Controlling rate of release of tsetse fly repellent blend by encapsulating in β-cyclodextrin nanoparticles.

Tsetse flies are major vectors of African trypanosomiasis, with devastating medical and veterinary consequences in sub-Saharan region of Africa. Insect repellents are promising tool for control of tsetse flies in the region. A four-component tsetse-repellent blend (δ-nonalactone, heptanoic acid, 4-methylguaiacol and geranyl acetone) previously formulated and optimized was encapsulated in β-cyclodextrin for a slow controlled release. Here, we explored various methods of microencapsulating (kneading, co-precipitation, heating or freeze-drying) tsetse fly repellent blend in β-cyclodextrin nanopar

Thiazolidinones and related analogues as efficient antitrypanosomal agents.

We designed and synthesised a series of thiazolidinediones and related analogues and evaluated their antiparasitic activity. A structure-activity relationship (SAR) study focused on modifications of specific parts of the molecule revealed derivatives that displayed significant activity against Trypanosoma brucei species. Notably, the analogue 6i exhibited exceptional activity, with an EC50 value of 30 nM and a selectivity index of >2000, against the protozoan Trypanosoma brucei rhodesiense, which causes human African trypanosomiasis. Additionally, compounds 6a, 6k, 7e, and 18 demonstrated anti

Pharmacokinetics of Racemic Eflornithine in Human Plasma and Cerebrospinal Fluid: Clinical Perspectives for L-eflornithine Against Human African Trypanosomiasis.

Intravenous dosing of L- and D-eflornithine in a racemic mixture is a currently recommended late-stage gambiense human African trypanosomiasis (g-HAT) treatment, either as 14-day monotherapy or in combination with oral nifurtimox for seven days. However, an oral eflornithine treatment against late-stage g-HAT would be preferable. Pharmacokinetics of eflornithine are enantioselective with different oral absorption of the enantiomers. L-eflornithine has a greater in vitro antitrypanosomal potency than D-eflornithine. This study aimed to integrate knowledge about in vitro potency and literature d

From pan-active to parasite-selective antiparasitic agents: A scaffold hopping approach.

Vector-borne parasitic diseases (VBPDs) represent a major global public health concern, with human African trypanosomiasis (HAT), Chagas disease, leishmaniasis, and malaria collectively threatening millions of people, particularly in developing regions. Climate change may further influence their transmission and geographic spread, increasing the global burden. As drug resistance continues to rise, there is an urgent need for novel therapeutic agents to expand treatment options and limit disease progression. Exploiting a cell-based phenotypic approach, we had previously developed 1,3,4-oxadiazo

Spatiotemporal changes in prevalence of Sodalis glossinidius, Spiroplasma spp. and trypanosome species in wild Glossina tachinoides from Sora-Mboum animal African trypanosomiasis focus in northern Cameroon.

Sterile Insect Technique (SIT) has proven effective to reduce tsetse population density in large infected areas where animal African trypanosomosis (AAT) and human African trypanosomiasis (HAT) elimination was difficult to achieve. However, the decrease in mass production of insectary-reared tsetse and the limited but incomplete knowledge on symbiont-trypanosome interaction over time, impede large-scale use of SIT. We investigated the spatiotemporal changes in symbiont prevalence and symbiont-trypanosome interactions in wild tsetse of Sora-Mboum AAT focus in northern Cameroon, collected in 201

Hidden diversity and host specificity of bat trypanosomes in East and Central Africa.

Bats are recognized as key reservoir hosts for a variety of trypanosomes, yet their full diversity and host associations remain underexplored in Africa. In this study, we investigated the diversity and phylogenetic relationships of trypanosomes infecting bats in Uganda and the Central African Republic (CAR) using molecular tools. A total of 22 distinct genotypes were identified, clustering into seven well-supported phylogroups, including known species such as Trypanosoma dionisii, T. livingstonei, T. vespertilionis, and T. erneyi, alongside several novel lineages. Notably, T. erneyi was restri

Molecular docking to homology models of human and Trypanosoma brucei ERK8 that identified ortholog-specific inhibitors.

Human African Trypanosomiasis (HAT), also known as sleeping sickness, is a lethal disease caused by two vector-borne parasites: Trypanosoma brucei gambiense and Trypanosoma brucei rhodesiense. The limited number of antitrypanosomal therapies for treating these deadly parasites suffer from toxicity, poor efficacy, and unspecified targets; thus, more and better medicines are needed. We used in silico methods to predict features of the bioactive compound AZ960 that make it an ortholog-specific inhibitor for the extracellular-signal regulated kinase 8 of T. brucei (TbERK8). Our homology models sho

Genome-wide overexpression screen reveals a novel transcription termination factor in Trypanosoma brucei.

