BACKGROUND: Cutaneous leishmaniasis (CL) is a major public health challenge in Ethiopia, with 20-30,000 new cases annually. Limited awareness and misconceptions contribute to delayed care seeking, stigma, prolonged healing, and reduced uptake of preventive measures in CL-endemic communities. To address these challenges, a participatory community dialogue (CD) intervention was implemented to evaluate a structured CD intervention in improving knowledge, attitudes, and preventive practices related to CL in Kalu District, Ethiopia.
METHODS: A convergent parallel mixed-methods study was conducted

INTRODUCTION: Cutaneous leishmaniasis (CL), caused by protozoa of the genus Leishmania , is a neglected tropical disease. Limited evidence exists to inform the management of CL in children and adolescents, and experience in non-endemic settings varies significantly. We report case data and treatment outcomes for 17 children and young people diagnosed with CL and treated at a specialist tropical medicine center in the United Kingdom.
METHODS: The electronic patient records of 17 patients less than 18 years old with CL who were treated at University College London Hospitals between October 2015

American cutaneous leishmaniasis (ACL) comprises a complex spectrum of immunopathological conditions in which Leishmania (V.) braziliensis and L. (L.) amazonensis elicit markedly divergent host responses. Mucosal leishmaniasis (ML) is characterized by a hyperergic, pro-inflammatory state, whereas anergic diffuse cutaneous leishmaniasis (ADCL) involves a profound impairment of cell-mediated immunity, respectively. The molecular sensors and metabolic circuits that dictate this immunological divergence remain poorly understood. We hypothesized that parasite species-specific programs orchestrate d

BACKGROUND: Cutaneous leishmaniasis (CL) is a neglected tropical disease with limited therapeutic options due to drug resistance, systemic toxicity, and prolonged treatment duration associated with pentavalent antimonials such as meglumine antimoniate (MAT). Lucilia sericata larvae produce hemolymph containing bioactive compounds with antimicrobial and immunomodulatory properties, suggesting potential as an alternative or adjunct therapy for CL.
METHODS: Hemolymph was extracted from sterile third-instar L. sericata larvae and characterized using SDS-PAGE and Fast Protein Liquid Chromatography

Cutaneous leishmaniasis (CL) is highly prevalent in northern Ethiopia but data on community knowledge, attitudes, and health-seeking behaviours remain limited. A cross-sectional survey was conducted between November and December 2022 in CL-endemic areas of Tigray using mixed sampling and a structured questionnaire administered to 512 households. Knowledge of CL transmission was poor: only 1% correctly identified sand flies as the vector, while 25% believed the disease was genetically acquired. Approximately 67% of participants perceived CL as stigmatizing, and 63.3% reported a preference for t

Cutaneous leishmaniasis (CL) is a vector-borne disease caused by Leishmania spp. Molecular methods such as conventional PCR (cPCR) and real-time PCR (qPCR) show high accuracy but remain restricted to reference centers. Loop-mediated isothermal amplification (LAMP) enables rapid DNA amplification under isothermal conditions. LAMP targeting 18S rDNA using WarmStart® Colorimetric LAMP 2× Master Mix and Bst 2.0 DNA Polymerase with SYBR® Green I (BstSg) was evaluated in swab samples (extracted by kit-SW-kit and direct boil-SW-DB) and specimens collected through biopsy from ulcerated lesions of pati

Leishmaniases are infectious diseases whose vectors are female sand flies that depend on the blood of vertebrates for egg maturation. In this study, we investigated the sand fly species and DNA from Leishmania spp. This entomological/epidemiological study was conducted in a rural area in Nova Andradina, Mato Grosso do Sul, Brazil. From March 2019 to February 2020, five modified Falcão light traps were installed at night once a month. The collected insects were separated by sex into pools of 10 specimens. A sample was used for species identification via microscopic analysis, and another sample

BACKGROUND: Skin NTDs cause mental ill health, primarily through social stigma and discrimination, overburdening services and undermining elimination efforts. A holistic One Health (OH) approach is essential for NTD elimination, but evidence and operational guidance lack. This scoping review documents the current state of integration of mental health and OH-based skin-NTD programmes. It forms part of a series of reviews on the role of OH in skin NTDs, led by the WHO skin NTD working groups.
METHODOLOGY: We searched four peer-reviewed databases for terms related to skin NTDs, OH, mental health

Macrophages are the primary host cells for Leishmania, and their polarization into either M1 (pro-inflammatory) or M2 (anti-inflammatory) phenotypes determines the course of the infection. This study aimed to investigate the effects of Leishmania tropica, the main agent of cutaneous leishmaniasis (CL) in Türkiye, on the polarization of primary human macrophages. Primary monocytes were isolated from human peripheral blood and differentiated into macrophages using GM-CSF. The cells were infected with L. tropica promastigotes, analyzed after 48 h. Polarization was evaluated through morphological

Cutaneous leishmaniasis is characterized by chronic inflammatory skin lesions in which CD8□ T cells exhibit paradoxical functions. While IFN-γ-producing CD8□ T cells contribute to the development of protective immunity in the draining lymph node, CD8□ T cells recruited to the infected skin lose their ability to produce IFN-γ and instead acquire cytotoxic functions that exacerbate tissue damage. We previously demonstrated that the hypoxic microenvironment of leishmanial lesions promotes CD8□ T cell cytotoxicity through induction of Blimp-1. Whether hypoxia also suppresses protective CD8□ T cell

Leishmaniasis is a disease caused by Leishmania species and transmitted via sandflies. Current control strategies against reservoir hosts and vectors are not eco-friendly. Using harmaline (HA) from Peganum harmala, and meglumine antimoniate (MA) could be a promising therapy. The study aimed to explore the potential treatment outcomes and action mechanisms of HA and MA against Leishmania major stages by investigating their effectiveness through molecular docking, anti-leishmanial effects, safety assessment, and apoptotic profile evaluations. According to the molecular docking results, the prote

Integrated One Health studies are essential for assessing the risks of leishmaniasis transmission. In Honduras, the most prevalent clinical presentation of leishmaniasis is the non-ulcerated cutaneous form caused by Leishmania infantum; however, research on its transmission dynamics remains limited. This study aimed to characterize the eco-epidemiology of leishmaniasis and identify potential zoonotic risk factors in the municipality of Ojos de Agua, Department of Comayagua, central Honduras, using a One Health approach. We collected sand flies using CDC light traps placed in intra- and peridom

Background/Objectives: American Tegumentary leishmaniasis typically presents as a single ulcerative lesion, but atypical clinical forms exist with diverse physiopathology and treatment responses. Disseminated cutaneous leishmaniasis is a less common presentation that is often challenging to treat and for which evidence-based clinical decisions are scant. A better understanding of treatment response may support the development of alternative therapeutic strategies. This study reports a retrospective cohort analysis using a time-to-event outcome to compare disseminated leishmaniasis with localiz

Leishmaniases, neglected tropical diseases comprising diverse clinical forms and manifestations depending upon the causative Leishmania species, remain a major public health concern globally. Broadly, the diseases are classified into cutaneous (CL), mucocutaneous (MCL) and visceral (VL) forms based on tissue invasion and pathogenesis. Conventional therapeutic agents, including antimonial compounds, amphotericin B, miltefosine, pentamidine, and paromomycin, are limited by high toxicity, emerging resistance, and poor bioavailability. Leishmania amastigotes proliferate within macrophages of the r
