Detection of antibodies against the African parasite Trypanosoma brucei using synthetic glycosylphosphatidylinositol oligosaccharide fragments.

Trypanosoma brucei (T. brucei) parasites cause two major infectious diseases in Africa: African trypanosomiasis in humans (HAT) and Nagana in animals. Despite the enormous economic and social impact, vaccines and reliable diagnostic measures are still lacking for these diseases. The main obstacle to developing accurate diagnostic methods and an active vaccine is the parasite’s ability for antigenic variation, impairment of B cell maturation, and loss of B cell memory which collectively prevent the development of a long-lasting, effective immune response. The antigenic variation is sustained by

Discovery of Novel CRK12 Inhibitors for the Treatment of Human African Trypanosomiasis: An Integrated Computational and Experimental Approach.

Background: Human African trypanosomiasis (HAT), caused by Trypanosoma brucei, is a neglected tropical disease with limited treatments, highlighting the pressing need for new drugs. Cell division cycle-2-related kinase 12 (CRK12), a pivotal protein involved in the cell cycle regulation of T. brucei, has emerged as a promising therapeutic target for HAT, yet effective CRK12 inhibitors remain lacking. Methods: An integrated strategy combining computational modeling, virtual screening, molecular dynamics (MD) simulations, and experimental validation was adopted to discover potential inhibitors ag

Synthesis and Combination Studies of Novel Dipeptide Nitriles with Curcumin for a Potent Synergistic Action Against Rhodesain, Cysteine Protease of Trypanosoma brucei rhodesiense.

Background/Objectives: Rhodesain is a cysteine protease crucial for the life cycle of Trypanosoma brucei rhodesiense, a parasite that causes the lethal form of human African trypanosomiasis. For these reasons, rhodesain is considered an important target for the drug discovery process of novel antitrypanosomal agents. Methods: In the present work, we carried out a combination study of two novel synthetic nitriles, Nitrile 1 and Nitrile 2, with curcumin, the golden multitarget nutraceutical obtained from Curcuma longa L., which we demonstrated to inhibit rhodesain in a non-competitive manner. We

Role of the nucleotide excision repair endonuclease XPF in the kinetoplastid parasite Trypanosoma brucei.

The nucleotide excision repair (NER) mechanism is responsible for removing bulky DNA damage, such as pyrimidine dimers induced by ultraviolet (UV) light. The NER pathway excises the damaged strand through incisions at the 5' and 3' ends of the damage, with the 5' incision catalyzed by the XPF-ERCC1 endonuclease complex. Here, we identify an XPF ortholog in Trypanosoma brucei, the causative agent of human African trypanosomiasis (sleeping sickness). XPF-deficient parasites exhibit hypersensitivity to UV irradiation and a slower rate of DNA damage repair. Consistent with its role in DNA repair,

Antiproliferative and Trypanocidal Activity of Ivermectin Bioconjugates.

Ivermectin (IVR), whose discovery has been Nobel-Prize-honored, is a 16-membered macrocyclic lactone used in medicine as an extremely effective and safe antiparasitic drug. In recent years, interest in this compound has grown due to its potential effectiveness in killing various types of cancer cells. However, research on the anticancer activity of IVR derivatives is limited. Additionally, the growing problem of drug resistance raises concerns about the effectiveness of this drug in the treatment of parasitic diseases. Therefore, in this work, we provide a detailed description of the synthesis

Key RNA-binding domains in the La protein establish tRNA modification levels in Trypanosoma brucei.

The RNA-binding protein La is found in most eukaryotes, and despite being essential in many organisms, its function is not completely clear. Trypanosoma brucei, the causative agent of human African trypanosomiasis, encodes a 'classical' La protein (TbLa) composed of a La-motif, two RNA recognition motifs (RRM1 and RRM2α), a C-terminal short basic motif (SBM), and a nuclear localization signal (NLS). In T. brucei, like in most eukaryotes, position 34 of tRNATyr, -Asp, -Asn and -His is modified with queuosine (Q34). The steady-state levels of queuosine-modified tRNA in the insect form (procyclic

A Cell Biologist's View on APOL1: What We Know and What We Still Need to Address.

