Furanocoumarin-rich extracts from Trichocline plicata as natural photosensitizers for the photodynamic inactivation of Leishmania amazonensis: Insights into ROS- and DNA-mediated mechanisms.

Cutaneous leishmaniasis (CL) is a neglected tropical disease with limited therapeutic options due to toxicity, resistance, and high costs. Photodynamic therapy offers a promising alternative; however, the systematic evaluation of furanocoumarin-rich plant extracts in Leishmania models remains limited. Despite the well-established photodynamic properties of individual furanocoumarins, no systematic evaluation of Trichocline plicata extracts has been performed in protozoan parasites. This study aimed to provide an integrated evaluation of the chemical composition, intrinsic photophysical propert

Activity of Luetzelburgia auriculata Lectin Against Leishmania major Promastigote Via Carbohydrate Recognition Domain.

INTRODUCTION: Leishmaniasis, an infectious disease, significantly impacts patients' quality of life. Current treatments primarily involve glucantime and amphotericin B. Although these drugs are effective, their limitations necessitate the exploration of alternative therapies. Lectins show promise in this context due to their established ability to interact with membrane glycans of various pathogens. OBJECTIVE: Luetzelburgia auriculata agglutinin (LAA) exhibits binding affinity for galactose and lactose carbohydrates. METHODS: Based on these properties, this study evaluated the in vitro anti-

Antigenic profiling of Leishmania amazonensis microsomal fraction to support vaccine development.

Vaccination remains the most promising long-term strategy for controlling leishmaniasis, as individuals who recover from primary infection generally develop protective immunity. However, no vaccine is currently available for human use. In this study, we identified two immunogenic proteins that may account for the protective effect previously observed with the Leishmania amazonensis microsomal fraction (MicF), which confers protection in murine models. Both proteins were expressed in Escherichia coli and used to immunize mice, eliciting specific humoral responses and promoting elevated producti

Development of a Recombinant Chimeric Antigen Expressing Different Epitopes of Leishmania spp. For Diagnosis of Human Cutaneous Leishmaniasis.

Development of highly sensitive and specific diagnostic tests for cutaneous leishmaniasis (CL) remains a significant challenge. Localized immune response in dermal leishmaniasis results in low systemic antibody production. Consequently, serologic diagnostic platform should focus on detecting Leishmania antigens directly from the skin lesions. Designing multiepitope-based proteins as an alternative to crude antigens can enhance sensitivity and specificity, lower production costs, and facilitate development of more field-amenable diagnostic tools. A chimeric multiepitope protein construct (THAG)

Myopericarditis and leishmaniasis: a case report.

BACKGROUND: Leishmaniasis belongs to a group of neglected tropical diseases worldwide, causing significant morbidity and mortality. Cardiovascular complications are rare; however, when they occur, they can be severe. Pericardial involvement with varying degrees of effusion is one of its main complications, whereas myocardial involvement remains poorly documented and not well established. CASE SUMMARY: In our case report, we present a young woman from the Amazon region, diagnosed with cutaneous leishmaniasis and exertional dyspnoea. Cardiac magnetic resonance imaging revealed active pericardit

Pilot study evaluating a newly developed nanoparticle-based lateral flow assay for the diagnosis of cutaneous leishmaniasis in Sri Lanka.

BACKGROUND: With the launch of Sri Lanka's National Strategic Plan for leishmaniasis control, case diagnosis has been identified as a key strategic intervention. However, the routine slit-skin smear (SSS) demonstrates variable sensitivity (33-78%), underscoring the need for rapid and sensitive diagnostic tools. This study evaluated the diagnostic performance of a newly developed nanoparticle-based anti-rKRP42 (recombinant kinesin-related protein antigen-42) IgG immunochromatographic test (rKRP42-ICT) for detecting cutaneous leishmaniasis caused by Leishmania donovani. METHODS: A cross-section

Development of a 17-DMAG-Loaded Carboxymethylcellulose Gel for In Vivo Treatment of Cutaneous Leishmaniasis.

