: VP1, VP2, and VP3) in experimental immunisation protocols for American Cutaneous Leishmaniasis and Chagas disease. METHODS: BALB/c mice were immunised with native antigens of Leishm

Platelet DKK1 promotes tolerogenic dendritic cells and non-healing responses in cutaneous leishmaniasis.

Activation of platelet TLR1/2 at the initiation of Leishmania major infection results in the release of the Wnt antagonist Dickkopf-1 (DKK1). To examine the role of platelet MyD88 and/or DKK1 in regulating the immune response, genetic approaches using BALB/c mice deficient in platelet MyD88 (MyD88PKO) or DKK1 (DKK1PKO) were used. Genetically deficient mice exhibit significant changes in the immune response throughout infection. Initially, the levels of activated neutrophils and neutrophil-platelet aggregates were reduced at the site of infection. Subsequently, an anti-leishmanial Th1-response

In vitro, In silico, and Scanning Electron Microscopy Investigations of Anti-leishmanial Efficacy of New Synthetic 3-Imidazolyl flavan oxime on Leishmania tropica Parasites.

INTRODUCTION: Antimonials have been traditional therapeutic agents for leishmaniasis for decades, but selecting the optimal treatment regimen remains challenging. Alternative systemic treatments, including liposomal amphotericin B and sodium stibogluconate, have shown unclear efficacy in Leishmania tropica cutaneous leishmaniasis. The objective of this study was to evaluate the efficacy of a new synthetic 3-Imidazolyl flavan oxime (IFO) against Leishmania tropica, using in vitro, in silico, and Scanning Electron Microscopy (SEM) analyses. METHODS: A standard strain of Leishmania tropica was c

Functional Effects of Cyclization on Li1 Peptide Activity against Metacyclic Leishmania amazonensis Internalization.

Leishmaniases are caused by protozoa of the genus Leishmania, whose metacyclic promastigote forms initiate infection in the mammalian host. Building upon previous work with Leishmania infantum, this study evaluated the cyclic peptide Li1 and its linear analogue Li1nc regarding their capacity to interfere with Leishmania amazonensis infection. Both peptides were synthesized by solid-phase peptide synthesis (SPPS-Fmoc) and tested after pre-exposure of metacyclic promastigotes prior to infection of murine peritoneal and THP-1-derived macrophages. In contrast to the previous study conducted by our

Molecular Detection of Leishmania Species on Microscopically Negative Smears in Gonbad-e-Kavus, Northeastern Iran.

BACKGROUND: The diagnosis of cutaneous leishmaniasis (CL) is typically based on microscopic examination of Giemsa-stained smear. However, this method can lead to false negative and false positive results. Therefore, the study aimed to identify L. major and L. tropica species by investigating microscopically negative smears suspected of CL using the multiplex nested polymerase chain reaction (PCR) method. MATERIALS AND METHODS: This study was conducted on 227 microscopically negative specimens from individuals suspected of having CL referred to the CL Treatment and Diagnosis Center in Gonbad-e

Differential microRNA expression profiles and predicted miRNA-mRNA regulatory networks in human macrophage-like cells infected with Leishmania infantum.

MicroRNAs (miRNAs) regulate gene expression and play a crucial role in numerous diseases, including infections. Leishmaniasis is a neglected infectious disease occurring in different forms (i.e., cutaneous, mucocutaneous, and visceral) caused by a protozoan belonging to the Leishmania genus. The parasite infects mainly the macrophages, establishing a niche permissive for its proliferation. Leishmania parasites are known to modulate host gene expression, including miRNAs; however, the specific role of these miRNAs in driving host transcriptomic changes remains to be fully characterized. The aim

Therapeutic challenges in relapsing cutaneous and visceral leishmaniasis caused by Leishmania (Mundinia) martiniquensis in patients with advanced HIV disease from Southern Thailand.

BACKGROUND: Autochthonous leishmaniasis has become increasingly recognized in Thailand, with Leishmania (Mundinia) martiniquensis identified as the predominant species, particularly among immunocompromised individuals. Infected immunosuppressed patients often present with complex clinical features, which can delay diagnosis and complicate treatment. Given the limited clinical data available and emerging reports of resistant infections, improved awareness, prompt diagnosis, and optimized management strategies are urgently needed to address this underrecognized pathogen in Thailand. Toward Eliminating Leishmaniasis as a Public Health Problem in Yemen: Advocating for a One Health Approach.

Leishmaniasis is among the most neglected tropical diseases in Yemen, where both cutaneous and visceral clinical forms represent a public health challenge. Leishmania tropica is the main cause of cutaneous leishmaniasis, whereas L. donovani and L. infantum cause visceral leishmaniasis in the country. Although leishmaniasis has been recognized for decades, its actual burden in Yemen remains poorly defined, largely due to weak surveillance, constraints in diagnostic services, and the lack of a national control program. Conflict-related disruptions in healthcare, poverty, poor housing, and malnut

Successful Treatment of Cutaneous Leishmania infantum Infection with Immunocryosurgery after Failure of the Respective Monotherapies.

Herein, we present a case of cutaneous leishmaniasis successfully treated with a modified immunocryosurgery protocol. A 46-year-old male Caucasian presented with two nodules on the forehead of eight-month duration. He reported an unsuccessful attempt to treat the lesions with a session of cryosurgery elsewhere. During the clinical examination, two adjacent hard mobile nodules that did not infiltrate the underlying tissues were notable. Their diameter was 10 mm and 15 mm, respectively. There were no clinical, laboratory or imaging data in favor of a systemic infection. Histology and polymerase

Characterization of GH18 chitinase in Leishmania braziliensis: expression, structural insights, and implications for vaccine and therapeutic development.

Chitinases, a group of glycosyl hydrolases (GHs), catalyze the degradation of chitin by releasing N-acetylglucosamine subunits. GHs can be found in all three domains of life. Among the three GH families (GH18, GH19, and GH20), GH18 chitinases are the most conserved and extensively studied. These enzymes have been implicated in nutrition, immune modulation, invasion, and virulence across diverse pathogens. In some parasitic protists, GH18 chitinases are essential for transmission. However, in kinetoplastids, including Leishmania spp., these enzymes remain poorly characterized. This study aimed