Trypanosoma brucei is a protozoan parasite that causes African trypanosomiasis. We performed an overexpression library screen to gain insights into essential cellular processes in T. brucei and identified genes whose overexpression impaired parasite growth. One of the three top-ranking candidates we validated exhibited a notable phenotype. Tb FOP, an uncharacterized protein, has an N-terminal arginine-rich domain and a C-terminal FOP (Friend Of PRMT1) domain. Tb FOP overexpression substantially increases antisense transcription across the genome, particularly at Transcription Termination Sites

Optimization of Quinazolines for Inhibition of Trypanosoma brucei Proliferation.

A large population of people remain at risk of contracting human African trypanosomiasis (HAT), which has significant health implications and leads to death. To prepare for future epidemics and provide alternative therapies in case of resistance to current treatment options for HAT, there is a need to continue developing novel therapeutics. Previously, we reported the repurposing and reoptimization of human kinase inhibitor, Lapatinib, toward inhibiting Trypanosoma brucei proliferation. These efforts improved the selectivity and potency; however, the physicochemical properties of derivatives s

Parasitological, serological and molecular evaluation of Trypanosoma vivax in cattle experimentally infected and early treated with isometamidium chloride in a region lacking the biological vector.

This study aimed to evaluate whether early treatment with isometamidium chloride (ISM) affects the production of anti-Trypanosoma vivax antibodies in cattle and whether it can induce parasitological cure, based on tissue analysis 546 days post-treatment in animals without evidence of parasitological relapse. Ten male cattle were experimentally infected with ≈ 1 × 106 viable T. vivax trypomastigotes (D-6) and treated with ISM six days post-infection (D0). The animals were euthanized 546 days post-treatment. During this period, the presence of T. vivax in blood was assessed using direct methods

IL-4Rα signalling influences behavioural and immune responses in Trypanosoma brucei-infected mice: Evidence for iloprost as a neuroprotective agent.

This study investigated the effects of Trypanosoma brucei infection on neuroinflammation, immune response, and behaviour in both wild-type (WT) and IL-4Rα inhibited (IL-4Rα┴) mice. To achieve this, 9-week-old WT and IL-4Rα┴ mice were infected with T. brucei intraperitoneally (5 ×102 parasites) and the treated groups received 200 μg/kg/day of Iloprost intraperitoneally. Results from infected animals showed that behavioural activity and inflammation were reduced in animals treated with Iloprost. Euthanasia was performed on 12 days post-infection (dpi), and prefrontal cortex (PFC), Hippocampus (H

Thermo-related degeneration of stumpy forms of Trypanosoma brucei, the pathogen of African sleeping sickness.

Trypanosoma brucei, the causative agent of African trypanosomiasis, develops from the long slender (LS) to the short stumpy (SS) form in the mammalian host. The SS trypanosomes are critical for transmission to the insect vector but face significant challenges within the vertebrate host. The role of the immune response in controlling the parasitaemia is well studied, however, the mechanism underpinning the rapid degeneration of SS trypanosomes during the first parasitaemic peak in mice remains somewhat elusive. We demonstrate that fever is a critical yet underexplored factor in facilitating the

Interventions for Neglected Diseases Caused by Kinetoplastid Parasites: A One Health Approach to Drug Discovery, Development, and Deployment.

Kinetoplastids are protozoa that possess a unique organelle called a kinetoplast. These include the parasites Trypanosoma cruzi, T. brucei and related African trypanosomes, and Leishmania spp. These parasites cause a variety of neglected tropical diseases in humans and livestock, with devastating consequences. In the absence of any vaccine, pharmaceutical interventions are the mainstay of control, but these have historically been underfunded, fragmented, and inadequately aligned with the complex zoonotic and ecological realities of the parasites' transmission dynamics. In this review, the land

Targets of the human antibody response to the variant surface glycoprotein of Trypanosoma brucei.

Human African Trypanosomiasis (HAT) is a severe disease endemic to Sub-Saharan Africa caused by the parasite Trypanosoma brucei. Diagnosis of the chronic form of HAT relies heavily on serological screening tests that detect specific antibodies. Trypanosoma brucei parasites rely on the antigenic variation of their variant surface glycoproteins (VSG) to evade host antibody recognition. Although VSG is the primary target of host antibodies, studies of the antibody response to VSG have been limited by low-throughput approaches that allow for the analysis of only a few VSG proteins at a time, prima

Extending Thioflavin T Fluorescence Probe to 2-Ethenyl-benzothiazole Derivatives: Drug-like Quadruplex Ligands with Potent Antitrypanosomatid Activity.

Thioflavin T (ThT) is a well-established fluorescence probe with selectivity for G-quadruplex (G4) structures. Over the past few years, G4 ligands have emerged as promising candidates for the development of antiparasitic agents. Building on this concept, we explored extending ThT's benzothiazole scaffold by introducing various 2-ethenyl aromatic and heteroaromatic moieties, aiming to enhance G4 binding affinity and potential therapeutic effect. A series of benzothiazolium derivatives were synthesized and evaluated for their antiproliferative and antiparasitic activity. Several 2-ethenyl benzot