Cutaneous leishmaniasis (CL) is a neglected tropical disease for which safer and more effective therapeutic options are urgently needed. Heat shock protein 90 (Hsp90) inhibitors have emerged as promising antileishmanial agents. Among geldanamycin derivatives, 17-dimethylaminoethylamino-17-demethoxygeldanamycin (17-DMAG) has previously demonstrated potent activity against Leishmania braziliensis. Here, we evaluated the therapeutic potential of a topical 17-DMAG formulation for CL. The compound exhibited low cytotoxicity toward human keratinocytes (HaCaT) and THP-1 macrophages cell-lines. Leishmaniasis in Morocco: Epidemiology, Transmission Dynamics, and the Potential of Artificial Intelligence for Disease Management.

Leishmaniases are neglected tropical diseases caused by Leishmania parasites and transmitted by infected phlebotomine sand flies, and they remain a major public health challenge in Morocco. The burden is dominated by cutaneous leishmaniasis (CL) and visceral leishmaniasis (VL), mainly associated with Leishmania major, Leishmania tropica, and L. infantum. Despite long-standing national control efforts aligned with the Sustainable Development Goals and the national ambition to eliminate leishmaniasis as a public health problem by 2030, transmission persists and continues to expand in some areas.

Habitat Associations Shape Phlebotomine Sand Fly Assemblages at the Andes-Amazon Interface in Southeastern Peru.

Phlebotomine sand flies remain poorly studied in southeastern Peru, a region with a high burden of cutaneous leishmaniasis (CL). Using modified ultraviolet (UV) light traps, we surveyed sand fly assemblages at Manu Biological Station during the wet season within secondary forest, Guadua bamboo forest, fruit crop plots, and peridomicile habitats. We collected 2641 sand flies representing 32 species. Habitat type was the primary driver of assemblage composition, with minimum nightly temperature as the strongest environmental correlate. Sand fly abundance was highest in secondary forest (n = 921)

Cutaneous Leishmaniasis in High-Altitude: A Case Report From Nepal.

Leishmaniasis is a parasitic infection caused by Leishmania and spread by the bite of a sandfly, most commonly Phlebotomus. Among the three subtypes of Leishmaniasis, cutaneous leishmaniasis is the most common form. Visceral Leishmaniasis is common in Nepal whereas few cases of cutaneous leishmaniasis have been reported in migrant Nepalese population. The most common lesion seen in cutaneous leishmaniasis is papulo-nodular although other morphological lesions can also be seen. A 30 years female currently residing in Humla presented with a history of multiple non-tender nodules with central nec

Light Photo Treatment at 405 nm Can Effectively Kill Leishmania Parasites.

Cutaneous leishmaniasis is a protozoan disease which is responsible for significant morbidity in humans. Currently, there is no clinically approved vaccine to prevent infections, and, therefore, treatments to cure skin lesions are required. Ideally, a treatment that can be self-administered to affected areas is desirable. In this study, the effect of violet-blue light, of wavelength in the region of 405 nm, on the survival of Leishmania major and Leishmania mexicana was determined using in vitro and in vivo models. Light treatment caused significant killing of both promastigotes and intracellu

Lupoid Cutaneous Leishmaniasis: A Case Report.

BACKGROUND: Lupoid Cutaneous Leishmaniasis(LCL) is rare type of cutaneous leishmaniasis that clinically and histologically mimics other granulomatous skin conditions like Lupus Vulgaris. This similarity often cause difficulty in diagnosis, especially in endemic areas. CASE PRESENTATION: We report a case of a 29-year-old male who presented with isolated plaque on right cheek persisting for 2 years. The lesion was slowly progressive, non-tender and had not responded completely to local therapy. Smear of initial lesion revealed +LD bodies but the recurrent lesion showed no amastigotes on smear.

Mechanistic evaluation of niosomal Rhoifolin in antileishmanial therapy and its additive potential with meglumine antimoniate.

Leishmaniasis is a neglected tropical disease that urgently requires safer and more effective therapeutic strategies. Rhoifolin (RF), a natural flavonoid with anti-inflammatory and antiparasitic potential, has shown promising biological activities against several infectious diseases. In the present study, the antileishmanial effects of free RF and its niosomal formulation (NRF), alone and in combination with meglumine antimoniate (MA), were investigated against Leishmania major. Niosomes were prepared using Span 40, Tween 40, and cholesterol, resulting in high encapsulation efficiency (90.9%)