APOL1 is the most recent member of the APOL gene family and is expressed exclusively in humans and a few higher primates. More than twenty years ago, it was discovered that APOL1 protects humans from infections by trypanosome subspecies that cause African sleeping sickness. Interestingly, by a co-evolutionary process between parasite and host, two APOL1 variants emerged, which, in addition to their trypanotoxic effects, are simultaneously associated with a significantly increased risk for various different kidney diseases, which are now summarized as APOL1-mediated kidney diseases (AMKDs). The

In Vitro Leishmanicidal Efficacy of Synthesized Arylidene Analogues of Glitazone.

Diabetes is a fast-growing health issue in low- and middle-income countries, with ~80% of diabetics living in the tropics and sub-tropics. It is a deadly condition claiming the lives of millions of individuals annually, with no therapeutic treatment available to date. The management of diabetes is thus limited to symptomatic relief by glycemic control. Furthermore, the geographical overlap of diabetes and neglected tropical diseases (NTDs) is of concern, as diabetes is known to increase infection susceptibility and severity. In contrast, diabetes-infection comorbidity can negatively affect tre

Managing African animal trypanosomiasis in Benin: Knowledge, attitudes, and practices of cattle owners in the West Atacora zone.

BACKGROUND AND AIM: African animal trypanosomiasis (AAT), transmitted by tsetse flies, severely constrains livestock productivity in sub-Saharan Africa. In Benin, limited governmental control initiatives and widespread drug misuse have raised concerns about emerging trypanocidal resistance. This study aimed to assess the knowledge, attitudes, and practices of cattle farmers in the Atacora and Donga departments of northern Benin and identify behaviors contributing to the persistence and drug resistance of AAT. MATERIALS AND METHODS: A cross-sectional survey was conducted from September to Dece

Can the tick Rhipicephalus microplus transmit Trypanosoma vivax to cattle by the transovarial and mechanical routes?

Due to the presence of Trypanosoma vivax DNA in ticks, transovarian and mechanical transmission is suspected. The present study aimed to evaluate (i) the presence of T. vivax in R. microplus that fed on animals with acute trypanosomosis; (ii) the transovarian transmission; and (iii) the ability of larvae and males to mechanically transmit T. vivax to cattle in a region without a cyclic vector. To this end, six calves were experimentally infected with T. vivax on day 0, and after different intervals, these animals were infested with cattle tick larvae. Before oviposition, the content of engorge

Geographical distribution of potential mechanical vectors implicated in Surra transmission in Spain: an entomological perspective.

BACKGROUND: Haematophagous Diptera can transmit a wide range of diseases to both humans and animals. Some species of the Trypanosoma genus rely on these vectors for transmission, either cyclically or mechanically. Trypanosoma evansi, the causative agent of Surra, is the only African-origin trypanosome species detected in Spain to date, which is mechanically transmitted. METHODS: To assess the occurrence and distribution of potential mechanical vectors at the national level, a systematic review was conducted on the Hippoboscidae, Muscidae and Tabanidae families. The review followed the methodo

Trypanosoma vivax elicits both Th1 and Th2 immunological responses in experimentally infected cattle.

Bovine trypanosomosis caused by Trypanosoma vivax is a health problem of economic importance in South America. In Ecuador, the presence of T. vivax was first reported in 2018; however, the isolates found in Ecuador are still being studied, mainly on issues related to virulence, pathogenicity, and immune response. To this end, this study aimed to evaluate the cellular and humoral adaptive immune response in vivo in experimentally infected cattle with T. vivax. The study lasted 42 days (with samples collected twice weekly) and was conducted in two cattle experimentally infected with an isolate o

Functional genomics of trypanotolerant and trypanosusceptible cattle infected with Trypanosoma congolense across multiple time points and tissues.

Human African trypanosomiasis (HAT), or sleeping sickness, is a neglected tropical disease caused by infection with trypanosome parasites (Trypanosoma spp.). These are transmitted by infected tsetse flies (Glossina spp.) and cause a similar disease in animals, known as African animal trypanosomosis (AAT), which is one of the largest constraints to livestock production in sub-Saharan Africa and causes a financial burden of approximately $4.5 billion annually. Some African Bos taurus cattle populations have an important evolutionary adaptation known as trypanotolerance, a genetically determined

Recrudescence of transmission of onchocerciasis in some endemic communities in Kaduna State, Nigeria: What is the way forward?

BACKGROUND: Onchocerciasis caused by the filarial parasite Onchocerca volvulus and transmitted by Simulium damnosum s.l. remains a public health concern in Nigeria. Infestation of S. damnosum s.l. along rivers Gurara and Kaduna; and heavy intensity of O. volvulus infection in Kaduna were documented in 1956. Control of onchocerciasis in Kaduna started 1954 with larviciding using dichlorodiphenyltrichloroethane reduced S. damnosum s.l. population by 94% in 1966. Diethylcarbamazine used for human treatment was discontinued due to toxicity. Kaduna State Ministry of Health and its partners built on

Spiroplasma, Wolbachia, Sodalis and trypanosome associations in Glossina Tachinoides from Yankari game reserve, Nigeria.

BACKGROUND: Tsetse flies are vectors of African trypanosomiasis, a disease that affects both humans and animals. Trypanosomiasis remains a threat to lives and it is an impediment to socio-economic development in sub-Saharan Africa. In spite of decades of chemotherapy and vector control, the disease has not been eradicated. Parasitic drug resistance has been developed to existing drugs, while vector control strategies are expensive and unsustainable. Therefore, there is a need to explore other control approaches, such as the transformation of tsetse fly endosymbionts to render the fly refractor

Classification of Trypanosoma brucei mammalian life cycle stages using Deep Learning Algorithms.

Accurate classification of Trypanosoma brucei bloodstream forms, slender and stumpy, is essential for understanding parasite biology and transmission dynamics. Traditional classification methods rely on flourescent transgenic parasites, as distinguishing these forms visually is highly challenging. To address this, we developed a semi-automated deep-learning pipeline that segments and classifies T. brucei bloodstream forms from unlabeled microscopic images. The pipeline consists of two key stages: (1) a segmentation step using the Cellpose algorithm, which detects and extracts individual parasi

Indigenous African chickens (Gallus gallus domesticus) lack an anti-Trypanosoma factor and have a prospect for zoonotic transmission of African trypanosomiasis.

African trypanosomes evade host immune response through antigenic variation and utilize other immune modulatory mechanisms to survive in the immunologically hostile mammalian bloodstream. However, indigenous African chicken (Gallus gallus domesticus) exhibits lower susceptibility to trypanosomes, suggesting unique resistance mechanisms; but the exact factor(s) of resistance remains elusive. This study aims to elucidate the mechanisms underlying the observed resistance of indigenous African chickens to T. brucei brucei infection, and to assess their potential role as cryptic reservoirs in zoono

Dermal trypanosomes in seropositive suspects of gambiense human African trypanosomiasis in Côte d'Ivoire.

In the population at risk of gambiense human African trypanosomiasis (gHAT), the prevalence of extravascular parasite carriage remains unclear. Here, we conducted an observational clinical study in the hypo-endemic gHAT foci of Sinfra and Bonon in Côte d'Ivoire from 2019 to 2022. A total of 74 individuals were enrolled, including 45 suspects previously found positive at least once in a serological test for gHAT and followed by the national elimination programme of Côte d'Ivoire, as well as 29 seronegative controls. No significant differences between groups were observed for any epidemiological

De novo rates of a Trypanosoma-resistant mutation in two human populations.

Mutation rates have long been measured as averages across many genomic positions. Recently, a method to measure the rates of individual mutations was applied to a narrow region in the human hemoglobin subunit beta (HBB) gene containing the site of the hemoglobin S (HbS) mutation as well as to a paralogous hemoglobin subunit delta (HBD) region, in sperm samples from sub-Saharan African and northern European donors [Melamed et al., Genome Res. 32, 488-498 (2022)]. The HbS mutation, which protects against malaria while causing sickle-cell anemia in homozygotes, originated de novo significantly mo
